    Analysis of Developmental Changes in Avian DNA Methylation Using a Novel Method for Quantifying Genome-wide DNA Methylation

    Individual differentiated somatic cells and undifferentiated stem cells have common genome, although their functions or morphological characters are very different. These differences are derived from difference of gene expression pattern. DNA methylation is generally key factor of Suppression of gene and its level is globally change during mammalian early development. But, in birds, whether genome-wide changes in DNA methylation occur during embryonic development is still unknown. Here, we show that genome-wide DNA methylation to assess occurrence during early chick embryonic development. We found that the methylation status at stage 1 was approximately 57%, after which it gradually decreases, reaching a minimum at stage 10 (33%). After stage 10, DNA methylation gradually increased.     Amphiregulin and Epiregulin mRNA expression in primary colorectal cancer and corresponding liver metastases

    <p>Abstract</p> <p>Background</p> <p>Amphiregulin (AREG) and Epiregulin (EREG), ligands of EGFR, are reported to be predictive biomarkers of colorectal cancer patients treated with Cetuximab, an anti-EGFR antibody. The purpose of this study is to determine the correlation of AREG and EREG expression between primary colorectal cancer and corresponding liver metastases.</p> <p>Methods</p> <p>One hundred twenty colorectal cancer patients with liver metastases (100 with synchronous metastases, 20 with metachronous) were evaluated. No patients had ever received anti-EGFR antibody agents. AREG and EREG mRNA expression from both the primary tumor and liver metastases were measured using real-time RT-PCR. KRAS codon 12, 13 mutation status was analyzed by direct sequencing.</p> <p>Results</p> <p>Modest, but significant, correlations were observed between primary tumor and corresponding liver metastases in both AREG mRNA expression (Rs = 0.54, p < 0.0001) and EREG mRNA expression (Rs = 0.58, p < 0.0001). AREG and EREG mRNA expression was strongly correlated in both the primary tumor (Rs = 0.81, p < 0.0001) and the liver metastases (Rs = 0.87, p < 0.0001). No significant survival difference was observed between low and high AREG or EREG patients when all 120 patients were analyzed. However, when divided by KRAS status, KRAS wild-type patients with low EREG mRNA levels in the primary site showed significantly better overall survival rates than those with high levels (p = 0.018). In multivariate analysis, low EREG expression was significantly associated with better overall survival (p = 0.006).</p> <p>Conclusions</p> <p>AREG and EREG expression showed a modest correlation between primary tumor and liver metastases. As EREG mRNA expression was associated with decreased survival, it is appeared to be a useful prognostic marker in KRAS wild-type patients who never received anti-EGFR therapy.</p

    Subjective intelligibility of speech sounds enhanced by ideal ratio mask via crowdsourced remote experiments with effective data screening

    It is essential to perform speech intelligibility (SI) experiments with human listeners to evaluate the effectiveness of objective intelligibility measures. Recently crowdsourced remote testing has become popular to collect a massive amount and variety of data with relatively small cost and in short time. However, careful data screening is essential for attaining reliable SI data. We compared the results of laboratory and crowdsourced remote experiments to establish an effective data screening technique. We evaluated the SI of noisy speech sounds enhanced by a single-channel ideal ratio mask (IRM) and multi-channel mask-based beamformers. The results demonstrated that the SI scores were improved by these enhancement methods. In particular, the IRM-enhanced sounds were much better than the unprocessed and other enhanced sounds, indicating IRM enhancement may give the upper limit of speech enhancement performance. Moreover, tone pip tests, for which participants were asked to report the number of audible tone pips, reduced the variability of crowdsourced remote results so that the laboratory results became similar. Tone pip tests could be useful for future crowdsourced experiments because of their simplicity and effectiveness for data screening.Comment: This paper was submitted to Interspeech 2022 (http://www.interspeech2022.org

    Mycosis fungoides, the most common subtype of cutaneous T-cell lymphoma, displays a broad spectrum of clinical, histological and phenotypic variants with different prognostic impacts. The classic immunophenotype is CD3+/CD4+/CD45RO+memory T cells. CD4/CD8 double-negative mycosis fungoides is rare. Here we describe the clinicopathological features of CD4/CD8 double-negative mycosis fungoides in a 55-year-old female with a review of the literature. Although the CD4/CD8 double-negative phenotype appears to be associated with an unusual clinical presentation, it does not appear to confer prognostic significance

    High-pressure synthesis of superconducting Nb{1-x}B2 (x = 0-0.48) with the maximum Tc = 9.2 K

    Superconductivity with Tc above 9 K was found in metal-deficient NbB2 prepared under 5 GPa, while no clear superconductivity was observed down to 3 K in stoichiometric NbB2. The superconductivity was observed above x = 0.04 in Nb1-xB2. and the lattice parameters also changed abruptly at x = 0.04. As x increased, the transition temperature Tc slightly rose and fell with the maximum value of 9.2 K at x = 0.24 for the samples sintered at 5 GPa and 1200 C. The Tc-value changed in the range from 7 K to 9 K, depending on the sintering pressure. A series of Ta1-xB2 (0 =< x =< 0.24) was also synthesized under high pressure to examine a special effect of high-pressure synthesis.Comment: 24 pages including 2 tables and 7 figures, accepted for publication in Physica
