    Antihypertrophic effect of hemin in deoxycorticosterone acetate-salt-induced hypertensive rat model

    The application of the synthetic mineralocorticoid, deoxycorticosterone acetate (DOCA)-salt, to unilaterally nephrectomised rats induces severe hypertension due to volume-overload, and mimics human primary aldosteronism. Importantly, DOCA-salt hypertension is characterized by severe cardiac and renal lesions triggered by nuclear factor kappa B (NF-kappaB), activating protein (AP-1), and transforming growth factor beta1 (TGF-beta1) leading to end-stage organ damage. Although DOCA-salt hypertension is a low renin model, local production of angiotensin-II and aldosterone in cardiac and renal tissues stimulate TGF-beta1, fibronectin and collagen-1 causing fibrosis and hypertrophy. Since TGF-beta1 gene promoter contains binding sites for NF-kappaB and AP-1, cross-talk between TGF-beta1, NF-kappaBƒnand AP-1 can be envisaged. Accordingly, the activation of TGF-beta1, fibronectin, collagen, NF-kappaB and AP-1 may constitute a potent destructive force in hypertension. Emerging evidence indicates that upregulation of the heme oxygenase (HO) system is cytoprotective with antioxidant, antihypertensive and antihypertrophic effects. Interestingly, the promoter region of HO-1 gene harbors consensus-binding sites for NF-kappaB and AP-1; therefore, the HO system may regulate these transcription factors to counteract tissue insults. However, the multifaceted interactions between the HO system, NF-kappaB, AP-1, TGF-beta1, fibronectin and collagen in mineralocorticoid-induced end-stage-organ damage have not been fully characterized. Similarly, the effect of the HO system on tissue angiotensin-II and aldosterone levels in mineralocorticoid-induced hypertension remains unclear. Therefore, the present study was designed to investigate the antihypertrophic effect of the HO system in cardiac and renal tissue of DOCA-salt hypertensive rats. In this study, the HO inducer, hemin, lowered blood pressure and attenuated cardiac/renal hypertrophy, whereas the HO inhibitor, chromium mesoporphyrin (CrMP), nullified the effects of hemin and exacerbated cardiac/renal injury the DOCA-salt hypertensive rats. The protective effect of hemin was associated with increased HO-1, HO activity, cyclic guanosine monophosphate (cGMP), superoxide dismutase activity, ferritin and the total antioxidant capacity in the cardiac and renal tissue. In contrast, angiotensin-II, aldosterone, 8-isoprostane, NF-kappaB and AP-1 were significantly downregulated. Furthermore, hemin therapy attenuated TGF-beta1 and extracellular matrix (ECM) proteins such as fibronectin and collagen, with corresponding reduction of cardiac histopathological lesions, including longitudinal/cross-sectional muscle fiber thickness, scarring, muscular hypertrophy, coronary arteriolar thickening and collagen deposition. Similarly, hemin attenuated structural lesions in the kidney such as glomerular hypertrophy, glomerular sclerosis, mononuclear cell infiltration, tubular cast formation, tubular dilation and renal arteriolar thickening with concomitant improvement of kidney function as evidenced by reduction of plasma creatinine, proteinuria, but enhanced creatinine clearance. Collectively, these results suggest that the HO system suppressed hypertension, cardiac and renal fibrosis, and hypertrophy in the DOCA-salt hypertensive rat by downregulating transcription factors such as NF-kappaB and AP-1, reducing ECM proteins such as fibronectin and collagen, decreasing local tissue production of angiotensin-II and aldosterone, and improved renal functional capacity

    Extending Shelf-life of Different Cut-flowers under Cold Room Conditions

    Uniform and healthy Rose cv. ‘Dutch'; Gerbera cv. ‘Lexington'; Gladiolus cv. ‘Top Secrate'; Tuberose cv. ‘Bizet' and Carnation cv. ‘Liberty' were used for the study in September 2016. Cut flowers were harvested at 7.00 am at proper stage, transported within 1.30 hours by AC car to the Agricultural Research Laboratory of Ecofrost Technologies Pvt. Ltd., Pune and then immediately prepared for post-harvest treatment and storage. The aim of this study was to determine the effectiveness of different storage conditions, i.e. room and cold storage conditions (10°C + 93 % RH) on the longevity of the cut flowers. The two treatments viz., holding flowers at room temperature (RT) (T1) and at cold room conditions (T2), were replicated twice. The result showed that keeping cut-flowers at cold storage in a holding-solution of tap water recorded the maximum storage-life (days) compared to room conditions

