    Scavengers protection of cells against ALA-based photodynamic therapy-induced damage

    The exogenously stimulated formation of intracellularly generated protoporphyrin IX, a precursor of haem, is becoming one of the fastest developing areas in the field of photodynamic therapy (PDT). We tested the action of several free radical scavengers, amino acids, antioxidants and sulphur-containing compounds as protectors from photodamage induced by 5-aminolaevulinic acid (ALA)-mediated PDT, employing the LM2 cell line, derived from a mammary murine adenocarcinoma. We exposed the cells to different concentrations of the compounds, 24 h before PDT, during PDT, and 19 h after treatment. We defined the protection grade (PG) as the ratio between cell survival after ALA-PDT treatment in the presence of the protector and cell survival of ALA-PDT treatment alone. We found that L-tryptophan (PG=9.2 at 2 mM), reduced glutathione (GSH) (PG=5.8 at 0.8 mM), N-acetyl-L-cysteine (PG=4.86 at 30 mM), melatonin (PG=4.5 at 8 mM) and L-methionine (PG=4.0 at 0.8 mM) are the best protectors from PDT damage, followed by L-cysteine (PG=2.8 at 0.8 mM), mannitol (PG=2.6 at 20 mM) and glycine (PG=2.4 at 40 mM) whereas oxidised glutathione and S-adenosyl-L-methionine do not exert any protection. We did not found any photoactive action of the protectors in absence of ALA. These results can be considered to modulate the photodamage induced by ALA-PDT.Fil: Perotti, Carlos Hernan.     Non frequent porphyrias in the argentinean population

    Las Porfirias son consecuencia de fallas en el metabolismo del Hemo. Se clasifican según el tipo de sintomatología clínica prevalente o el órgano donde se expresa preferencialmente la falla metabólica. En general la deficiencia enzimática  está asociada a mutaciones en los genes que codifican para cada una de las enzimas. Están descriptos 7 tipos de Porfiria diferentes. Se transmiten por carácter autosómico dominante a excepción de la PCE, la PHE y la NPA que son recesivas. Sin embargo, están reportadas variantes homocigotas para el resto de las Porfirias de pronóstico y evolución mucho más grave que la forma heterocigota. La descripción de estos casos poco frecuentes, sus tratamientos y evolución, facilitarían tanto el diagnóstico diferencial de la Porfiria como el conocimiento de las posibilidades terapéuticas en cada caso. Asimismo para las Porfirias heterocigotas con manifestación infantil, su identificación temprana y tratamiento aseguraría una mejor evolución minimizando los riesgos asociados. Se han diagnosticado 5 casos de Porfirias Agudas en niñas: 2 de PAI, 2 de PV y 1 de CPH. Entre las Porfirias Cutáneas se presentan 25 casos de PCT infantil, el primer caso de PHE en Argentina, 4 casos de PCE infantil y 1 en un adulto y 2 casos de PPE con compromiso hepatobiliar.The Porphyrias are a group of diseases resulting from partial deficiencies in one of the heme biosynthetic enzymes. These disorders can be classified on the basis of their clinical manifestations or according the organ where the metabolic deficiency is mainly expressed. In general this enzyme deficiency is associated with mutations in the genes which codify each enzyme. There are 7 types of Porphyrias. They are autosomal dominant disorders with the exception of PCE, PHE and NPA which are recessive. However, some rare and severe cases with recessive inheritance have also been reported. The description of these infrequent cases and their treatments and evolution would make easier the differential diagnosis of Porphyrias as well as the therapeutic possibilities to be applied in each case. Moreover, it is very important the early identification and treatment of infantile heterozygous porphyrias to avoid the risks of associatedd complications. In the CIPYP we have diagnosed 5 cases of infantil Acute Porphyrias: 2 PAI, 2 PV and 1 CPH. In the group of Cutaneous Porphyrias we present 25 cases of infantil PCT, the first case of PHE in Argentina, 4 cases of infantil PCE and 1 adult PCE and 2 cases of PPE with hepatic failure.Fil: Melito, Viviana Alicia.     Sevoflurane: Its action on heme metabolism and phase I drug metabolizing system

