    Development and characterization of microsatellite markers for Piptadenia Gonoacantha (fabaceae)

    Microsatellite primers were designed for Piptadenia gonoacantha (Fabaceae) and characterized to estimate genetic diversity parameters. The species is a native tree from the Atlantic Forest biome commonly used in forest restoration; it has medicinal potential and the wood is economically useful. Twenty-eight microsatellite loci were identified from an enriched genomic library. Fifteen loci resulted in successful amplifications and were characterized in a natural population of 94 individuals. Twelve loci were polymorphic, with allele numbers ranging from three to 15 per locus, and expected and observed heterozygosities ranging from 0.2142 to 0.8325 and 0.190 to 0.769, respectively.     The Cabreúva tree, Myroxylon peruiferum, is an endangered tropical species from Brazil used in forest restoration projects. It is known for its medicinal properties. Eleven microsatellite markers were developed for this species, from a microsatellite-enriched library.     Population genetic structure of an estuarine and a reef fish species exploited by brazilian artisanal fishing

    In this study, we used microsatellite markers to examine the genetic structures of Centropomus undecimalis (Bloch, 1792) and Epinephelus marginatus (Lowe, 1834) populations collected from artisanal fishing sites along a stretch of coastline in southeastern Brazil. Based on F-statistics, there was no significant genetic differentiation evident in any C. undecimalis samples (F-ST=0.012). However, Bayesian clustering, principal component analysis (PCA) and discriminant analysis of principal components (DAPC) results suggested that there were most likely two clusters, with no relation to geographic areas. The bottleneck results showed no significant values and the effective population sizes (Ne) for the two genetically differentiated groups were large and similar. In contrast, for E. marginatus populations, the microsatellite loci showed no population subdivisions. The F-ST value was low and non-significant (F-ST=0.008), a Bayesian analysis indicated one cluster, and a PCA showed that all samples from different geographical sites shared the same genetic structure. The bottleneck results exhibited significant differences, and a low Ne was observed.     Genome analysis to identify SNPs associated with oil content and fatty acid components in soybean

    The nutritional value, flavor and stability of soybean oil are determined by its five dominant fatty acids: saturated palmitic and stearic, monounsaturated oleic, and polyunsaturated linoleic and linolenic acids. Identifying molecular markers or quantitative trait loci associated with these components has the potential to facilitate the development of improved varieties and thus improve soybean oil content and quality. In this study, we used the BARCSoySNP6K BeadChip array to conduct a genome analysis of diverse soybean accessions evaluated for 2 years under Brazilian field conditions. The results demonstrated high broad-sense heritability, suggesting that the soybean genotype panel could be useful for oil trait breeding programs. Moreover, the range of oil trait variation among the plant introductions (PIs) was superior to that among the Brazilian cultivars in this study, indicating that a PI population could be used to find genes controlling these traits. The genome analysis showed that the genetic structure of the soybean germplasm comprised two main genetic groups, and it revealed linkage disequilibrium decay of approximately 300 kb. A total of 19 single-nucleotide polymorphism (SNP) loci on ten different chromosomes significantly associated with palmitic acid, oleic acid and total oil contents were discovered. Analysis of the SNP annotations revealed enzymes associated with several oil-related physiological metabolisms. Loci and specific alleles in our soybean panel that contributed to lower palmitic acid contents and higher oleic acid and total oil contents were identified.
