    A Description of an Improved Homodyne Laser Interferometer

    First appearing on the commercial market in the mid 1980’s, diode-pumped, continuous-wave (cw) Nd:YAG lasers have more recently been used to obtain visible output, by the incorporation of frequency doubling optics in the laser cavity.The laser diode pumping of a Nd:YAG laser rod is selective and highly efficient, resulting in compact, high power, spatial mode lasers. Frequency-doubling processes are non-linear and lead to doubling TEM00 only of the high energy fundamental temporal mode, resulting in operation of the 532 nm laser in a single spatial and single longitudinal mode. The technology is rapidly advancing, and green lasers with energies of up to 1W could soon be available. The beam properties of the lasers described above are highly desirable in the field of interferometry, where such lasers are now in direct competition with the much larger Argon lasers, which have already been employed in high power interferometric systems. We describe here the performance of a modified Michelson interferometer [1–4], built to incorporate a 90 mW ADLAS 300 diode-pumped Nd:YAG laser. In previous versions of the Michelson interferometer, we have used HeNe lasers with a few milliwatts output, requiring mirror-quality surfaces on our samples. A 90 mW laser power enables us to make displacement measurements on metal surfaces with little or no preparation. The laser could also, of course, be used in other, more elaborate interferometer types, such as the confocal Fabry-Perot, which are better suited to industrial environments

    Pasture Yield and Animal Performance from Aeschynomene Americana Cultivars Glenn and Lee

    An on farm demonstration site was used to promote and compare the performance of the tropical legumes Glenn American jointvetch and Lee American jointvetch (Aeschynomene americana). Both legumes are widely sown in tropical Australia and are being increasingly sown in South-East Asian pasture projects. Glenn and Lee both grew well and persisted in grass/legume pastures over a 5 year period. Liveweight gain of steers, at a stocking rate of 1.5/ha, slightly favoured the Glenn pasture and gains of 0.49 kg/day for a 595 day period were achieved. Soil seed reserves were much higher under the annual Glenn than for the later flowering perennial Lee. Current recommendations are to sow equal portions of Glenn and Lee in legume/grass pasture mixture

    More Aeschynomene Pasture Legumes for the Tropics and Sub-Tropics

    Two cultivars of Aeschynomene villosa were released in Queensland in 1995. Cultivars ‘Reid’ (previously CPI 91209) and ‘Kretschmer’ (previously CPI 93621) are both native to Mexico and are the first cultivars of A. villosa to be released as pasture legumes. They will complement the three Aeschynomene cultivars already in use in tropical and sub-tropical northern and eastern Australia, namely cultivars ‘Glenn’ and ‘Lee’ (A. americana) and ‘Bargoo’ (A. falcata). Characteristics of Reid and Kretschmer jointvetch include, early seeding perennials within high seed yields, prostrate to semi-erect growth habit, adaptation to a wide range of soil types in medium rainfall areas, tolerance of temporary waterlogging, palatability of high quality forage, responsive to added phosphorus, and tolerance of light frosts. Commercial seed will be available for sale about October, 1997. Reid and Kretschmer villose jointvetch are widely adapted high quality pasture legumes which will complement current legume cultivars and will broaden the scope for sown pasture development in sub-tropical and tropical northern Australia

    Partnerships in sanitation service management and infrastructure provision in Kumasi, Ghana

    This paper investigates the potentials of the private sector in the development and management of sanitation infrastructure and services within three Sub metros in the Kumasi Metropolitan Assembly (KMA). A case study approach was used and data on public toilets under Build-Operate-Transfer (BOT) and franchise management were collected. Results showed that users of BOT facilities were generally satisfied with the services they receive but the KMA-owned toilets under franchised management receive low patronage because of the poor management and political interference. Some factors plaguing the partnership include mistrust between the partners, political interference and socio-cultural behaviour of the users

