    A new approach to onset detection: towards an empirical grounding of theoretical and speculative ideologies of musical performance

    This article assesses aspects of the current state of a project which aims, with the help of computers and computer software, to segment soundfiles of vocal melodies into their component notes, identifying precisely when the onset of each note occurs, and then tracking the pitch trajectory of each note, especially in melodies employing a variety of non-standard temperaments, in which musical intervals smaller than 100 cents are ubiquitous. From there, we may proceed further, to describe many other “micro-features” of each of the notes, but for now our focus is on the onset times and pitch trajectories

    A microtonal wind controller building on Yamaha’s technology to facilitate the performance of music based on the “19-EDO” scale

    We describe a project in which several collaborators adapted an existing instrument to make it capable of playing expressively in music based on the microtonal scale characterised by equal divsion of the octave into 19 tones (“19-EDO”). Our objective was not just to build this instrument, however, but also to produce a well-formed piece of music which would exploit it idiomatically, in a performance which would provide listeners with a pleasurable and satisfying musical experience. Hence, consideration of the extent and limits of the playing-techniques of the resulting instrument (a “Wind-Controller”) and of appropriate approaches to the composition of music for it were an integral part of the project from the start. Moreover, the intention was also that the piece, though grounded in the musical characteristics of the 19-EDO scale, would nevertheless have a recognisable relationship with what Dimitri Tymoczko (2010) has called the “Extended Common Practice” of the last millennium. So the article goes on to consider these matters, and to present a score of the resulting new piece, annotated with comments documenting some of the performance issues which it raises. Thus, bringing the project to fruition involved elements of composition, performance, engineering and computing, and the article describes how such an inter-disciplinary, multi-disciplinary and cross-disciplinary collaboration was co-ordinated in a unified manner to achieve the envisaged outcome.     The near-shore, soft-substrate bivalves of the Swan Estuary: An assessment of their use as biological indicators

    The soft-substrate bivalves of the Swan Estuary were evaluated for their suitability as biological indicators. The major requirement of a biological indicator is that a response to changing conditions can be shown. Demonstrating that bivalve community and populations respond to changing environmental conditions within The Estuary would fulfill this criteria. To make this assessment the study conducted a quantitative sampling program which established the current structure of the near-shore soft-substrate bivalve community and this was compared with the community established in 1973-74. To evaluate the predictive potential of bivalve structure as an indicator, major environmental conditions in the Estuary were measured and related to bivalve distribution, abundance and biomass. TI1e utility of bivalves as indicator species in causal experiments was also assessed by means of a toxicity test conducted in the laboratory. Tills study found significant differences in the current bivalve community compared with that established in 1973-74. Three species have been added to the Middle Estuary community, two of which now dominate. Possible relationships between these changes and alterations to environmental conditions within the Estuary over the same period i.e nutrient enrichment and changes in hydrology, were examined but conclusions were speculative only. The current bivalve community appears to represent the major biotic divisions within The Estuary and relationships were found with the presence of seagrass, phytoplankton biomass, fine particulate organic matter and sediment grain-size composition. A causal relationship to the toxicity of chromium was demonstrated in the laboratory. The processes of measuring population and community structure, and of handling and manipulating bivalves both in the field and the laboratory were shown to be time- and cost-efficient. Their ease of sampling, abundance and widespread distribution affords bivalves potential as biological indicators within the Swan Estuary

    Developing a Culture of Publication: a joint enterprise writing retreat

    Purpose: Many students irrespective of level of study produce excellent course work which, if given support and encouragement could clearly be of a publishable standard. Academic staff are expected to produce quality publications meeting peer review standards although they may be relatively novice authors. All are engaged in some aspects of academic writing practices but not as frequently involved in co-production of publications emanating from student work. This activity is still at the margins of much of the student experience. Design/methodology: Mindful of these issues, we designed and offered a writing programme including a writing retreat. This brought together undergraduate and postgraduate students from a range of applied disciplines (health and art, design and architecture) and their supervisors with the aim of co-producing publications and participating in a community of scholarly practice. The project was delivered over nine months. It involved four days ‘compulsory’ attendance and included a preparatory workshop, a two day off campus writing retreat and a dissemination event. Student and supervisors applied to participate as a team. Kirkpatrick’s (2006) four-stage classic model: reaction, learning, changes in behaviour and real world results was used as a framework for the educational evaluation. Key findings organised thematically were: Supervisor-supervisee relationships; space and time; building confidence enabling successful writing and publication. Originality/Value: This paper will provide an overview of the design, content and approaches used for successful delivery of this innovative project. It will draw on examples that illustrate the different types of joint enterprise that emerged, illuminate experiences of co-production and co-authorship along with recommendations for future ventures

    Access to Justice for All: Towards an \u27Expansive Vision\u27 of Justice and Technology

