    Smoking counseling practices of physicians in the United Arab Emirates

    Background. Despite the cost effectiveness of physician smoking cessation counseling, many physicians do not adhere to current clinical practice guidelines. Methods. A cross-sectional mail survey was conducted in a sample of general practitioners in the United Arab Emirates (UAE), emirates of Sharjah, Dubai and Abu Dhabi, to document cessation-counseling practices and identify correlates of these activities. Results. Of 391 eligible GPs, 218 (56%) completed the questionnaire. Despite favorable beliefs/attitudes about cessation counseling, less than half of GPs (47%) indicated that they have the skills to help their patients quit smoking and only 24% knew of community resources to refer patients to. GPs who perceived physician barriers in offering smoking counseling as very important were significantly less likely to ascertain smoking status of most of their patients (OR: 0.28, 95% CI: 0.09,0.88). Moreover, high self-efficacy and favorable beliefs/attitudes were significantly associated with high level of counseling completeness (OR: 4.44, 95% CI: 1.21, 16.37; OR: 3.78, 95% CI: 1.25, 11.35; respectively). Conclusion. Support, training and intervention programs to overcome lack of awareness and knowledge, unfavorable beliefs/attitudes, and low self-efficacy could increase and enhance cessation-counseling practices among general practitioners in the UAE. © 2010, SAGE Publications.     TOPICAL AND SYSTEMIC DERMAL CARRIERS FOR PSORIASIS

    Psoriasis is a non-infectious, dry, inflammatory (autoimmune) skin disorder. Treatment approaches include phototherapy, topical, oral and other systemic drug delivery. However, owing to the side effects and incomplete cure accompanying the oral administration as well as phototherapy, the topical route seemed to be more satisfactory for the medical team. Dermal treatment ensuring percutaneous penetration is now highly recommended in topical indications for psoriatic patients, which can be achieved using pharmaceutical carriers. Several carrier systems loaded with antipsoriatic drugs have demonstrated promising results, with some of them strictly being confined to the skin and others allowing for systemic involvement also. The evolution in this area will present a more useful and safer therapy by minimizing the drugs' degradation and loss, and increasing their bioavailability and effectiveness. Since patients require at least three topical applications for almost a 1-year period to gain health benefit, a reduction in the cost of the treatment will be of real value. A distinction of these carriers is made in the current review, to allow the choice of the most suitable pharmaceutical carrier for psoriatic patients requiring either local and/or systemic involvement

    Characterization of silver nanoparticles prepared by wet chemical method and their antibacterial and cytotoxicity activities

    Purpose: To investigate the efficiency of silver nanoparticles synthesized by wet chemical method, and evaluate their antibacterial and anti-cancer activities.Methods: Wet chemical method was used to synthesize silver nanoparticles (AgNPs) from silver nitrate, trisodium citrate dehydrate (C6H5O7Na3.2H2O) and sodium borohydride (NaBH4) as reducing agent. The AgNPs and the reaction process were characterized by UV–visible spectrometry, zetasizer, transmission electron microscopy (TEM) and scanning electron microscopy (SEM) equipped with energy dispersive spectroscopy (EDS). The antibacterial and cytotoxic effects of the synthesized nanoparticles were investigated by agar diffusion method and MTT assay respectively.Results: The silver nanoparticles formed were spherical in shape with mean size of 10.3 nm. The results showed good antibacterial properties, killing both Gram-positive and Gram-negative bacteria, and its aqueous suspension displayed cytotoxic activity against colon adenocarcinoma (HCT-116) cell line.Conclusion: The findings indicate that silver nanoparticles synthesized by wet chemical method demonstrate good cytotoxic activity in colon adenocarcinoma (HCT-116) cell lines and strong antibacterial activity against various strains of bacteria.Keywords: Wet chemical, Silver nanoparticles, Characterization, Diffusion method, Antibacterial, Cytotoxic activit

    Novel Green Synthesis and Characterization of Nanopolymer Porous Gold Oxide Nanoparticles

    Purpose: To develop a novel approach to green synthesis of nano-polymer porous gold oxide nanoparticles, and examine the effects of the temperatures on their surface.Methods: Green synthesis of nano-polymer porous gold oxide nanoparticles (GONPs) using cetyle trimethylammonium bromide (CTAB) surfactant with a mixture of Olea europaea fruit and Acacia Nilotica extracts, was performed using sol-gel method. The nanoporous particles were characterized by UV (ultraviolet (UV) visible spectroscopy and dynamic light scattering (DLS) while a zetasizer was applied to determine their average particle size. Their surface morphology and shape were assessed by transmission electron microscopy (TEM) and scanning election microscopy (SEM) while surface area was measured using nitrogen gas adsorption method.Results: TEM and SEM images showed a smooth, cylindrical or spherical, and cluster shapes, and porous surface morphology. Increase in calcination temperature resulted in increase in surface area and pore volume of nanoparticles. This feature yielded GONPs that were unique with a high surface area of 146.706 m2/g.Conclusion: The approach used in this study constitutes a new and rapid green synthesis of porous nanoparticles of gold oxide under simple conditions.     Association between polymorphisms of SLC6A3 and DRD1 genes and autism among Saudi Arabia Taif population using PCR-restriction fragment length polymorphism (PCR- RFLP)