    Normal Cones and Thompson Metric

    The aim of this paper is to study the basic properties of the Thompson metric dTd_T in the general case of a real linear space XX ordered by a cone KK. We show that dTd_T has monotonicity properties which make it compatible with the linear structure. We also prove several convexity properties of dTd_T and some results concerning the topology of dTd_T, including a brief study of the dTd_T-convergence of monotone sequences. It is shown most of the results are true without any assumption of an Archimedean-type property for KK. One considers various completeness properties and one studies the relations between them. Since dTd_T is defined in the context of a generic ordered linear space, with no need of an underlying topological structure, one expects to express its completeness in terms of properties of the ordering, with respect to the linear structure. This is done in this paper and, to the best of our knowledge, this has not been done yet. The Thompson metric dTd_T and order-unit (semi)norms ∣⋅∣u|\cdot|_u are strongly related and share important properties, as both are defined in terms of the ordered linear structure. Although dTd_T and ∣⋅∣u|\cdot|_u are only topological (and not metrical) equivalent on KuK_u, we prove that the completeness is a common feature. One proves the completeness of the Thompson metric on a sequentially complete normal cone in a locally convex space. At the end of the paper, it is shown that, in the case of a Banach space, the normality of the cone is also necessary for the completeness of the Thompson metric.Comment: 36 page

    Asymptotic behaviour of random tridiagonal Markov chains in biological applications

    Discrete-time discrete-state random Markov chains with a tridiagonal generator are shown to have a random attractor consisting of singleton subsets, essentially a random path, in the simplex of probability vectors. The proof uses the Hilbert projection metric and the fact that the linear cocycle generated by the Markov chain is a uniformly contractive mapping of the positive cone into itself. The proof does not involve probabilistic properties of the sample path and is thus equally valid in the nonautonomous deterministic context of Markov chains with, say, periodically varying transitions probabilities, in which case the attractor is a periodic path.Comment: 13 pages, 22 bibliography references, submitted to DCDS-B, added references and minor correction

    TAp73 contributes to the oxidative stress response by regulating protein synthesis.

    TAp73 is a transcription factor that plays key roles in brain development, aging, and cancer. At the cellular level, TAp73 is a critical homeostasis-maintaining factor, particularly following oxidative stress. Although major studies focused on TAp73 transcriptional activities have indicated a contribution of TAp73 to cellular metabolism, the mechanisms underlying its role in redox homeostasis have not been completely elucidated. Here we show that TAp73 contributes to the oxidative stress response by participating in the control of protein synthesis. Regulation of mRNA translation occupies a central position in cellular homeostasis during the stress response, often by reducing global rates of protein synthesis and promoting translation of specific mRNAs. TAp73 depletion results in aberrant ribosomal RNA (rRNA) processing and impaired protein synthesis. In particular, polysomal profiles show that TAp73 promotes the integration of mRNAs that encode rRNA-processing factors in polysomes, supporting their translation. Concurrently, TAp73 depletion causes increased sensitivity to oxidative stress that correlates with reduced ATP levels, hyperactivation of AMPK, and translational defects. TAp73 is important for maintaining active translation of mitochondrial transcripts in response to oxidative stress, thus promoting mitochondrial activity. Our results indicate that TAp73 contributes to redox homeostasis by affecting the translational machinery, facilitating the translation of specific mitochondrial transcripts. This study identifies a mechanism by which TAp73 contributes to the oxidative stress response and describes a completely unexpected role for TAp73 in regulating protein synthesis

    Genomic comparisons of Escherichia coli ST131 from Australia.

