    Herbal medicine IMOD suppresses LPS-induced production of proinflammatory cytokines in human dendritic cells

    Traditional medicines that stimulate or modulate the immune system can be used as innovative approaches to treat immunological diseases. The herbal medicine IMOD has been shown to strongly modulate immune responses in several animal studies as well as in clinical trials. However, little is known about the mechanisms of IMOD to modulate immunity. Here we have investigated whether IMOD modulates the immunological function of human dendritic cells (DCs). IMOD alone did not induce DC maturation nor production of cytokines. Notably, IMOD decreased the production of pro-inflammatory cytokines IL-6, IL-12 p70 and TNFα by LPS-activated DCs at both mRNA and protein levels in a dose dependent manner. In contrast, treatment with IMOD did not affect LPS induced-production of the anti-inflammatory cytokine IL-10. Furthermore, IMOD inhibited T cell activation/proliferation by LPS-treated DCs and skewed T-cells responses towards the T helper type 2 polarization. These data strongly indicate that IMOD has a potent immunomodulatory ability that affects TLR signaling and thereby modulates DC function. Insight into the immunomodulatory effect of herbal medicine IMOD may provide innovative strategies to affect the immune system and to help combat various disease

    Discovery of the neutron star nature of SLX 1737-282

    SLX 1737-282 is a persistent moderately bright X-ray source 1.2 deg from the Galactic center. X-ray observations with the Wide Field Cameras on BeppoSAX have for the first time revealed the true nature of SLX 1737-282: a 15-min long thermonuclear flash was detected exposing the source as a neutron star in a binary system. The flash was Eddington-limited constraining the distance to between 5 and 8 kpc. We analyze BeppoSAX, ROSAT, and RXTE data on SLX 1737-282. The persistent 0.5-200 keV luminosity is close to or less than 1% of the Eddington limit which implies a rarely-seen circumstance for thermonuclear flash activity.Comment: Accepted for publication in A&A Letter

    Fast X-ray Transients and Their Connection to Gamma-Ray Bursts

    Fast X-ray transients (FXTs) with timescales from seconds to hours have been seen by numerous space instruments. We have assembled archival data from Ariel-5, HEAO-1 (A-1 and A-2), WATCH, ROSAT, and Einstein to produce a global fluence-frequency relationship for these events. Fitting the log N-log S distribution over several orders of magnitude to simple power law we find a slope of -1.0. The sources of FXTs are undoubtedly heterogeneous, the -1 power law is an approximate result of the summation of these multiple sources. Two major contributions come from gamma-ray bursts and stellar flares. Extrapolating from the BATSE catalog of GRBs, we find that the fraction of X-ray flashes that can be the X-ray counterparts of gamma-ray bursts is a function of fluence. Certainly most FXTs are not counterparts of standard gamma-ray bursts. The fraction of FXTs from non-GRB sources, such as magnetic stars, is greatest for the faintest FXTs. Our understanding of the FXT phenomenon remains limited and would greatly benefit from a large, homogeneous data set, which requires a wide-field, sensitive instrument.Comment: 36 pages, 8 figure

    Dimensional Implicit Memory Priming Deficits in Young ADHD Adults

    The experiment explored the difference between production and identification processes and conceptual and perceptual processes in long-term implicit memory. The first phase consisted of prescreening ADHD (Attention Deficit Hyperactivity Disorder) and non- ADHD Texas A&M undergraduate students. The second phase consisted of a study task and a filler task. The third phase consisted of four tests containing two of the four processes. The participants performed a category verification test (conceptualidentification cross), category generation test (conceptual-production cross), a flashing words test (perceptual-identification cross), and a stem completion test (perceptualproduction cross). No statistically significant difference in priming was found between ADHD and non-ADHD. However, looking at the sample as a whole (ADHD and non- ADHD combine), there was significant priming in the category generation test, flashing words test, and the stem completion test but not in the category verification test

    A randomized controlled trial comparing tibial migration of the ATTUNE cemented cruciate-retaining knee prosthesis with the PFC-sigma design

    AimsThe primary objective of this study was to compare migration of the cemented ATTUNE fixed bearing cruciate retaining tibial component with the cemented Press-Fit Condylar (PFC)-sigma fixed bearing cruciate retaining tibial component. The secondary objectives included comparing clinical and radiological outcomes and Patient Reported Outcome Measures (PROMs).MethodsA single blinded randomized, non-inferiority study was conducted including 74 patients. Radiostereometry examinations were made after weight bearing, but before hospital discharge, and at three, six, 12, and 24 months postoperatively. PROMS were collected preoperatively and at three, six, 12, and 24 months postoperatively. Radiographs for measuring radiolucencies were collected at two weeks and two years postoperatively.ResultsThe overall migration (mean maximum total point motion (MPTM)) at two years was comparable: mean 1.13 mm (95% confidence interval (CI), 0.97 to 1.30) for the ATTUNE and 1.16 mm (95% CI, 0.99 to 1.35) for the PFC-sigma. At two years, the mean backward tilting was -0.43 degrees (95% CI, -0.65 to -0.21) for the ATTUNE and 0.08 degrees (95% CI -0.16 to 0.31), for the PFC-sigma. Overall migration between the first and second postoperative year was negligible for both components.The clinical outcomes and PROMs improved compared with preoperative scores and were not different between groups. Radiolucencies at the implant-cement interface were mainly seen below the medial baseplate: 17% in the ATTUNE and 3% in the PFC-sigma at two weeks, and at two years 42% and 9% respectively (p = 0.001).ConclusionIn the first two postoperative years the initial version of the ATTUNE tibial component was not inferior with respect to overall migration, although it showed relatively more backwards tilting and radiolucent lines at the implant-cement interface than the PFC-sigma.     The resolution of racemates through their diastereomeric salts can be positively affected by the addition of small amounts of suitable nucleation inhibitors. This discovery is a logical extension of “Dutch Resolution”, in which equimolar amounts of resolving agents that are members of the same family (i.e., structurally related) are used. We conducted a systematic search for nucleation inhibitors of the resolving agent 1-phenylethylamine. A wide range of amines that bear possible family resemblances to 1-phenylethylamine was investigated. It was found that (R)-1-phenylbutylamine is a good inhibitor of (R)-1-phenylethylamine. Results of turbidity measurements showed that, for the model case of mandelic acid resolution, the chief effect of this inhibitor was to widen the metastable zone for the more soluble diastereomer. This observation is in accordance with previous experience. Further scouting for possible family members revealed a wide variation in the effectiveness of inhibitors, dependent on their structure. By far the most effective inhibitors are bifunctional 1-phenylethylamine and/or 1-phenylbutylamine analogues. The effect of racemic inhibitors was found to approach that of enantiomerically pure inhibitors of the same absolute configuration of the 1-phenylethylamine used for resolution. The most effective inhibitors were tested for the resolution of a structural variety of racemates, and were shown to be broadly applicable.