    The prevalence of autism in Saudi Arabia is 18 per 10,000, higher than the 13 per 10,000 reported in developed  countries. The etiology of autism is still not completely understood. Different studies support the involvement  of dopaminergic neurotransmitter system in the etiology of autism. Several lines of evidences suggest the role of some dopamine related genes, such as DRD1 and SLC6A3 in the etiology of autism. The aim of the present  work was to study the possible role of rs2550936 A/C polymorphism at SLC6A3 locus as well as rs4532 A/G  polymorphism at DRD1 locus in the etiology of autism among Saudi population. The polymorphisms of DRD1  and LC6A3 were genotyped in the case-control study using polymerase chain reaction-restriction fragment  length polymorphism (PCR-RFLP) technique. Significant association as risk factor was found between autism  and GA genotype of DRD1 [OR = 3.5 CI (1.04, 12.41*)] as well as CA genotype of SLC6A3 [OR = 2.53 CI  (1.03, 6.26*)], while CC genotype of SLC6A3 revealed protective effect. In conclusion, possible risk  genotypes  for autism in the DRD1 and SLC6A3 genes were observed. This is the first report in Saudi Arabia  population and Arab world. Therefore further investigations of these markers and other SNPs of SLC6A3 and  DRD1 genes are considered in large replication samples with other causal factors to enable positive  identification of risk genotypes and generalize obtained results.Key words: Etiology, polymorphism, autism, genotype

    The significance of Epstein Barr Virus (EBV) & DNA Topoisomerase II alpha (DNA-Topo II alpha) immunoreactivity in normal oral mucosa, Oral Epithelial Dysplasia (OED) and Oral Squamous Cell Carcinoma (OSCC)

    <p>Abstract</p> <p>Background</p> <p>Head and neck cancer including oral cancer is considered to develop by accumulated genetic alterations and the major pathway is cancerization from lesions such as intraepithelial dysplasia in oral leukoplakia and erythroplakia. The relationship of proliferation markers with the grading of dysplasia is uncertain. The involvement of EBV in oral carcinogenesis is not fully understood.</p> <p>Aim</p> <p>The present study was designed to investigate the role of EBV and DNA Topoisomerase II∝ (DNA-Topo II∝) during oral carcinogenesis and to examine the prognostic significance of these protein expressions in OSCCs.</p> <p>Methods</p> <p>Using specific antibodies for EBV and DNA-Topo II∝, we examined protein expressions in archival lesion tissues from 16 patients with oral epithelial dysplasia, 22 oral squamous cell carcinoma and 20 normal oral mucosa by immunohistochemistry. Clinical information was obtained through the computerized retrospective database from the tumor registry.</p> <p>Results</p> <p>DNA-Topo II∝ was expressed in all examined specimens. Analysis of Variance ANOVA revealed highly significant difference (P < 0.01) in young aged labial tissues and significant (P ≀ 0.05) in gingival and not significant (P > 0.05) in inferior surface of tongue and in hard palatal tissues. Significant differences were observed between OEDs and NSE (P < 0.001) and SCCs and controls (P < 0.001), also, significant differences could be observed between SCCs and OEDs. DNA-Topo II∝ expression was significantly higher in tumors of low differentiation versus tumors of moderate and high differentiation (P < 0.001), DNA-Topo II∝ expression was correlated with age, tumor size, tumor stage, node metastasis and tumor differentiation, but not with gender and tumor site. None of normal squamous epithelium (NSE) expressed EBV. Heterogenous reactivity for EBV was observed through the series of dysplasia and squamous cell carcinoma. Its expression increased progressively with lymph node metastasis and low tumor differentiation, but no significant association could be observed with other clinicopathological parameters. EBV protein expression was increased with elevated Topo II-∝ LI in OEDs and OSCCs. A tendency to positive correlation between EBV and Topo II∝ expression was observed in OEDs but not in OSCCs.</p> <p>Conclusion</p> <p>EBV and DNA Topo II-αLI expression are possible indicators in oral carcinogenesis and may be valuable diagnostic and prognostic indices in oral carcinoma.</p

    ANTIOXIDANT AND HEPATOPROTECTIVE EFFECTS OF JUSTICIA SPICIGERA ETHYL ACETATE FRACTION AND CHARACTERIZATION OF ITS ANTHOCYANIN CONTENT

