Inhibitory activity of jackfruit (Atrocarpus heterophyllus Lam.) leaf extract on the growth of Porphyromonas gingivalis bacteria
Background: Periodontitis is an inflammation that affects the periodontal tissue, causing pocket formation, loss of attachment, and tooth mobility. Periodontitis caused by a complex interaction between the body's immune response and microorganisms in dental plaque. One of the main agents in the etiology of chronic periodontitis is Porphyromonas gingivalis. Jackfruit (Atrocarpus heterophyllus Lam.) leaf contains active compounds such as saponin, flavonoid, and tannin.
Objective: This study is aimed to determine the inhibition of jackfruit leaf extract (Atrocarpus heterophyllus Lam.) on the growth of Porphyromonas gingivalis bacteria.
Method: This research is an in vitro laboratory experiment. Jackfruit leaf extraction was carried out by maceration method with 70% ethanol solvent, then diluted serially by dilution to obtain various concentrations. Concentrations used include 100%; 50%; 25%; 12.5%; 6.25%; 3.125%; 1.56%; and 0.78%. Inhibition test against Porphyromonas gingivalis bacteria was carried out by spectrophotometric method and colony count.
Result: Jackfruit leaf extract with a concentration of 12.5%; 25%; 50%; and 100% can kill Porphyromonas gingivalis bacterial colonies resulting in 0 colonies. A concentration of 6.25% inhibited the growth of bacteria by 91.77%. The results of statistical tests obtained a significance value of 0.001 (P <0.05).
Conclusion: Jackfruit leaf extract (Atrocarpus heterophyllus Lam.) has an inhibitory effect on the growth of Porphyromonas gingivalis bacteria with MIC values at a concentration of 6.25% and MBC at a concentration of 12.5%
Viability Testof Lemongrass Extract (Cymbopogon Citratus) on BHK-21 Fibroblast Cell Culture
Background: Lemongrass are found in Indonesia thus it lemongrass can be used as an alternative in healing
inflammation by conducting a preliminary study in the form of BHK-21 fibroblast cell viability test after 24
hours of extract lemongrass.
Objective: To determine the viability of extract lemongrass against BHK-21 fibroblast cell cultures. Method: BHK-21 fibroblast cells were taken from neonatal hamsters’ kidneys and planted in 96 well plates. Extracts
lemongrass with concentrations of 100%, 50%, 25%, 12.5%, 6.25%, 3.13% and 1.56% were included in each well prepared and incubated for 24 hours. MTT Assay was performed to see the viability of extract lemongrass on fibroblast cells.
Results: Extract lemongrass viable against BHK-21 fibroblast cells at a concentration of 100%. There is a
decrease in viability at concentrations of 50% to 1.56%.
Conclusion: The 100% concentration showed the highest viability of BHK-21 fibroblast cells and extract lemongrass had potential in wound healing
Peningkatan Kesehatan Periodontal Ibu Hamil Dalam Upaya Mengurangi Resiko Bayi Dengan Berat Badan Lahir Rendah
OPG and RANKL Signal Transduction in Osteoblast Regulation Post Application Extract Collagen in Osteogenesis
Background and Aim: The current goal of periodontal therapy is to achieve periodontal regeneration. Important factor for periodontal regeneration is to promote bone formation, nowadays used bone replacement. The used of xenograft materials as gold standard for periodontal tissue regeneration using type I collagen bone graft has been widely developed. The main organic component in fish scales is type I fibril collagen, which are same as with component in bones. Specific markers of bone formation are the expression of osteoblast, osteoclast, osteoprotegerin (OPG), and receptor activator of nuclear factor κB ligands (RANKL). The purpose of this study is to assess the expression of OPG and RANKL after application of extract collagen from gourami (Osphronemus gouramy) scales. Materials and Methods: Thirty-two experimental 3-month-old male Wistar albino rats (150g and 200g) were randomly divided into four groups: 7-day control group, 7-day fish collagen group, 14-day control group and 14-day fish collagen group. The sockets were filled with blood (control group), whereas 10 mg to 30 mg extract collagen was added until the sockets were fully occupied (treatment group). Results: The ANOVA test showed a significance level of 0.000 (p <0.05). Conclusion: Expression of OPG enhanced and expression of RANKL lowered after application of type I collagen from gourami scales, accelerating osteogenesis
Viability of Nigella sativa Toothpaste with SLS Compared Non-SLS on Fibroblast Cell Culture
beneficial effects in infectious disease, such as gingival inflammation. One of the most widely used
synthetic detergents in toothpaste is Sodium lauryl sulphate (SLS). SLS is used to decrease water's
surface tension. The side effects of SLS include oral epithelial sloughing, ulcerations, inflammation,
protein denaturation, and membrane expansion. This study aimed to determine the viability of
human gingival fibroblast (HGFs) cultured with Nigella sativa toothpaste extract containing 2% SLS
and Nigella sativa toothpaste without SLS. HGFs were grown in DMEM medium then challenged
with Nigella sativa toothpaste with 2% SLS (Nigella sativa-SLS group), Nigella sativa toothpaste
without SLS (non-SLS group), and control group. Based on the present study, the cell viability of
Nigella sativa-2% SLS, non-SLS, and control group were 92.33%, 96.30%, and 98.99%,
respectively. In conclusion, both Nigella sativa with 2% SLS and non-SLS were non-toxic to HGFs.
Thus, Nigella sativa toothpaste is potential to treat gingivitis and oral diseases. However, future in
vivo study for this is needed
