    Use of synchrotron tomographic techniques in the assessment of diffusion parameters for solute transport in groundwater flow

    This technical note describes the use of time-resolved synchrotron radiation tomographic energy dispersive diffraction imaging (TEDDI) and tomographic X-ray fluorescence (TXRF) for examining ion diffusion in porous media. The technique is capable of tracking the diffusion of several ion species simultaneously. This is illustrated by results which compare the movement of Cs+, Ba2+ and La3+ ions from solution into a typical sample of English chalk. The results exhibited somewhat anomalous (non-Fickian) behaviour and revealed heterogeneities (in 1D) on the scale of a few millimetres

    RR Lyrae Variables in M33. I. Evidence For a Field Halo Population

    We present observations of RR Lyrae variables in the Local Group late-type spiral galaxy M33. Using the Advanced Camera for Surveys on the Hubble Space Telescope, we have identified 64 ab-type RR Lyraes in M33. We have estimated reddenings for these stars based on their minimum light V-I colors and metallicities based on their periods. From the distributions of these properties, we conclude that the RR Lyraes belong to two populations - one associated with the halo of M33 and the other with its disk. Given that RR Lyraes are produced by populations older than ~10 Gyr, this suggests that not only does the field halo of M33 contain an old component, but so does its disk. This is one of the best pieces of evidence for the existence of a halo field component in M33. Using a relation between RR Lyrae absolute magnitude and metallicity (Mv(RR) = 0.23[Fe/H] + 0.93), we estimate a mean distance modulus of = 24.67 +/- 0.08 for M33. This places M33 approximately 70 kpc beyond M31 in line-of-sight distance.Comment: 20 pages, 17 figures, accepted for publication in The Astronomical Journa

    Unfurling Fern Biology in the Genomics Age

    Twenty-first century technology is addressing many of the questions posed by 20th-century biology. Although the new approaches, especially those involving genomic data and bioinformatic tools, were first applied to model organisms, they are now stretching across the tree of life. Here, we review some recent revelations in the ferns. We first examine how DNA sequence data have contributed to our understanding of fern phylogeny. We then address evolution of the fern plastid genome, including reports of high levels of RNA editing. Recent studies are also shedding light on the evolution of fern nuclear genomes. Initial analyses of genomic data suggest that despite their very high chromosome numbers homosporous ferns may have experienced relatively few rounds of genome duplication. Genomic data are enabling researchers to examine speciation rates and the mechanisms underlying the formation of new fern species. We also describe genetic tools that have been used to study gene function and development in ferns. Recent findings in fern biology are providing insights that are not only pertinent to this major component of the land flora but can also help to provide an improved evolutionary context for research on flowering plants

    Superconducting and normal-state properties of the noncentrosymmetric superconductor Re6Zr

    We systematically investigate the normal and superconducting properties of non-centrosymmetric Re6_{6}Zr using magnetization, heat capacity, and electrical resistivity measurements. Resistivity measurements indicate Re6_{6}Zr has poor metallic behavior and is dominated by disorder. Re6_6Zr undergoes a superconducting transition at Tc=(6.75±0.05)T_{\mathrm{c}} = \left(6.75\pm0.05\right) K. Magnetization measurements give a lower critical field, μ0Hc1=(10.3±0.1)\mu_{0}H_{\mathrm{c1}} = \left(10.3 \pm 0.1\right) mT. The Werthamer-Helfand-Hohenberg model is used to approximate the upper critical field μ0Hc2=(11.2±0.2)\mu_{0}H_{\mathrm{c2}} = \left(11.2 \pm 0.2\right) T which is close to the Pauli limiting field of 12.35 T and which could indicate singlet-triplet mixing. However, low-temperature specific-heat data suggest that Re6_{6}Zr is an isotropic, fully gapped s-wave superconductor with enhanced electron-phonon coupling. Unusual flux pinning resulting in a peak effect is observed in the magnetization data, indicating an unconventional vortex state.Comment: 11 pages, 7 figures, 2 table

    Probing the superconducting ground state of the noncentrosymmetric superconductors CaTSi3 (T = Ir, Pt) using muon-spin relaxation and rotation

    The superconducting properties of CaTSi3 (where T = Pt and Ir) have been investigated using muon spectroscopy. Our muon-spin relaxation results suggest that in both these superconductors time-reversal symmetry is preserved, while muon-spin rotation data show that the temperature dependence of the superfluid density is consistent with an isotropic s-wave gap. The magnetic penetration depths and upper critical fields determined from our transverse-field muon-spin rotation spectra are found to be 448(6) and 170(6) nm, and 3800(500) and 1700(300) G, for CaPtSi3 and CaIrSi3 respectively. The superconducting coherence lengths of the two materials have also been determined and are 29(2) nm for CaPtSi3 and 44(4) nm for CaIrSi3.Comment: 6 pages, 7 figure

    Changes in muscle cell cation regulation and meat quality traits are associated with genetic selection for high body weight and meat yield in broiler chickens

