Mary Anning’s legacy to French vertebrate palaeontology

peer reviewedThe real nature of marine reptile fossils found in England in between the 1700s to the beginning of the 1900s remained enigmatic, until Mary Anning's incredible fossil discoveries and their subsequent study by eminent English and French scientists. In 1820, Georges Cuvier acquired several ichthyosaur specimens found by Mary Anning, now kept or displayed in the Palaeontology Gallery of the MNHN in Paris. Four years later, Cuvier obtained a plesiosaur specimen from Mary Anning, only the second ever discovered. Cuvier was fascinated by these fossils and their study allowed him to apply his comparative anatomical method and to support his catastrophist theory. We re-examined these important specimens from an historical point of view and herein describe them taxonomically for the first time since Cuvier’s works. The Paris specimens belong to two different ichthyosaur genera (Ichthyosaurus and Leptonectes) and one plesiosaur genus (Plesiosaurus)

Jeu de rôle pédagogique dans un projet universitaire

International audienceDans nos formations scientifiques et techniques, nous devons sensibiliser les étudiants à la nécessité de transmettre leurs connaissances. Cette situation de transmission des savoirs ne conduira pas les étudiants vers l'enseignement mais leur sera utile face à un interlocuteur (client ou supérieur). Elle consistera à présenter, expliquer leur produit ou travail à cet interlocuteur. Dans ces conditions, les étudiants sont confrontés à des publics assez éloignés de leurs compétences techniques de base. Cependant, dans le cadre universitaire, trouver un client candide n'est pas facile. Nous avons proposé des sujets de projets atypiques à des étudiants de 2ème année en DUT GEII. Les étudiants devaient imaginer, penser, proposer et livrer des démonstrateurs d'une plate-forme informatique en direction d'un public jeune (élèves de primaire ou secondaire). Le niveau technique des sujets n'étant pas très important (pour eux), ils doivent prendre de la hauteur pour quitter leur rôle d'apprenant afin de coopérer avec ce public inhabituel et de se projeter dans son univers de pensées pour réussir à convaincre. En les bousculant ainsi, nous espérions leur ouvrir l'esprit sur un autre aspect du travail technique (complémentaire aux compétences techniques). Ils devaient apprendre alors, à communiquer simplement, précisément et efficacement afin de capter l'attention et obtenir l'adhésion à leur projet

Une Association de Compétences entre Elektron et Hippocrate

International audienceThis paper presents an association of skills that was established a few weeks ago, between a particular medical sector (therapies adopted in neu roscience) and a scientific and technological department at the University of Orléans (training department in Electrical Engineering and Industrial Computing (Geii)). Throughout the project, a common working strategy was adopted and led to the creation of an operational prototype in a context of providing progressive skills for the students who were in charge of the project. The various constraints that have been managed in terms of cost, time and performance are detailed in this paper. The working scenario adopted with L2 students (pedagogy by project) shows the great interest of this working approach.Cet article présente une association de compétences qui a été établie il y a quelques semaines, entre un secteur médical particulier (les thérapies adoptées en neuroscience) et un département scientifique et technologique à l’Université d’Orléans (département de formation en DUT Génie Électrique et Informatique Industrielle (Geii)). Tout au long du projet, une stratégie de travail commune a été adoptée et a permis la réalisation d’u n prototype opérationnel dans un contexte d’apport de compétences progressives pour les étudiants qui étaient en charge du projet. Les différentes contraintes qui ont été gérées en matière de coût, délai, performance sont détaillées dans cet article. Le scénario de travail adopté avec des étudiants en L2 (pédagogie par projet) montre tout l’intérêt de cette démarche de travail

Des exemples de briques technologiques dans le cadre d'une application pour l'industrie du futur

Tous les jours nous en avons la démonstration au travers de nouveaux modes d'organisation autant marchands qu'industriels de la société. Nous sommes confrontés à des ruptures majeures aussi bien liées à l'évolution des technologies qu'à la mondialisation de l'économie avec l'émergence et la maturité de nouveaux acteurs : le défi de la transition énergétique, la révolution du numérique, la protection de notre planète, les convergences entre les sciences du vivant et des sciences dures… Cet article présente des applications implémentées dans le cadre de la mise en place de technologie " Internet of Things " par l'utilisation d'un réseau Profinet. Ces applications peuvent être utilisées sur l'ensemble des processus de l'entreprise dans le cadre de l'Industrie du Futur. Le standard OPC est apparu au milieu des années 90 pour faciliter les échanges entre le monde des automatismes et celui de la supervision basée sur PC. Les dernières spécifications d'OPC UA (Unified Architecture) sont aujourd'hui validées. Avec cette nouvelle version, la fondation OPC révolutionne son standard de communication entre équipements industriels. OPC UA rompt les liens qui le rendaient indissociable de Windows, pour se déployer sur tous types de plateformes

