    Determination of polycyclic aromatic hydrocarbons (PAHs) in Yerba-mate herbal drink

    Yerba-mate (Y-M), known as llex-paraquariensis is traditional South American herbal drink that is consumed as an aqueous extract from the Y-M leaves. This drink is thought to have long-term health benefits and thus it has recently gained more popularity. However, health hazards of polyaromatic hydrocarbons (PAHs) deposited in Y-M during drying process has been linked to the development of different types of cancer in particular oesophageal cancer. The current study developed and validated an extraction and analytical method to determine the presence of one of the PAH compounds, namely; benzo(b)fluoranthene [Bbf] as an indicator for the PAH(s) occurrence in Y-M in commercial samples of the dried leaves. Since the expected level of contamination with this compound is at trace level thus a sensitive and selective analytical method was needed. Gas Chromatography-Mass Spectrometry GC-MS method was developed using selected ion monitoring (SIM) with split-less mode focusing on 126 ion and 252 ion (main) for [Bbf]. The content of [Bbf] using different extraction methods was found in a range 0.02 to 0.09 μg/g. GC-MS results indicated a good repeatability of the peak area with strong correlation and linearity between mean peak area and concentration from the calibration curve. Soxhlet extraction using hexane as a solvent gave the highest yield of [Bbf] concentration. However, ultrasonic extraction using hot distilled water as a solvent is recommended as it better simulates the way of making and drinking

    'Managing My Patellofemoral Pain’:the creation of an education leaflet for patients

    STUDY DESIGN: Qualitative, including consultation with international experts and patients. PURPOSE: Develop a brief yet comprehensive evidence-based education leaflet to be used as an adjunct in the management of patellofemoral pain (PFP) through consultation with both experts (clinical academics) and individuals with PFP. BACKGROUND: Appropriate patient education is an essential component of effective PFP management. However, there are currently no published educational resources for clinicians and researchers treating individuals with PFP to help translate current evidence into clinical practice. METHODS: A preliminary education leaflet titled ‘Managing My Patellofemoral Pain’ was created using information from the ‘Best Practice Guide to Conservative Management of Patellofemoral Pain’ and educational content used in published research. Feedback was sought from 21 experts (clinical academics) for accuracy, adequacy and clarity of the information in the leaflet using a semistructured questionnaire, and a number of suggested modifications were made as a result. Further feedback was sought from 20 patients diagnosed with PFP regarding the clarity and adequacy of information contained in the leaflet, and to determine additional educational resource needs. RESULTS: The leaflet created is titled ‘Managing My Patellofemoral Pain’ and the main topics of the leaflet are ‘What might cause my knee pain?’ and ‘Treatment options’, which are divided into exercise and additional treatments. Patient feedback was positive, and included a number of considerations for further education resource development. CONCLUSIONS: The ‘Managing My Patellofemoral Pain’ education leaflet may provide a valuable resource for patients, clinicians and researchers to assist the provision of education and translation of the current evidence

    Reconstruction of haplotype-blocks selected during experimental evolution.

    The genetic analysis of experimentally evolving populations typically relies on short reads from pooled individuals (Pool-Seq). While this method provides reliable allele frequency estimates, the underlying haplotype structure remains poorly characterized. With small population sizes and adaptive variants that start from low frequencies, the interpretation of selection signatures in most Evolve and Resequencing studies remains challenging. To facilitate the characterization of selection targets, we propose a new approach that reconstructs selected haplotypes from replicated time series, using Pool-Seq data. We identify selected haplotypes through the correlated frequencies of alleles carried by them. Computer simulations indicate that selected haplotype-blocks of several Mb can be reconstructed with high confidence and low error rates, even when allele frequencies change only by 20% across three replicates. Applying this method to real data from D. melanogaster populations adapting to a hot environment, we identify a selected haplotype-block of 6.93 Mb. We confirm the presence of this haplotype-block in evolved populations by experimental haplotyping, demonstrating the power and accuracy of our haplotype reconstruction from Pool-Seq data. We propose that the combination of allele frequency estimates with haplotype information will provide the key to understanding the dynamics of adaptive alleles

    In heutigen Unternehmen werden im Kern alle Aufgaben durch Anwendungssysteme direkt oder durch diese unterstützt erledigt. Folglich beschreiben betriebliche Anwendungssysteme heute im Grunde, welche Aufgaben in Unternehmen überhaupt zu lösen sind und welche davon automatisiert und somit durch Software erledigt bzw. unterstützt werden können. Die Arbeit an und mit Betrieblichen Anwendungssystemen ist gekennzeichnet durch eine große thematische Breite und demonstriert die für die Wirtschaftsinformatik charakteristische Nutzung von Ansätzen der Betriebswirtschaftslehre sowie der Informatik. Deswegen erwarteten die Herausgeber sehr heterogene Themenvorschläge und sie wurden nicht enttäuscht. Die letztlich ausgewählten Themen stellen aktuelle Entwicklungs- und anwendungsorientierte Forschungsprojekte zu Geschäftsprozessen, Standardsoftware, Softwareentwicklung und Betrieb von Anwendungssystemen vor. Dadurch beschreiben sie das heute existierende Berufsbild von Wirtschaftsinformatikern und -innen in der industriellen Praxis

In a first step the implementation of an Arbitrary Lagrangian Eulerian (ALE) method into AVBP is described, allowing to move solid boundaries. Then the principles and implementation of the Conditioned Temporal Interpolation (CTI) mesh management technique is described, and some specific adaptations for LES simulations are discussed. Finally a first validation of the so obtained LES IC engine code is presented by comparing predictions with findings on the square piston experiment

    AlmostSignificant:simplifying quality control of high-throughput sequencing data

    Motivation: The current generation of DNA sequencing technologies produce a large amount of data quickly. All of these data need to pass some form of quality control (QC) processing and checking before they can be used for any analysis. The large number of samples that are run through Illumina sequencing machines makes the process of QC an onerous and time-consuming task that requires multiple pieces of information from several sources. Results: AlmostSignificant is an open-source platform for aggregating multiple sources of quality metrics as well as run and sample meta-data associated with DNA sequencing runs from Illumina sequencing machines. AlmostSignificant is a graphical platform to streamline the QC of DNA sequencing data, to store these data for future reference together with extra meta-data associated with the sequencing runs not typically retained. This simplifies the challenge of monitoring the volume of data produced by Illumina sequencers. AlmostSignificant has been used to track the quality of over 80 sequencing runs covering over 2500 samples produced over the last three years.
