    Investigation of porcupine bezoar extract (Hystrix Brachyuran) on hela cell combined with electroporation method

    Electroporation (EP) is a technique whereby the cell membrane is exposed to increase the intensity of electric field pulses that can lead to a variety of biophysical and biochemical responses. It is a molecular biology technique that stimulates pores through cell membrane, to increase the permeability of the cell, allowing chemicals, and drugs or Deoxyribonucleic acid (DNA) to be introduced into the cell. In this research, the HeLa cell behavior was investigated via EP. The purpose of this study is to investigate the best parameter of EP to be applied on HeLa cell treated with the best concentration of PBE which was 80.0ug/ml. Thus, the experiment is conducted for 48 hours and is initiated by identifying the best parameter for EP process. The range of electric field used is commenced between 100V/cm and 500V/cm with the increment of 100 V/cm interval sand for the pulse duration from 100ฮผs, 500ฮผs, 1ms, 2ms and 5ms. Based on the parameter used, the best result obtained is when the parameter was set to 500 V/cm for the electric field and 100ฮผs for pulse duration, where it inhibits the lowest proliferation rate percentage of HeLa cell. For the natural resource compound or extract which is available in Malaysia Porcupine Bezoar (Hystrix Brachyuran) were used. This extract has the potential as an anti-proliferation agent, which is useful in anti-cancer application. The experiment suggests that the best concentration for Porcupine Bezoar Extract (PBE) was at 80.0ug/ml. This combined method of EP and PBE revealed the process inhibited the proliferation of HeLa cell to 8.48%. In comparison to the study without this combined technique of EP and PBE, the inhibition process reached 55.35%.The study has successfully demonstrated the contribution towards a new method for anti-cancer treatment due to its minimal side effects compared to other conservative methods such as Electrochemotherapy (ECT)

    Electroporation (EP) is a technique whereby the biophysical changes on the cells that induced external high-intensity electrical field pulses in order to enhance applications in the medical field. It is a molecular biology technique in order to create pores through a cell wall membrane, boost the permeability of the cell membrane, and support chemicals, drugs or DNA to be imported into Hela cells. While by combining electroporation (EP) technique with porcupine bezoar (PB) extract might reduce the proliferation of HeLa cells because this compound extract has the ability of anti-proliferation and also anti-angiogenesis properties for controlling cancer cell growth. This research concentrate on reviewing and analyses the basic concepts and methods of combining electroporation and porcupine bezoar (PB) extract as applied in cancer treatment application. The combination of this technique might be a new alternative for anti-cancer treatment.     Investigation of porcupine bezoar extract combined with electroporation on HeLa cell

    Current chemo-preventive agents will causes a long term side effects in cancer sufferer. Therefore, this research focusses on the benefits of combine both technique of Electroporation (EP) method and with natural animal extract in process to inhibit the proliferation of cancer cell, as a solution to reduce or adverse effects of orthodox drugs. The present study indicates on anti-cancer potentially of Porcupine Bezoar Extract (PBE), also known as Hystrix Brachyuran against HeLa cell. Cell viability of HeLa cell were determined after HeLa cell treated with 500 V/cm and pulse duration of 100ยตs before the concentration of 80.0 ยตg/ml of Porcupine Bezoar Extract (PBE) was added into the cell. The cell viability and cell growth were monitored up to 48 hours with comparative to untreated cell as a control group.This study result proved that by combine both technique; Electroporation (EP) and with natural animal extract Porcupine Bezoar Extract (PBE) might open the door and it has an ability in supressing the growth of HeLa cell

    In order to bring up a cells or tissue with a gene or DNA, Electroporation parameter such as pulse duration, field strength and number of pulse plays the most important aspect. Thus, the aim of this paper is to optimize the pulse duration parameter for HeLa cells line. This is very important because nowadays, research on EP factors is still in limited mode. Moreover, suitable and applicable pulse duration parameter would ensure successful treatment with least possible side effect. In this paper, HeLa cells or known as cervical cancer cells was used to induce into a single pulse electric field. Therefore, to achieve the aim of this study, field strength of 500V/cm was preferred and the pulse duration was varied in the range of 100ฮผs, 500ฮผs, 1ms, 2ms and 5ms to obtain lowest proliferation rate and inhibit growth of HeLa cells. The result obtained shows that HeLa cell induced to 500V/cm with pulse duration of 100ฮผs, induced the lowest proliferation rate (45%) as compared to initial reading density (control) after 48 hours of culture. Thus, further research based on this work may pave the way for an anti-cancer application through the study of the effect of pulse duration parameter on HeLa cells

    Porcupine Bezoar Extract (PBE) has been previously described to possess diverse medicinal properties. In any case, its potential and ability as anti-cancer agent on HeLa cell line are still poorly studied and understood. Hence, in this research, Porcupine Bezoar Extract (PBE) aqueous extract was figure out for its potential as a safe anti-cancer agent. Initially, Porcupine Bezoar Extract (PBE) was infused with distilled water for 18 hours to get PBE aqueous solution in order to confirm the presence of bioactive agents as antioxidants. Later, PBE aqueous solution was subjected to HeLa cell line and was observed for 48 hours exposures with comparative to the untreated cell to evaluate its anticancer potential and ability. The data from this study propose the potential agents for improving human health, especially in the anti-cancer application
