    Physico-chemical, proximate, sensory and storage quality attributes analysis of Papaver somniferum (poppy) fortified chevon nuggets

    Papaver somniferum(Poppy) seeds contain poly-phenols like tannic acid, ellagitannin that act as antioxidant, fat replacer, sedative, analgesic and anti-tussive properties, disease preventing and health promoting proper-ties. Efficacy of ground P. somniferum poppy seed was used in chevon nuggets was analyzed. The use of ground poppy seed in chevon nuggets formulation had no effect on moisture as well as pH content of poppy seed incorporated chevon nuggets. However, ground poppy seed incorporation in chevon nuggets had significantly (p<0.05) lower fat content, higher protein content, higher emulsion stability and subsequently higher cooking yield. Chevon nuggets having 10% ground poppy seed were having significantly (p<0.05) higher sensory scores. The ground poppy seed treated chevon nuggets were having significantly (p<0.05) lower TBA and FFA value. The microbial load of developed product was significantly (p<0.05) lower during refrigeration storage. The chevon nuggets prepared with fortification of ground poppy seed was found to be suitable for consumption till 21st day during refrigeration storage based on TBA, FFA, microbiological and sensory profile. Thus, chevon nuggets with good to very good acceptability were developed with incorporation of ground poppy (P. somniferum) seed in it. The chevon nuggets developed consisted of higher amount of antioxidants and polyunsaturated fatty acid with better sensory scores and longer shelf-life

    Extrahepatic undifferentiated embryonal sarcoma, cytology as an accurate method for early diagnosis: an extremely rare case report in an Indian child

    Undifferentiated embryonal sarcoma has been described in the liver, a rare malignant mesenchymal neoplasm, that occurs primarily in children and teenagers. Approximately 260 cases have been reported arising in the liver since 1978 when this disease was first described. Its pathogenesis is still obscure. Authors presented a case of extrahepatic undifferentiated embryonal sarcoma in a 9-year-old female presenting with upper abdominal dull pain. Ultrasound and CT Scan showed normal liver architecture, with liver pushed upwards due to compression by tumor arising in the retroperitoneum. To the best of our knowledge, this is first case of extrahepatic undifferentiated embryonal sarcoma diagnosed on cytomorphology and confirmed by histopathology and immunohistochemistry markers

    Disaster Preparedness Information Needs of Individuals Attending an Adult Literacy Center : An Exploratory Study

    Being prepared with accurate, credible, and timely information during a disaster can help individuals make informed decisions about taking appropriate actions. Unfortunately, many people have difficulty understanding health and risk-related resources. This exploratory, mixed methods study assessed disaster information-seeking behaviors and comprehension of public health disaster preparedness resources by individuals at an adult literacy center. A convenience pilot sample of 20 adult learners (mean age: 53.1) was recruited. Health literacy was assessed using Newest Vital Sign (NVS) and modified Cloze (multiple choice) tests on biological terrorism and avian influenza information. In-person interviews were conducted to determine participants’ knowledge, perceptions, and information needs about disasters. Thematic analysis of interviews was conducted using NVivo7. Mean NVS was 3.11/6.00 implying limited health literacy. Mean Cloze scores revealed marginal disaster comprehension (avian flu: .46/1.00; biological terrorism: .48/1.00). Over half of participants with inadequate Cloze comprehension self-rated their understanding as “good.” Key themes emerging from interviews were: multiple perceptions about disasters, limited access to preparedness resources, need for visuals and plain language information, and importance of knowing where to go during a disaster. Study findings advocate for multimedia, plain language, and visual communication to influence adult learners’ literacy practices and self-efficacy in interpreting instructions and acting appropriately in preparing for and responding to disasters

    The Enhanced Liver Fibrosis test maintains its diagnostic and prognostic performance in alcohol-related liver disease: a cohort study