    Acute attacks of porphyria are most commonly precipitated by events that decrease heme concentrations. Enzyme inducing-drugs are the most important triggering factors, particularly in relation to anaesthesia. We have reported previously that Enflurane and Isoflurane produced significant heme metabolism alterations, indicating that the use of these anaesthetics in porphyric patients should be avoided. The aim of this work was to evaluate the effect of the anaesthetic Sevoflurane on heme pathway and drug metabolizing Phase I system in mice. To this end, animals received different doses of the anaesthetic (1-2 ml/kg) and were sacrificed at different times (5-60 min). Data revealed important alterations in the enzymes involved in Acute Intermittent Porphyria, such as an induction in hepatic 5-Aminolevulinic acid synthetase activity and a diminished Porphobilinogen deaminase activity in liver and blood 20 minutes after Sevoflurane administration to mice in a dose of 1.5 ml/kg. Heme oxygenase activity was also induced, indicating the onset of oxidative stress. Total CYP levels and CYP2E1 expression were enhanced. As a consequence of these events, heme free pool would be depleted. In conclusion, our results in mice would suggest that Sevoflurane should be used with caution and very careful control in porphyric patients.Fil: Sampayo, Rocío Guadalupe.     Trypanocidal activity of tioamide-sustituted imidazoisoquinolinones: Electrochemical properties and biological effects

    Three thioamide-substituted imidazoisoquinolinones, which possess an heterocyclic center similar to tryptanthrin and are named C1, C2 and C3, were studied regarding: a) its in vitro anti-Trypanosoma cruzi activity, b) its cytotoxicity and electrochemical behaviour and c) its effect on cell viability, redox state and mitochondrial function . The assayed compounds showed a significant activity against the proliferative forms, but only C1 showed activity on the trypomastigote form (for C1, IC50 epi = 1.49 µM; IC50 amas = 1.74 µM and IC50 try = 34.89 µM). The presence of an antioxidant compound such as ascorbic acid or dithiotreitol induced a three-fold increase in the antiparasitic activity, whereas glutathione had a dual effect depending on its concentration. Our results indicate that these compounds, which exhibited low toxicity to the host cells, can be reduced inside the parasite by means of the pool of low molecular weight thiols, causing oxidative stress and parasite death by apoptosis. The antiparasitic activity of the compounds studied could be explained by a loss of the capacity of the antioxidant defense system of the parasite to keep its intracellular redox state.     Clinical, biochemical, and genetic characterization of acute hepatic porphyrias in a cohort of Argentine patients

    Background: Acute Hepatic Porphyrias (AHPs) are characterized by an acute neuroabdominal syndrome including both neuropsychiatric symptoms and neurodegenerative changes. Two main hypotheses explain the pathogenesis of nervous system dysfunction: (a) the ROS generation by autooxidation of 5-aminolevulinic acid accumulated in liver and brain; (b) liver heme deficiency and in neural tissues that generate an oxidative status, a component of the neurodegenerative process. Methods: We review results obtained from Acute Intermittent Porphyria (AIP) and Variegate Porphyria (VP) families studied at clinical, biochemical, and molecular level at the CIPYP in Argentina. The relationship between the porphyric attack and oxidative stress was also evaluated in AHP patients and controls, to identify a marker of neurological dysfunction. Results: We studied 116 AIP families and 30 VP families, 609 and 132 individuals, respectively. Genotype/phenotype relation was studied. Oxidative stress parameters and plasma homocysteine levels were measured in 20 healthy volunteers, 22 AIP and 12 VP individuals. Conclusion: No significant difference in oxidative stress parameters and homocysteine levels between the analyzed groups were found.Fil: Martinez, María del Carmen.     Heme metabolism, oxidative and nitrosative markers in a mouse model of Hemochromatosis: Effect of Isoflurane, ethanol and 5-aminolevulinic acid

