Parede celular e digestibilidade in vitro da matéria seca da silagem de gliricídia aditivada com coprodutos.
O objetivo do trabalho foi avaliar a digestibilidade in vitro da matéria seca (DIVMS), a fibra detergente neutro (FDN), fibra detergente ácido (FDA), hemicelulose, celulose e lignina da silagem de Gliricídia in natura, emurchecida e aditivada com 10% do peso verde com sorgo (grão moído), raspa de mandioca e co-produto de vitivinícola com um, três, cinco, sete, 14, 28 e 56 dias de fermentação em silos experimentais de PVC (50x10cm). O delineamento experimental utilizado foi inteiramente casualizado. As médias foram comparadas pelo teste SNK (P<0,01) e os dias de abertura foram submetidos ao estudo de regressão para as variáveis avaliadas. Os valores de DIVMS entre os tratamentos variaram de 46,7 % (co-produto de vitivinícola) a 55,8% (Raspa de mandioca) e não houve diferença entre a DIVMS do material original e das silagens com 56 dias de fermentação, evidenciando-se uma boa preservação do material ensilado

A 3D printed drug delivery implant formed from a dynamic supramolecular polyurethane formulation
Using a novel molecular design approach, we have prepared a thermo-responsive supramolecular polyurethane as a matrix material for use in drug eluting implants. The dynamic supramolecular polyurethane (SPU) is able to self-assemble through hydrogen bonding and π-π stacking interactions, resulting in an addressable polymer network with a relatively low processing temperature. The mechanical properties of the SPU demonstrated the material was self-supporting, stiff, yet flexible thus making it suitable for hot-melt extrusion processing, inclusive of related 3D printing approaches. Cell-based toxicity assays revealed the SPU to be non-toxic and therefore a viable candidate as a biocompatible polymer for implant applications. To this end, the SPU was formulated with paracetamol (16 %w/w) and 4 wt% or 8 wt% poly(ethylene glycol) (PEG) as an excipient and hot melt extruded at 100 °C to afford a 3D printed prototype implant to explore the extended drug release required for an implant and the potential manipulation of the release profile. Furthermore, rheological, infra-red spectroscopy, powder X-ray diffraction and scanning electron microscopy studies revealed the chemical and physical properties and compatibility of the formulation components. Successful release of paracetamol was achieved from in vitro dissolution studies and it was predicted that the drug would be released over a period of up to 8.5 months with hydrophilic PEG being able to influence the release rate. This extended release time is consistent with applications of this novel dynamic polymer as a drug eluting implant matrix
Teores de matéria seca e proteína bruta, nitrogênio amoniacal e pH de silagem de gliricídia aditivados com diferentes co-produtos.
Objetivou-se avaliar os teores de matéria seca (MS), proteína bruta (PB), nitrogênio amoniacal (N-NH3/N-T) e pH da silagem de Gliricídia in natura, emurchecida e aditivada com 10% do peso verde com sorgo (grão moído), raspa de mandioca e co-produto de vitivinícola com um, três, cinco, sete, 14, 28 e 56 dias de fermentação em silos experimentais de PVC (50x10cm). O delineamento experimental utilizado foi inteiramente casualizado. As médias foram comparadas pelo teste SNK (P<0,01) e os dias de abertura foram submetidos ao estudo de regressão para as variáveis avaliadas. A adição do sorgo, raspa de mandioca e co-produto de vitivinícola reduziram os valores de pH (4,64; 4,69 e 4,33, respectivamente) e de PB (24,13; 20,63 e 24,23%). O conteúdo de N-NH3/N-T aumentou com o decorrer dos processos fermentativos e a proteólise não estabilizou durante os 56 dias de fermentação
Detection of pathogenic Bacteria by Electrochemical Impedance Spectroscopy: Influence of the immobilization strategies on the sensor performance
Electrochemical impedance spectroscopy (EIS) is applied to detect pathogenic E. coli O157:H7 bacteria via a label free immunoassay-based detection method. Polyclonal anti-E.coli antibodies (PAb) are immobilized onto gold electrodes following two different strategies, via chemical bond formation between antibody amino groups and a carboxylic acid containing self-assembled molecular monolayer (SAM) and alternatively by linking a biotinylated anti-E. coli to Neutravidin on a mixed-SAM. Impedance spectra for sensors of both designs for increasing concentrations of E. coli are recorded in phosphate buffered saline (PBS). The Nyquist plots can be modeled with a Randle equivalent circuit, identifying the charge transfer resistance RCT as the relevant concentration dependent parameter. Sensors fabricated from both designs are able to detect very low concentration of E. coli with limits of detection as low as 10-100 cfu/ml. The influence of the different immobilization protocols on the sensor performance is evaluated in terms of sensitivity, dynamic range and resistance against nonspecific absorption
Predicting the onset and persistence of episodes of depression in primary health care. The predictD-Spain study: Methodology
Background:
The effects of putative risk factors on the onset and/or persistence of depression remain unclear. We aim to develop comprehensive models to predict the onset and persistence of episodes of depression in primary care. Here we explain the general methodology of the predictD-Spain study and evaluate the reliability of the questionnaires used.