    The geometry of the 2-nitroso-1,naphthol in solid state and 1-2 naphthoquinine-2, oxime in solution have studied by employing Hatree Fock ab initio calculations using 6-31 G* level. IR wave number of 2-nitroso -1, naphthol is calculated and compared with experimental date. Further, 1-2, naphthaquinine-2, oxime in solution, NMR chemical shifts of 1H and 13C are calculated by HF method and 6.31 G* level, experimental data was compared.     A bridge project from its conception to completion involves various stages such as planning, design, approval/sanction, tendering and execution. Also inspections, maintenance & repairs are continuing activities for enhancing the service life of the structure. The selection of superstructure type for bridges plays a vital role to execute the project. The time and cost factor for completion of the bridges shall have great impact on finalization of the superstructure type. The type of superstructure has numbers of aspects in line with the design constraints, geometrical aspects, existing features of old bridges, etc. The standard specifications for design also play a key role to freeze the type of superstructure. The basic idea of study is to increase the cost-effectiveness of bridges by increasing their durability (i.e., useful life). There may be many ways to achieve more durable, less expensive and rapidly constructed structures, however, the methods for execution matters, in cost point of view. A proper method selected will help to save in time and cost of the work. Recently, there has been increased interest in constructing bridges that last longer, are less expensive, and take less time to construct. The results of the study are evaluated for different effective spans by considering support conditions, constituent materials, casting/fabrication methodologies etc. to reach at best economical option for superstructure of the river bridge

    The design and maintenance of an aero-engine generates a significant amount of documentation. When designing new engines, engineers must obtain knowledge gained from maintenance of existing engines to identify possible areas of concern. Firstly, this paper investigate the use of advanced business intelligence tenchniques to solve the problem of knowledge transfer from maintenance to design of aeroengines. Based on data availability and quality, various models were deployed. An association model was used to uncover hidden trends among parts involved in maintenance events. Classification techniques comprising of various algorithms was employed to determine severity of events. Causes of high severity events that lead to major financial loss was traced with the help of summarization techniques. Secondly this paper compares and evaluates the business intelligence approach to solve the problem of knowledge transfer with solutions available from the Semantic Web. The results obtained provide a compelling need to have data mining support on RDF/OWL-based warehoused data

    How informed are bronchial asthma patients: a questionnaire based study in a tertiary care hospital

    Background: Asthma has been considered as one of the most common chronic diseases worldwide. Asthma due to its effects on bronchial passage compromises the respiration and intern impairs the quality of life. Aim was to evaluate the knowledge of asthma among patients attending our tertiary care hospital.Methods: The patients were subjected to chest X-ray and spirometry followed by the knowledge on asthma questionnaire which consisted of 28 questions.Results: Out of 75 patients, aged between 18-80 years, majority were males. Out of 75 patients, 60 (80%) patients had obtained a total score below 10 out of the total of 28 score. The causative factors for asthma, 60 (80%) patients believed that asthma was due to allergy. Only 2-3 patients (3-4%) had knowledge of the underlying pathologic process during an asthma attack. Fifty nine (79%) patients were aware of the symptoms of asthma. However, only 10 (13%) patients knew about the early signs of worsening of asthma and only 14 (19%) patients could judge the severity of asthma. Only 5 (7%) patients knew which drug was for regular use and which was to be used if breathlessness occurred. Merely around 4-10 (5 to 13%) patients knew that asthma could be prevented by avoiding trigger factors and by taking medication regularly.Conclusions: A good level of knowledge about asthma and better practices are rigorously important to prevent asthma exacerbations. More comprehensive, regular and patient centred counselling programs will be beneficial in improving awareness of asthma