Claudin Loss-of-Function Disrupts Tight Junctions and Impairs Amelogenesis

Claudins are a family of proteins that forms paracellular barriers and pores determining tight junctions (TJ) permeability. Claudin-16 and -19 are pore forming TJ proteins allowing calcium and magnesium reabsorption in the thick ascending limb of Henle's loop (TAL). Loss-of-function mutations in the encoding genes, initially identified to cause Familial Hypomagnesemia with Hypercalciuria and Nephrocalcinosis (FHHNC), were recently shown to be also involved in Amelogenesis Imperfecta (AI). In addition, both claudins were expressed in the murine tooth germ and Claudin-16 knockout (KO) mice displayed abnormal enamel formation. Claudin-3, an ubiquitous claudin expressed in epithelia including kidney, acts as a barrier-forming tight junction protein. We determined that, similarly to claudin-16 and claudin-19, claudin-3 was expressed in the tooth germ, more precisely in the TJ located at the apical end of secretory ameloblasts. The observation of Claudin-3 KO teeth revealed enamel defects associated to impaired TJ structure at the secretory ends of ameloblasts and accumulation of matrix proteins in the forming enamel. Thus, claudin-3 protein loss-of-function disturbs amelogenesis similarly to claudin-16 loss-of-function, highlighting the importance of claudin proteins for the TJ structure. These findings unravel that loss-of-function of either pore or barrier-forming TJ proteins leads to enamel defects. Hence, the major structural function of claudin proteins appears essential for amelogenesis

Therapeutic depletion of CCR8+ tumor-infiltrating regulatory T cells elicits antitumor immunity and synergizes with anti-PD-1 therapy.

BACKGROUND: Modulation and depletion strategies of regulatory T cells (Tregs) constitute valid approaches in antitumor immunotherapy but suffer from severe adverse effects due to their lack of selectivity for the tumor-infiltrating (ti-)Treg population, indicating the need for a ti-Treg specific biomarker. METHODS: We employed single-cell RNA-sequencing in a mouse model of non-small cell lung carcinoma (NSCLC) to obtain a comprehensive overview of the tumor-infiltrating T-cell compartment, with a focus on ti-Treg subpopulations. These findings were validated by flow cytometric analysis of both mouse (LLC-OVA, MC38 and B16-OVA) and human (NSCLC and melanoma) tumor samples. We generated two CCR8-specific nanobodies (Nbs) that recognize distinct epitopes on the CCR8 extracellular domain. These Nbs were formulated as tetravalent Nb-Fc fusion proteins for optimal CCR8 binding and blocking, containing either an antibody-dependent cell-mediated cytotoxicity (ADCC)-deficient or an ADCC-prone Fc region. The therapeutic use of these Nb-Fc fusion proteins was evaluated, either as monotherapy or as combination therapy with anti-programmed cell death protein-1 (anti-PD-1), in both the LLC-OVA and MC38 mouse models. RESULTS: We were able to discern two ti-Treg populations, one of which is characterized by the unique expression of Ccr8 in conjunction with Treg activation markers. Ccr8 is also expressed by dysfunctional CD4+ and CD8+ T cells, but the CCR8 protein was only prominent on the highly activated and strongly T-cell suppressive ti-Treg subpopulation of mouse and human tumors, with no major CCR8-positivity found on peripheral Tregs. CCR8 expression resulted from TCR-mediated Treg triggering in an NF-κB-dependent fashion, but was not essential for the recruitment, activation nor suppressive capacity of these cells. While treatment of tumor-bearing mice with a blocking ADCC-deficient Nb-Fc did not influence tumor growth, ADCC-prone Nb-Fc elicited antitumor immunity and reduced tumor growth in synergy with anti-PD-1 therapy. Importantly, ADCC-prone Nb-Fc specifically depleted ti-Tregs in a natural killer (NK) cell-dependent fashion without affecting peripheral Tregs. CONCLUSIONS: Collectively, our findings highlight the efficacy and safety of targeting CCR8 for the depletion of tumor-promoting ti-Tregs in combination with anti-PD-1 therapy

This squalid world on the outskirts of New York testifies to the mechanistic destruction of the land. However, the writer’s apprehension of this augean non-place gives it an aesthetic dimension. The artistic gesture transforms the anthropocenic landscape into a readable object. In this coupling of the real and the imaginary, what Edward Soja calls 'third-space,' geopoetic perspectives appear and transform what seemed to be a 'non-landscape' into an allegorical, transfigured territory

Habiter un lieu nécessite une démarche d’identification au territoire. Les liens qui unissent identité et territorialité sont ici étudiés à travers l'examen de deux démarches en apparence opposées. Les gated communities, enclaves résidentielles fermées qui ont fait leur apparition il y a une dizaine d’années dans le Montana, répondent à la volonté pour certains de se forger une identité collective basée sur l’entre-soi. Ici, le besoin d'habiter un espace cerné, borné permet de renforcer le sentiment d'appartenance et d'existence. Pour d'autres, tel l'écrivain Rick Bass, la construction identitaire nécessite un rapport fusionnel entre soi et un espace que l'on souhaite garder inviolé, territoire de l’intime et de la contemplation. Cependant, une analyse plus approfondie montre que, quelle que soit la démarche envisagée, l’individu a besoin de borner son espace pour s’identifier à lui. Qu’il soit naturel ou construit, le balisage ancre le lieu et territorialise celui qui s’y installe
