    Infliximab therapy in rheumatoid arthritis and ankylosing spondylitis-induced specific antinuclear and antiphospholipid autoantibodies without autoimmune clinical manifestations: a two-year prospective study

    Treatment of rheumatoid arthritis (RA) with infliximab (Remicade(®)) has been associated with the induction of antinuclear autoantibodies (ANA) and anti-double-stranded DNA (anti-dsDNA) autoantibodies. In the present study we investigated the humoral immune response induced by infliximab against organ-specific or non-organ-specific antigens not only in RA patients but also in patients with ankylosing spondylitis (AS) during a two-year followup. The association between the presence of autoantibodies and clinical manifestations was then examined. The occurrence of the various autoantibodies was analyzed in 24 RA and 15 AS patients all treated with infliximab and in 30 RA patients receiving methotrexate but not infliximab, using the appropriate methods of detection. Infliximab led to a significant induction of ANA and anti-dsDNA autoantibodies in 86.7% and 57% of RA patients and in 85% and 31% of AS patients, respectively. The incidence of antiphospholipid (aPL) autoantibodies was significantly higher in both RA patients (21%) and AS patients (27%) than in the control group. Most anti-dsDNA and aPL autoantibodies were of IgM isotype and were not associated with infusion side effects, lupus-like manifestations or infectious disease. No other autoantibodies were shown to be induced by the treatment. Our results confirmed the occurrence of ANA and anti-dsDNA autoantibodies and demonstrated that the induction of ANA, anti-dsDNA and aPL autoantibodies is related to infliximab treatment in both RA and AS, with no significant relationship to clinical manifestations

    Abstractα1-Acid glycoprotein (AGP) was purified to homogeneity by a 3-step procedure using pseudo-ligand affinity chromatography on immobilized Cibacron blue F3GA, Procion red HE3B, and preparative column isoelectric focusing. The overall yield of the combined techniques was 88%. Analysis of the purified AGP by lectin affinity chromatography on immobilized Con A and immunoaffinoelectrophoresis indicated that the most acidic form did not interact with the lectin, while the two more basic fractions possessed different affinities for Con A. In addition, 3 different populations of AGP were clearly separated by Con A affinity chromatography

    Diversity of farming systems integrating fish pond aquaculture in the province of Kinshasa in the Democratic Republic of the Congo

    Agriculture and aquaculture systems are used by many farmers in various tropical countries of Asia, America and Africa. They have proven their relevancy to increase the productivity of farms by optimising nutrient fluxes and reducing requirements for external fertilisers. This article analysed the current state of fish farming and the way it is integrated with other farm subsystems in the urban/peri-urban and rural areas of Kinshasa, Democratic Republic of Congo. More precisely, it examined the allocation of resources at the farm level, the recovery of helophytes plants, and the fate of fish production choices and it explored the possibility of intensifying these existing integrated farming systems. After a census of ponds in the urban and rural areas of Kinshasa, an on-site survey was conducted on 150 fish pond farms to assess the different activities practiced on farms, the impact of integrating crops and livestock to fish pond aquaculture and the constraints of the system. A total of three thousand and twenty (3020) fish ponds were recorded in the urban and rural areas of Kinshasa. Among these farms integrated aquaculture-agriculture systems exist with a wide diversity of practices (about 79% of farms combined fish with livestock and/or vegetable production). No striking differences between fish farms according to the allocation of resources, fish production method such as monoculture or polyculture, the recovery of helophytes plants and the fate of fish production choice were found depending on the location. However, fish farms were differently managed when combined with agriculture and/or livestock. Regarding the integration of the different subsystems through nutrient fluxes, 11 different movements of material between subsystems were found in integrated farms. However, not all fluxes are equally used in all farms and therefore improvements cannot be generalised. Improvements to be explored are such as making better use of manure pond mud and helophyte plants. For this purpose, proper training of farmers might be critical. Finally, bringing farmers together in cooperatives could also contribute to reduce the cost of purchase and transportation of fish fry and feed

    Synaptic Maturation at Cortical Projections to the Lateral Amygdala in a Mouse Model of Rett Syndrome

    Rett syndrome (RTT) is a neuro-developmental disorder caused by loss of function of Mecp2 - methyl-CpG-binding protein 2 - an epigenetic factor controlling DNA transcription. In mice, removal of Mecp2 in the forebrain recapitulates most of behavioral deficits found in global Mecp2 deficient mice, including amygdala-related hyper-anxiety and lack of social interaction, pointing a role of Mecp2 in emotional learning. Yet very little is known about the establishment and maintenance of synaptic function in the adult amygdala and the role of Mecp2 in these processes. Here, we performed a longitudinal examination of synaptic properties at excitatory projections to principal cells of the lateral nucleus of the amygdala (LA) in Mecp2 mutant mice and their wild-type littermates. We first show that during animal life, Cortico-LA projections switch from a tonic to a phasic mode, whereas Thalamo-LA synapses are phasic at all ages. In parallel, we observed a specific elimination of Cortico-LA synapses and a decrease in their ability of generating presynaptic long term potentiation. In absence of Mecp2, both synaptic maturation and synaptic elimination were exaggerated albeit still specific to cortical projections. Surprisingly, associative LTP was unaffected at Mecp2 deficient synapses suggesting that synaptic maintenance rather than activity-dependent synaptic learning may be causal in RTT physiopathology. Finally, because the timing of synaptic evolution was preserved, we propose that some of the developmental effects of Mecp2 may be exerted within an endogenous program and restricted to synapses which maturate during animal life

    Development of innovative technology reducing the levels of Polycyclic Aromatic Hydrocarbons in grilled meat and smoked fish

Recent studies revealed that traditional equipments led to high contamination of end-products by polycyclic aromatic hydrocarbons (PAHs). This study aimed to assess the performances of new equipments and reduce the level of PAHs in end-products. Thirty-six controlled production trials were performed using three improved and new equipments in combination with charcoal and wood as fuel. The temperature at the core of grilled pork (77.5 ± 3.8 °C) and smoked fish (76.4 ± 2.8 °C) showed the highest thermal efficiency of QUALISANI equipment (FAQ) used with charcoal compared to other combinations fuel-equipment. The benzo[a]pyrene (BaP) (0.2 ± 0.1–1.9 ± 2.1 µg/Kg) and sum of PAH4 (2.4 ± 0.2 µg/Kg) in grilled pork obtained with FAQ-charcoal used with or without filter were the lowest levels of PAHs in compliance with Beninese and European standards. Smoked and smoked-dried fishes obtained from FAQ-charcoal with or without filter, FAQ-wood-filter, and the modified barrel kiln (FBM) used with charcoal-filter had ranges of BaP (0.1–0.2 µg/Kg) and PAH4 (0.4–2.9 µg/Kg) significantly lower than the maximal limited values of 2 µg/Kg (BaP) and 12 µg/Kg (PAH4) admitted by European Commission