Methods:
This is a prospective cohort study. A systematic random sample of general practice attendees aged 18 to 75 has been recruited in seven Spanish provinces. Depression is being measured with the CIDI at baseline, and at 6, 12, 24 and 36 months. A set of individual, environmental, genetic, professional and organizational risk factors are to be assessed at each follow-up point. In a separate reliability study, a proportional random sample of 401 participants completed the test-retest (251 researcher-administered and 150 self-administered) between October 2005 and February 2006. We have also checked 118,398 items for data entry from a random sample of 480 patients stratified by province.
Results:
All items and questionnaires had good test-retest reliability for both methods of administration, except for the use of recreational drugs over the previous six months. Cronbach's alphas were good and their factorial analyses coherent for the three scales evaluated (social support from family and friends, dissatisfaction with paid work, and dissatisfaction with unpaid work). There were 191 (0.16%) data entry errors.
Conclusion:
The items and questionnaires were reliable and data quality control was excellent. When we eventually obtain our risk index for the onset and persistence of depression, we will be able to determine the individual risk of each patient evaluated in primary health car
Atlantic reef fish biogeography and evolution
Copyright © 2007 The Authors.Journal compilation © 2007 Blackwell Publishing Ltd.AIM: To understand why and when areas of endemism (provinces) of the tropical Atlantic Ocean were formed, how they relate to each other, and what processes have contributed to faunal enrichment. RESULTS: Phylogenetic (proportion of sister species) and distributional (number of shared species) patterns are generally concordant with recognized biogeographical provinces in the Atlantic. The highly uneven distribution of species in certain genera appears to be related to their origin, with highest species richness in areas with the greatest phylogenetic depth. Diversity buildup in Atlantic reef fishes involved (1) diversification within each province, (2) isolation as a result of biogeographical barriers, and (3) stochastic accretion by means of dispersal between provinces. The timing of divergence events is not concordant among taxonomic groups. The three soft (non-terrestrial) inter-regional barriers (mid-Atlantic, Amazon, and Benguela) clearly act as ‘filters’ by restricting dispersal but at the same time allowing occasional crossings that apparently lead to the establishment of new populations and species. Fluctuations in the effectiveness of the filters, combined with ecological differences among provinces, apparently provide a mechanism for much of the recent diversification of reef fishes in the Atlantic
Revealing potential functions of hypothetical proteins induced by genistein in the symbiosis island of Bradyrhizobium japonicum commercial strain SEMIA 5079 (= CPAC 15).
BACKGROUND: Bradyrhizobium japonicum strain SEMIA 5079 (= CPAC 15) is a nitrogen-fixing symbiont of soybean broadly used in commercial inoculants in Brazil. Its genome has about 50% of hypothetical (HP) protein-coding genes, many in the symbiosis island, raising questions about their putative role on the biological nitrogen fixation (BNF) process. This study aimed to infer functional roles to 15 HP genes localized in the symbiosis island of SEMIA 5079, and to analyze their expression in the presence of a nod-gene inducer. RESULTS: A workflow of bioinformatics tools/databases was established and allowed the functional annotation of the HP genes. Most were enzymes, including transferases in the biosynthetic pathways of cobalamin, amino acids and secondary metabolites that may help in saprophytic ability and stress tolerance, and hydrolases, that may be important for competitiveness, plant infection, and stress tolerance. Putative roles for other enzymes and transporters identified are discussed. Some HP proteins were specific to the genus Bradyrhizobium, others to specific host legumes, and the analysis of orthologues helped to predict roles in BNF. CONCLUSIONS: All 15 HP genes were induced by genistein and high induction was confirmed in five of them, suggesting major roles in the BNF process. The complete sequence of a 9000 bp fragment of the right arm of Saccharomyces cerevisiae chromosome VII contains four previously unknown open reading frames
We report the sequence of a 9000 bp fragment from the right arm of Saccharomyces cerevisiae chromosome VII. Analysis of the sequence revealed four complete previously unknown open reading frames, which were named G7587, G7589, G7591 and G7594 following standard rules for provisional nomenclature. Outstanding features of some of these proteins were the homology of the putative protein coded by G7589 with proteins involved in transcription regulation and the transmembrane domains predicted in the putative protein coded by G7591
Population expansion of the invasive Pomacentridae Chromis limbata (Valenciennes, 1833) in Southern Brazilian coast: long-term monitoring, fundamental niche availability and new records
Human-mediated species invasions are recognized as a leading cause of global biotic homogenization and extinction. Studies on colonization events since early stages, establishment of new populations and range extension are scarce because of their rarity, difficult detection and monitoring. Chromis limbata is a reef-associated and non-migratory marine fish from the family Pomacentridae found in depths ranging between 3 and 45 m. The original distribution of the species encompassed exclusively the eastern Atlantic, including the Azores, Madeira and the Canary Islands. It is also commonly reported from West Africa between Senegal and Pointe Noire, Congo. In 2008, vagrant individuals of C. limbata were recorded off the east coast of Santa Catarina Island, South Brazil (27° 41' 44″ S, 48° 27' 53″ W). This study evaluated the increasing densities of C. limbata populations in Santa Catarina State shoreline. Two recent expansions, northwards to São Paulo State and southwards to Rio Grande do Sul State, are discussed, and a niche model of maximum entropy (MaxEnt) was performed to evaluate suitable C. limbata habitats. Brazilian populations are established and significantly increasing in most sites where the species has been detected. The distributional boundaries predicted by the model are clearly wider than their known range of occurrence, evidencing environmental suitability in both hemispheres from areas where the species still does not occur. Ecological processes such as competition, predation and specially habitat selectivity may regulate their populations and overall distribution range.