    BACKGROUND: Alcohol is the main cause of chronic liver disease. The Enhanced Liver Fibrosis (ELF) test is a serological biomarker for fibrosis staging in chronic liver disease, however its utility in alcohol-related liver disease warrants further validation. We assessed the diagnostic and prognostic performance of ELF in alcohol-related liver disease. METHODS: Observational cohort study assessing paired ELF and histology from 786 tertiary care patients with chronic liver disease due to alcohol (n = 81) and non-alcohol aetiologies (n = 705). Prognostic data were available for 64 alcohol patients for a median of 6.4 years. Multiple ELF cut-offs were assessed to determine diagnostic utility in moderate fibrosis and cirrhosis. Survival data were assessed to determine the ability of ELF to predict liver related events and all-cause mortality. RESULTS: ELF identified cirrhosis and moderate fibrosis in alcohol-related liver disease independently of aminotransferase levels with areas under receiver operating characteristic curves of 0.895 (95% CI 0.823-0.968) and 0.923 (95% CI 0.866-0.981) respectively, which were non-inferior to non-alcohol aetiologies. The overall performance of ELF was assessed using the Obuchowski method: in alcohol = 0.934 (95% CI 0.908-0.960); non-alcohol = 0.907 (95% CI 0.895-0.919). Using ELF < 9.8 to exclude and ≧ 10.5 to diagnose cirrhosis, 87.7% of alcohol cases could have avoided biopsy, with sensitivity of 91% and specificity of 85%. A one-unit increase in ELF was associated with a 2.6 (95% CI 1.55-4.31, p < 0.001) fold greater odds of cirrhosis at baseline and 2.0-fold greater risk of a liver related event within 6 years (95% CI 1.39-2.99, p < 0.001). CONCLUSIONS: ELF accurately stages liver fibrosis independently of transaminase elevations as a marker of inflammation and has superior prognostic performance to biopsy in alcohol-related liver disease

    Electronic Cigarettes As a Smoking-Cessation Tool Results from an Online Survey

    Background: Electronic cigarettes (e-cigarettes) are battery-powered devices that deliver nicotine without any combustion or smoke. These devices have generated much publicity among the smoking-cessation community and support from dedicated users; however, little is known about the effıcacy of the device as a smoking-cessation tool. Purpose: This study aimed to examine the effectiveness of e-cigarettes for smoking cessation using a survey of smokers who had tried e-cigarettes. Methods: Using as a sampling frame a cohort of all fırst-time purchasers of a particular brand of e-cigarettes during a 2-week period, a cross-sectional, online survey was conducted in 2010 to describe e-cigarette use patterns and their effectiveness as a smoking-cessation tool. There were 222 respondents, with a survey response rate of 4.5%. The primary outcome variable was the point prevalence of smoking abstinence at 6 months after initial e-cigarette purchase. Results: The primary fınding was that the 6-month point prevalence of smoking abstinence among the e-cigarette users in the sample was 31.0% (95% CIϭ24.8%, 37.2%). A large percentage of respondents reported a reduction in the number of cigarettes they smoked (66.8%) and almost half reported abstinence from smoking for a period of time (48.8%). Those respondents using e-cigarettes more than 20 times per day had a quit rate of 70.0%. Of respondents who were not smoking at 6 months, 34.3% were not using e-cigarettes or any nicotine-containing products at the time. Conclusions: Findings suggest that e-cigarettes may hold promise as a smoking-cessation method and that they are worthy of further study using more-rigorous research designs.     LncRNA VEAL2 regulates PRKCB2 to modulate endothelial permeability in diabetic retinopathy

    Long non‐coding RNAs (lncRNAs) are emerging as key regulators of endothelial cell function. Here, we investigated the role of a novel vascular endothelial‐associated lncRNA (VEAL2) in regulating endothelial permeability. Precise editing of veal2 loci in zebrafish (veal2 (gib005Δ8/+)) induced cranial hemorrhage. In vitro and in vivo studies revealed that veal2 competes with diacylglycerol for interaction with protein kinase C beta‐b (Prkcbb) and regulates its kinase activity. Using PRKCB2 as bait, we identified functional ortholog of veal2 in humans from HUVECs and named it as VEAL2. Overexpression and knockdown of VEAL2 affected tubulogenesis and permeability in HUVECs. VEAL2 was differentially expressed in choroid tissue in eye and blood from patients with diabetic retinopathy, a disease where PRKCB2 is known to be hyperactivated. Further, VEAL2 could rescue the effects of PRKCB2‐mediated turnover of endothelial junctional proteins thus reducing hyperpermeability in hyperglycemic HUVEC model of diabetic retinopathy. Based on evidence from zebrafish and hyperglycemic HUVEC models and diabetic retinopathy patients, we report a hitherto unknown VEAL2 lncRNA‐mediated regulation of PRKCB2, for modulating junctional dynamics and maintenance of endothelial permeability