    Objective: The antioxidant and hepatoprotective activities of ethyl acetate (EA) fraction of the dried aerial part of Justicia spicigera were evaluated and the characterization of its anthocyanin content was done. Methods: Hepatic fibrosis was induced by carbon tetrachloride (CCl4) in rats. The ethyl acetate fraction was obtained by successive liquid/liquid fractionation of the crude cold ethanolic extract and the pigments were characterized by HPLC technique. The in vitro studies were carried out through evaluation of the EA fraction on the attenuation of 1, 1-Diphenyl-2-picrylhydrazyl (DPPH) free radicals. The in vivo biological evaluation was done in CCl4 injured rats through determination of liver function indices, oxidative stress markers and the histopathological picture of the treated liver.Results: The phenolic content in the EA fraction was 42.94 mg/g. Twelve anthocyanins were identified, the major of which are peonidin 3, 5-diglucoside (64.30%), malvidin 3, 5-diglucoside (10.59%) and petunidin 3,5-diglucoside (4.71%). Treatment of CCl4 intoxicated rats with EA fraction recorded improvement in the liver function indices and oxidative stress markers. The histopathological observations confirmed our results.Conclusion: The ethyl acetate fraction of the dried aerial part of Justicia spicigera recorded antioxidant and hepato protective activities.Â

    Preparation, characterization and antibacterial effects of eco-friendly gold nanorods

    Purpose: To synthesize and characterize eco-friendly gold nanorods (Au-NRs) and to assess their effects against two bacterial strains.Methods: Synthesis of eco-friendly gold nanorods was done from an aqueous solution of chloroauric acid and cetyltrimethylammonium bromide by mixing Olea europaea fruit and Acacia nilotica husk extracts with the latter as a reducing agent. The synthesis was monitored by ultraviolet–visible (UV) spectrophotmetry and a zetasizer, while the morphology of the resulting nanorods was assessed by transmission electron microscopy (TEM) and scanning electron microscopy (SEM) coupled with an energy-dispersive spectrophotometer (EDS). The effect of the prepared eco-nanorods on growth of Escherichia coli and Staphylococcus aureus bacteria were investigated by agar diffusion method.Results: The synthesized Au-NRs were mostly rod-shaped, with mean size of 96 nm. They exhibited a potent antibacterial activity against Gram-positive and  Gram-negative microorganisms (Escherichia coli and Staphylococcus aureus).Conclusion: The findings suggest that the nanoformulation of the biomaterial mix of Olea europaea fruit and Acacia nilotica husk extracts is a cost-effective, eco-friendly, and non-toxic synthesis of Au- NRs which has therpeautic potential.Keywords: Olea europaea, Acacia nilotica, Gold nanorods, Eco-friendly synthesis, Gram-positive and Gram-negative microorganism

    MicroRNA-34a: A Key Regulator in the Hallmarks of Renal Cell Carcinoma

    Renal cell carcinoma (RCC) incidence has increased over the past two decades. Recent studies reported microRNAs as promising biomarkers for early cancer detection, accurate prognosis, and molecular targets for future treatment. This study aimed to evaluate the expression levels of miR-34a and 11 of its bioinformatically selected target genes and proteins to test their potential dysregulation in RCC. Quantitative real-time PCR for miR-34a and its targets; MET oncogene; gene-regulating apoptosis (TP53INP2 and DFFA); cell proliferation (E2F3); and cell differentiation (SOX2 and TGFB3) as well as immunohistochemical assay for VEGFA, TP53, Bcl2, TGFB1, and Ki67 protein expression have been performed in 85 FFPE RCC tumor specimens. Clinicopathological parameter correlation and in silico network analysis have also implicated. We found RCC tissues displayed significantly higher miR-34a expression level than their corresponding noncancerous tissues, particularly in chromophobic subtype. MET and E2F3 were significantly upregulated, while TP53INP2 and SOX2 were downregulated. ROC analysis showed high diagnostic performance of miR-34a (AUC = 0.854), MET (AUC = 0.765), and E2F3 (AUC = 0.761). The advanced pathological grade was associated with strong TGFB1, VEGFA, and Ki67 protein expression and absent Tp53 staining. These findings indicate miR-34a along with its putative target genes could play a role in RCC tumorigenesis and progression

    Green Synthesis, Characterization, and Antibacterial Activity of Silver/Polystyrene Nanocomposite

    A novel, nontoxic, simple, cost-effective and ecofriendly technique was used to synthesize green silver nanoparticles (AgNPs). The AgNPs were synthesized using orange peel extract as a reducing agent for silver nitrate salt (AgNO3). The particle size distribution of AgNPs was determined by Dynamic Light Scattering (DLS). The average size of silver nanoparticles was 98.43 nm. The stable dispersion of silver nanoparticles was added slowly to polystyrene solution in toluene maintaining the temperature at 70°C. The AgNPs/polystyrene (PS) nanocomposite solution was cast in a petri dish. The silver nanoparticles encapsulated within polymer chains were characterized by X-ray diffraction (XRD) and Scanning Electron Microscopy (SEM) equipped with Energy Dispersive Spectroscopy (EDS) in addition to Transmission Electron Microscopy (TEM). The green AgNPs/PS nanocomposite film exhibited antimicrobial activity against Gram-negative bacteria Escherichia coli, Klebsiella pneumoniae and Salmonella, and Gram-positive bacteria Staphylococcus aureus. Thus, the key findings of the work include the use of a safe and simple AgNPs/PS nanocomposite which had a marked antibacterial activity which has a potential application in food packaging