    Between-breed genetic variation for muscle and meat quality traits was determined at eight weeks of age in 34 lines of purebred commercial broiler and layer lines and traditional breeds (categories) of chickens. Between-breed genetic variation for plasma ion concentrations and element concentration in muscle dry matter and ash were determined. Plasma from broilers had higher concentrations of Na+, K+, Mg++, total and free Ca++ and lower free:total Ca++ than plasma from layer and traditional lines. Muscle from broilers contained more Na and higher concentrations of K, Mg and Ca per mg of ash but not of dry matter compared with layer and traditional lines. In comparison with layer and traditional lines, broiler genotypes were over three times heavier, their plasma creatine kinase activity (CK), a marker of muscle tissue damage, was higher, their breast muscle colour was lighter (L*) and less red (a*) and yellow (b*) in appearance, the initial and final pH of their muscles were lower, the pH change was higher and their breast muscle was more tender. Thus, genetic selection for broiler traits has markedly altered cation regulation in muscle cells and may be associated with changes in muscle cell function and the development of pathology and meat quality problems

    Testing self-report time-use diaries against objective instruments in real time

    This study provides a new test of time-use diary methodology, comparing diaries with a pair of objective criterion measures: wearable cameras and accelerometers. A volunteer sample of respondents (n = 148) completed conventional self-report paper time-use diaries using the standard UK Harmonised European Time Use Study (HETUS) instrument. On the diary day, respondents wore a camera that continuously recorded images of their activities during waking hours (approximately 1,500–2,000 images/day) and also an accelerometer that tracked their physical activity continuously throughout the 24-hour period covered by the diary. Of the initial 148 participants recruited, 131 returned usable diary and camera records, of whom 124 also provided a usable whole-day accelerometer record. The comparison of the diary data with the camera and accelerometer records strongly supports the use of diary methodology at both the aggregate (sample) and individual levels. It provides evidence that time-use data could be used to complement physical activity questionnaires for providing population-level estimates of physical activity. It also implies new opportunities for investigating techniques for calibrating metabolic equivalent of task (MET) attributions to daily activities using large-scale, population-representative time-use diary studies

    Local Population Structure and Patterns of Western Hemisphere Dispersal for Coccidioides spp., the Fungal Cause of Valley Fever.

    UnlabelledCoccidioidomycosis (or valley fever) is a fungal disease with high morbidity and mortality that affects tens of thousands of people each year. This infection is caused by two sibling species, Coccidioides immitis and C. posadasii, which are endemic to specific arid locales throughout the Western Hemisphere, particularly the desert southwest of the United States. Recent epidemiological and population genetic data suggest that the geographic range of coccidioidomycosis is expanding, as new endemic clusters have been identified in the state of Washington, well outside the established endemic range. The genetic mechanisms and epidemiological consequences of this expansion are unknown and require better understanding of the population structure and evolutionary history of these pathogens. Here we performed multiple phylogenetic inference and population genomics analyses of 68 new and 18 previously published genomes. The results provide evidence of substantial population structure in C. posadasii and demonstrate the presence of distinct geographic clades in central and southern Arizona as well as dispersed populations in Texas, Mexico, South America, and Central America. Although a smaller number of C. immitis strains were included in the analyses, some evidence of phylogeographic structure was also detected in this species, which has been historically limited to California and Baja, Mexico. Bayesian analyses indicated that C. posadasii is the more ancient of the two species and that Arizona contains the most diverse subpopulations. We propose a southern Arizona-northern Mexico origin for C. posadasii and describe a pathway for dispersal and distribution out of this region.ImportanceCoccidioidomycosis, or valley fever, is caused by the pathogenic fungi Coccidioides posadasii and C. immitis The fungal species and disease are primarily found in the American desert southwest, with spotted distribution throughout the Western Hemisphere. Initial molecular studies suggested a likely anthropogenic movement of C. posadasii from North America to South America. Here we comparatively analyze eighty-six genomes of the two Coccidioides species and establish local and species-wide population structures to not only clarify the earlier dispersal hypothesis but also provide evidence of likely ancestral populations and patterns of dispersal for the known subpopulations of C. posadasii

    Elevated levels of inflammatory cytokines predict survival in idiopathic and familial pulmonary arterial hypertension

    BACKGROUND: Inflammation is a feature of pulmonary arterial hypertension (PAH), and increased circulating levels of cytokines are reported in patients with PAH. However, to date, no information exists on the significance of elevated cytokines or their potential as biomarkers. We sought to determine the levels of a range of cytokines in PAH and to examine their impact on survival and relationship to hemodynamic indexes. METHODS AND RESULTS: We measured levels of serum cytokines (tumor necrosis factor-alpha, interferon-gamma and interleukin-1beta, -2, -4, -5, -6, -8, -10, -12p70, and -13) using ELISAs in idiopathic and heritable PAH patients (n=60). Concurrent clinical data included hemodynamics, 6-minute walk distance, and survival time from sampling to death or transplantation. Healthy volunteers served as control subjects (n=21). PAH patients had significantly higher levels of interleukin-1beta, -2, -4, -6, -8, -10, and -12p70 and tumor necrosis factor-alpha compared with healthy control subjects. Kaplan-Meier analysis showed that levels of interleukin-6, 8, 10, and 12p70 predicted survival in patients. For example, 5-year survival with interleukin-6 levels of >9 pg/mL was 30% compared with 63% for patients with levels < or = 9 pg/mL (P=0.008). In this PAH cohort, cytokine levels were superior to traditional markers of prognosis such as 6-minute walk distance and hemodynamics. CONCLUSIONS: This study illustrates dysregulation of a broad range of inflammatory mediators in idiopathic and familial PAH and demonstrates that cytokine levels have a previously unrecognized impact on patient survival. They may prove to be useful biomarkers and provide insight into the contribution of inflammation in PAH