    The Canadian dialogue regarding access to justice has taken an important turn in the last few years, more robustly conceptualizing what is to be accessed (“deliverables”) and who is intended to benefit (“beneficiaries”), as well as recognizing that access to justice initiatives that benefit some citizens or groups of citizens cannot be presumed to benefit all citizens. Recent initiatives shift focus away from particular kinds of deliverables (e.g. access to lawyers and courts) and/or particular groups of beneficiaries (e.g. the middle class) toward a more “expansive vision” of access to justice. The expansive vision not only integrates and prioritizes a variety of deliverables, but also explicitly recognizes that socioeconomic and other structural differences among citizens affect their respective abilities to benefit both from the justice system itself and from initiatives designed to improve access to justice. These developments hold important promise not just for responding to what has been called an access to justice “crisis” in Canada, but also for tailoring responses in ways that are mindful of the differing needs and situations of all citizens. In this paper, and in keeping with this expansive vision, we argue that this more robust approach to access to justice must also be brought to bear on the specific dialogue around access to justice and technology

    Structure-function analysis of the Bacillus megaterium GerUD spore germinant receptor protein.

    Germination of Bacillus spores is triggered by the interaction of germinant molecules with specialized receptor proteins localized to the spore inner membrane. Germinant receptors (GRs) are comprised typically of three interacting protein subunits, each of which is essential for receptor function. At least some GRs appear to have a fourth component, referred to as a D-subunit protein. A number of D-subunit proteins were shown previously to be capable of modulating the activity of associated GRs. Here, we investigate the topology and structure-function relationships of the Bacillus megaterium QM B1551 GerUD protein, which is associated with the GerU GR. The presented data demonstrate that GerUD can be subjected to relatively extensive structural modifications while retaining function. Indeed, the presence of either of the two transmembrane spanning domains is sufficient to modulate an efficient GerU-mediated germinative response. The precise function of D-subunit proteins has yet to be established, although they may act as molecular chaperones within the spore inner-membrane environment.S.G was the recipient of a RA Fisher bursary award from Gonville and Caius College, University of Cambridge.     Structure and functional motifs of GCR1, the only plant protein with a GPCR fold?

    Whether GPCRs exist in plants is a fundamental biological question. Interest in deorphanizing new G protein coupled receptors (GPCRs), arises because of their importance in signaling. Within plants, this is controversial as genome analysis has identified 56 putative GPCRs, including GCR1 which is reportedly a remote homologue to class A, B and E GPCRs. Of these, GCR2, is not a GPCR; more recently it has been proposed that none are, not even GCR1. We have addressed this disparity between genome analysis and biological evidence through a structural bioinformatics study, involving fold recognition methods, from which only GCR1 emerges as a strong candidate. To further probe GCR1, we have developed a novel helix alignment method, which has been benchmarked against the the class A – class B - class F GPCR alignments. In addition, we have presented a mutually consistent set of alignments of GCR1 homologues to class A, class B and class F GPCRs, and shown that GCR1 is closer to class A and /or class B GPCRs than class A, class B or class F GPCRs are to each other. To further probe GCR1, we have aligned transmembrane helix 3 of GCR1 to each of the 6 GPCR classes. Variability comparisons provide additional evidence that GCR1 homologues have the GPCR fold. From the alignments and a GCR1 comparative model we have identified motifs that are common to GCR1, class A, B and E GPCRs.     Differential gene expression in multiple neurological, inflammatory and connective tissue pathways in a spontaneous model of human small vessel stroke

    Aims: Cerebral small vessel disease (SVD) causes a fifth of all strokes plus diffuse brain damage leading to cognitive decline, physical disabilities and dementia. The aetiology and pathogenesis of SVD are unknown, but largely attributed to hypertension or microatheroma. Methods: We used the spontaneously hypertensive stroke-prone rat (SHRSP), the closest spontaneous experimental model of human SVD, and age-matched control rats kept under identical, non-salt-loaded conditions, to perform a blinded analysis of mRNA microarray, qRT-PCRand pathway analysis in two brain regions (frontal and midcoronal) commonly affected by SVD in the SHRSP at age five, 16 and 21 weeks. Results: We found gene expression abnormalities, with fold changes ranging from 2.5 to 59 for the 10 most differentially expressed genes, related to endothelial tight junctions (reduced), nitric oxide bioavailability (reduced), myelination (impaired), glial and microglial activity (increased), matrix proteins (impaired), vascular reactivity (impaired) and albumin (reduced), consistent with protein expression defects in the same rats. All were present at age 5 weeks thus pre-dating blood pressure elevation. ‘Neurological’ and ‘inflammatory’ pathways were more affected than ‘vascular’ functional pathways. Conclusions: This set of defects, although individually modest, when acting in combination could explain the SHRSP's susceptibility to microvascular and brain injury, compared with control rats. Similar combined, individually modest, but multiple neurovascular unit defects, could explain susceptibility to spontaneous human SVD