    Hereditary hemochromatosis (HH) is characterized by iron homeostasis alterations. Association between HH and Porphyria Cutanea Tarda has been reported. The aim was to characterize oxidative and nitrosative stress status and its relationship with heme metabolism in a hemochromatosis mouse model (Hfe-/-), and to evaluate the effects of Isoflurane, ethanol and 5-aminolevulinic acid (ALA). Male and female Hfe−/− and wild-type C57BL/6J mice received Isoflurane (2 ml/kg); ethanol (30%) or ALA (40 mg/kg). In male Hfe-/-, reduced glutathione (GSH) was diminished respect to C57BL/6J mice. Female Hfe-/- showed higher levels of GSH and total antioxidant capacity than male Hfe-/-. Catalase activity was lower in male and female Hfe-/- than in controls. 5-Aminolevulinic acid synthetase activity was higher in male and female Hfe-/- than in controls. In male Hfe-/-, Porphobilinogen deaminase (PBG-D) activity was augmented and Heme oxygenase (HO) activity was diminished probably to avoid iron increase. Isoflurane and ethanol reduced PBG-D and increased HO activities. HO induction would indicate oxidative stress instauration being more striking due to ethanol that also induced Superoxide dismutase activity. Isoflurane reduced Nitric Oxide Synthase expression. ALA altered antioxidant system. In Hfe-/- mice different metabolisms were altered being more affected by the drugs studied. Findings here described would contribute to increase the knowledge about the association between HH and the Porphyrias and about the effects of volatile anaesthetics on different metabolisms in genetic models of Porphyrias and associated diseases.Fil: Buzaleh, Ana Maria.     The effect of nitric oxide on Heme Oxygenase gene expression in soybean leaves

    Heme oxygenase (HO, EC 1.14.99.3) catalyzes the oxidative conversion of heme to biliverdin IXalpha with the concomitant release of carbon monoxide and iron. Recently, HO has been involved in the protection against oxidative stress in plants. The fact that nitric oxide (NO), an endogenous signaling molecule in animals and plants mediates responses to abiotic and biotic stresses, prompted us to study whether this molecule could modulate HO-1 gene transcription. To fulfill this objective leaves of soybean (Glycine max L.) plants were stimulated with Cd, employing an acute intoxication model. Cadmium caused dehydration, chlorophyll loss and ion leakage. Semi-quantitative RT-PCR analysis showed no augmentation of HO-1 transcript levels with respect to controls. Pretreatment with 100 uM sodium nitroprussiate (SNP), a well-known NO donor, prevented the effects caused by Cd. When the HO-1 mRNA levels were analyzed, a significant augmentation (54%) was observed with respect to Cd-treated plants. On the other hand, 50 or 300 uM SNP did not fully prevent the effects elicited by Cd. When HO-1 transcript levels were analyzed, no significant enhancement or a down-regulation was observed. The potassium salt of 2-(4-carboxylphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (cPTIO), a specific NO scavenger, arrested NO-mediated protective effects against to Cd-induced oxidative damage. These data provide an understanding of one of the possible roles that NO can play against an oxidative insult. NO is cytoprotective depending on its concentration, and it was further demonstrated that this protection could be, at least in part, mediated by an enhancement of HO-1 mRNA, as it happens with genes associated with the antioxidant defense system.Fil: Noriega, Guillermo Osvaldo.     Acute Intermittent Porphyria in a Man with Dual Enzyme Deficiencies