    Integrated Pest Management (IPM) for Reducing Pesticide Residues in Crops and Natural Resources

    Investigation on the pesticide residues during 2006–2009 in various crops and natural resources (soil and water) in the study village (Kothapally, Telangana State (TS)) indicated the presence of a wide range of insecticidal residues. Pooled data of the 80 food crop and cotton samples, two rice grain samples (3 %) showed beta endosulfan residues, and two (3 %) soil samples showed alpha and beta endosulfan residues. In vegetables of the 75 tomato samples, 26 (35 %) were found contaminated with residues of which 4 % had residues above MRLs. Among the 80 brinjal samples, 46 (56 %) had residues, of these 4 % samples had residues above MRLs. Only 13 soil samples from vegetable fields were found contaminated. The frequency of contamination in brinjal fields was high and none of the pulses and cotton samples revealed any pesticide contamination. IPM fields showed substantial reduction sprays which in-turn reflected in lower residues. Initial studies on water analysis indicated the presence of residues in all water sources with higher in bore wells compared to open wells, however, by 2009 the water bodies reflected no residues above the detectable level

    Gene Bionetwork Analysis of Ovarian Primordial Follicle Development

    Ovarian primordial follicles are critical for female reproduction and comprise a finite pool of gametes arrested in development. A systems biology approach was used to identify regulatory gene networks essential for primordial follicle development. Transcriptional responses to eight different growth factors known to influence primordial follicles were used to construct a bionetwork of regulatory genes involved in rat primordial follicle development. Over 1,500 genes were found to be regulated by the various growth factors and a network analysis identified critical gene modules involved in a number of signaling pathways and cellular processes. A set of 55 genes was identified as potential critical regulators of these gene modules, and a sub-network associated with development was determined. Within the network two previously identified regulatory genes were confirmed (i.e., Pdgfa and Fgfr2) and a new factor was identified, connective tissue growth factor (CTGF). CTGF was tested in ovarian organ cultures and found to stimulate primordial follicle development. Therefore, the relevant gene network associated with primordial follicle development was validated and the critical genes and pathways involved in this process were identified. This is one of the first applications of network analysis to a normal developmental process. These observations provide insights into potential therapeutic targets for preventing ovarian disease and promoting female reproduction

    Preventive Effects of Omega-3 and Omega-6 Fatty Acids on Peroxide Mediated Oxidative Stress Responses in Primary Human Trabecular Meshwork Cells

    Pathologic processes in glaucoma include increased apoptosis, accumulation of extracellular material in the trabecular meshwork and optic nerve, condensations of the cytoskeleton and precocious cellular senescence. Oxidative stress was shown to generate these alterations in primary ocular cells. Fatty acids omega-3 and -6 are alleged to constitute a prophylaxis against these deleterious effects. Here, we tested actual preventive effects omega-3 and -6 against peroxide induced stress responses in primary human trabecular meshwork cells. Changes of mitochondrial activity, proliferation, heat shock proteins, extracellular matrix components, and inflammatory markers were evaluated. Alterations of the cytoskeleton were evaluated by phalloidin labeling. Here we report a repressive effect of omega-6 on metabolic activity and proliferation, which was not detected for omega-3. Both agents were able to prevent the anti-proliferative effect of H2O2, but only omega-3 prevented metabolic repression. Expression of heat shock protein 27 was unaltered by both fatty acids, whereas heat shock protein 90 was significantly induced by both. Omega-6 increased fibronectin and connective tissue growth factor synthesis, as well as the amount of secreted fibronectin. Omega-3, instead, induced plasminogen activator inhibitor 1 synthesis. H2O2 further increased fibronectin production in omega-6 supplemented cells, which was not the case in omega-3 treated cells. H2O2 stimulation of plasminogen activator inhibitor 1 and connective tissue growth factor was repressed by both fatty acids. Both fatty acids appeared to abolish H2O2 mediated stimulation of nuclear factor κB and IL-6, but not IL-1α and IL-8. H2O2 induced formation of cross-linked actin networks and stress fibers, which was reduced by preemptive application of omega-3. Omega-6, in contrast, had no protective effect on that, and even seemed to promote condensation. Based on the observed side effects of omega-6, omega-3 appears to be the more beneficial fatty acid in respect of prophylactic intake for prevention of a glaucomatous disease