    Escherichia coli ST131 is a globally dispersed extraintestinal pathogenic E. coli lineage contributing significantly to hospital and community acquired urinary tract and bloodstream infections. Here we describe a detailed phylogenetic analysis of the whole genome sequences of 284 Australian ST131 E. coli isolates from diverse sources, including clinical, food and companion animals, wildlife and the environment. Our phylogeny and the results of single nucleotide polymorphism (SNP) analysis show the typical ST131 clade distribution with clades A, B and C clearly displayed, but no niche associations were observed. Indeed, interspecies relatedness was a feature of this study. Thirty-five isolates (29 of human and six of wild bird origin) from clade A (32 fimH41, 2 fimH89, 1 fimH141) were observed to differ by an average of 76 SNPs. Forty-five isolates from clade C1 from four sources formed a cluster with an average of 46 SNPs. Within this cluster, human sourced isolates differed by approximately 37 SNPs from isolates sourced from canines, approximately 50 SNPs from isolates from wild birds, and approximately 52 SNPs from isolates from wastewater. Many ST131 carried resistance genes to multiple antibiotic classes and while 41 (14 %) contained the complete class one integron-integrase intI1, 128 (45 %) isolates harboured a truncated intI1 (462-1014 bp), highlighting the ongoing evolution of this element. The module intI1-dfrA17-aadA5-qacEΔ1-sul1-ORF-chrA-padR-IS1600-mphR-mrx-mphA, conferring resistance to trimethoprim, aminoglycosides, quaternary ammonium compounds, sulphonamides, chromate and macrolides, was the most common structure. Most (73 %) Australian ST131 isolates carry at least one extended spectrum β-lactamase gene, typically bla CTX-M-15 and bla CTX-M-27. Notably, dual parC-1aAB and gyrA-1AB fluoroquinolone resistant mutations, a unique feature of clade C ST131 isolates, were identified in some clade A isolates. The results of this study indicate that the the ST131 population in Australia carries diverse antimicrobial resistance genes and plasmid replicons and indicate cross-species movement of ST131 strains across diverse reservoirs

    GSMN-TB : a web-based genome-scale network model of Mycobacterium tuberculosis metabolism

    An impediment to the rational development of novel drugs against tuberculosis (TB) is a general paucity of knowledge concerning the metabolism of Mycobacterium tuberculosis, particularly during infection. Constraint-based modeling provides a novel approach to investigating microbial metabolism but has not yet been applied to genome-scale modeling of M. tuberculosis. Results GSMN-TB, a genome-scale metabolic model of M. tuberculosis, was constructed, consisting of 849 unique reactions and 739 metabolites, and involving 726 genes. The model was calibrated by growing Mycobacterium bovis bacille Calmette Guérin in continuous culture and steady-state growth parameters were measured. Flux balance analysis was used to calculate substrate consumption rates, which were shown to correspond closely to experimentally determined values. Predictions of gene essentiality were also made by flux balance analysis simulation and were compared with global mutagenesis data for M. tuberculosis grown in vitro. A prediction accuracy of 78% was achieved. Known drug targets were predicted to be essential by the model. The model demonstrated a potential role for the enzyme isocitrate lyase during the slow growth of mycobacteria, and this hypothesis was experimentally verified. An interactive web-based version of the model is available. Conclusion The GSMN-TB model successfully simulated many of the growth properties of M. tuberculosis. The model provides a means to examine the metabolic flexibility of bacteria and predict the phenotype of mutants, and it highlights previously unexplored features of M. tuberculosis metabolism

    JISTIC: Identification of Significant Targets in Cancer

    <p>Abstract</p> <p>Background</p> <p>Cancer is caused through a multistep process, in which a succession of genetic changes, each conferring a competitive advantage for growth and proliferation, leads to the progressive conversion of normal human cells into malignant cancer cells. Interrogation of cancer genomes holds the promise of understanding this process, thus revolutionizing cancer research and treatment. As datasets measuring copy number aberrations in tumors accumulate, a major challenge has become to distinguish between those mutations that drive the cancer versus those passenger mutations that have no effect.</p> <p>Results</p> <p>We present JISTIC, a tool for analyzing datasets of genome-wide copy number variation to identify driver aberrations in cancer. JISTIC is an improvement over the widely used GISTIC algorithm. We compared the performance of JISTIC versus GISTIC on a dataset of glioblastoma copy number variation, JISTIC finds 173 significant regions, whereas GISTIC only finds 103 significant regions. Importantly, the additional regions detected by JISTIC are enriched for oncogenes and genes involved in cell-cycle and proliferation.</p> <p>Conclusions</p> <p>JISTIC is an easy-to-install platform independent implementation of GISTIC that outperforms the original algorithm detecting more relevant candidate genes and regions.