    Porphyrias are a heterogeneous group of metabolic disorders that result from the altered activity of specific enzymes of the hemebiosynthetic pathway and are characterized by accumulation of pathway intermediates. Porphyria cutanea tarda (PCT) is the mostcommon porphyria and is due to deficient activity of uroporphyrinogen decarboxylase (UROD). Acute intermittent porphyria(AIP) is the most common of the acute hepatic porphyrias, caused by decreased activity of hydroxymethylbilane synthase(HMBS). An Argentinean man with a family history of PCT who carried the UROD variant c.10_11insA suffered severe abdominalpain. Biochemical testing was consistent with AIP, and molecular analysis of HMBS revealed a de novo variant:c.344 + 2_ + 5delTAAG. *is is one of the few cases of porphyria identified with both UROD and HMBS mutations and the firstconfirmed case of porphyria with dual enzyme deficiencies in Argentina.Fil: Cerbino, Gabriela Nora.     Tumor cell lines resistant to ALA-mediated photodynamic therapy and possible tools to target surviving cells

    We isolated and characterized cell lines resistant to aminolevulinic acid (ALA)-mediated photodynamic therapy (PDT) derived from a murine adenocarcinoma and studied cross resistance with other injuries. The most resistant clones were numbers 4 and 8, which exhibited 6.7- and 4.2-fold increase in resistance respectively. Several characteristics were altered in these clones. A 2-fold increase in cell volume, higher cell spreading, and a more fibroblastic, dendritic pattern, were the morphology features that led us to think they could have different adhesive, invasive or metastatic phenotypes. The amount of porphyrins synthesized per cell in the resistant clones was similar to the parental line but, when it was expressed per mg protein, there was a 2-fold decrease, with a higher proportion of hydrophilic porphyrins. These cells were not cross-resistant to photosensitization with Benzoporphyrin derivative and Merocyanine 540, but exhibited a slight resistance to exogenous protoporphyrin IX treatment. Both clones displayed higher protein content and increased number of mitochondria, together with a higher oxygen consumption. The distinctive features found in the resistant lines led as to think how to exploit the changes induced by PDT treatment to target surviving cells. Those hypoxic cells can be also a preferential target of bioreductive drugs and hypoxia-directed gene therapy, and would be sensitive to treatment with other photosensitizers.Fil: Casas, Adriana Gabriela.     Haplotype analysis of single nucleotide polymorphisms of cyp1a1 and cyp1a2 and its relationship with the development of porphyria cutanea tarda

    Abstract : Objective: Porphyria Cutanea Tarda results from decrease hepatic uroporphyrinogen decarboxylase activity. There are twoprincipal types: sporadic or acquired and familial or hereditary Porphyria Cutanea Tarda. Hereditary Porphyria Cutanea Tarda is observed in 25-30% of the patients in whom one allele of the uroporphyrinogen decarboxylase gene reduces the enzyme activity by approximately 50% in all tissues. There are experimental evidences that suggest a possible relationship between CYP1A1, CYP1A2 andPorphyria Cutanea Tarda symptomatology development but the results described in dif erent populations are very conflictive.The aim ofthis work is provide additional evidences about the participation of the CYP system on the manifestation of this disease. Subjets/Methods: We analyse the polymorphisms already described in these CYPs isoforms in 112 Porphyria Cutanea Tarda Argentinianpatients and 89 controls. Of these 64 were used for haplotype analysis. The polymorphisms were detected by RFLP-PCR or sequencing. Results: For CYP1A2*1F polymorphism C allele (wt) was exceptionally the less frequent while the polymorphic A allele, with an increasedtranscriptional activity, resulted to be the risk allele. For CYP1A1 m4 polymorphism, for which are not previous reports about this effect, our results indicated that the A variant would be arisk factor to develop the disease. For m2 polymorphism we obtained contradictory results depending on the groups compared. The risk haplotype calculated with the snpSTATs was m4-m2-m1-1A2: C-G-C-C. Conclusions: These results indicated that, among other factors, these polymorphisms would be related with Porphyria Cutanea Tardadevelopment.Fil: Gordillo, Diego Miguel.
