    The development and the progress of media communication at the present is a fact of the knowledge and the technology development that must be accepted. It presence like the flowing water which has a fast current that brings also two influences both positive and negative that must be accounted for the members of the Catholic Students Community Of St. Martinus Ende (KMK St. Martinus Ende). Both positive and negative influences the media community like a kinetic energy or a power attraction that attract  them in a tiring ambiquity. Let them walk alone without escort of a decisive compass where they should have a rightist attitude and responsible. On the point, the guidance and assistance of the church is an  offering  if the church will be born a generation  of the future  of the  church  that is mature and has a certain quality  based  on the growth  and the development  of acuteness and inner  to determine the attitude to the development of media communication. The process of sharpening of mind and the sharpeness of the participants can be realized by giving some activities such as: awareness, deepening and even  the sharpeness of the actor of  media communication as an  alternative of reporting work of the God Kingdom for human beings. It becomes the main moving spirit or activator  for the board of KMK Of St. Martinus Ende  to plan and boring  about the activity of catechism. The activity rise the method of Amos.  By this method, the participants are invited to build a deeply reflection that based on thein real experiences about the media communication, while keep on self opening to the God planning will come  to them  and  give them via  the commandment of God.  The commandment  of God  come to light, inspiration, motivate, power and critics to the  participants about the using of the media communication as a media of the commandment of the kingdom of God  to the world that is more progress and development lately

    Marriage preparation education and training for prospective married couples also have been known in Indonesia as marriage preparation training courses (KPP). Like the other parishes in every diocese all over the world, St. Joseph Parish of Onekore Ende which belong to the vicariate pastoral ministry area of Ende, and the Archdiocese of Ende cares and pays more attention to the needs of parish community members who are planning to build a family up for living together as husband and wife. So, responding to the needs of these prospective couples and based on the pastoral activity planning that has been designed in an annual plenary pastoral assembly, the Parish Pastoral Council, through the Family Pastoral Section of the Parish, carries out an educational and training program for the premarital marriage preparation activities under the coordination of the Parish Pastor and his fellow co-pastor. The facilitators use the acronym term PMPET in this article. PMPET is shortened from Premarital Marriage Preparation Education and Training. The purpose is to know whether PMPET as an approach and model be able to enhance the knowledge and skill of the participants about the nature of marriage which is discussed from various perspectives or points of view

    The communication media  as a work of human beings reason from time to time has a very rapidly progress together with the growth and the development of new innovations in the world of the digital technology. This progress is a  certainty that must be received by the human beings who are in a strong mind and conscience awareness that communication media is so sophisticated but remains as a means of the development of the human beings civilization. This awareness gradually can be expected to encourage human beings in order to place themselves appropriately as the subject of the  development of the communication media itself through a rational and an accurate filtering process against negative influences that can threaten with human beings civilization. One of the institutions is threatened with the negative  influences of the communicaton media is family. At first the family gets a respected  position as a socialization agent about the values of life is now experiencing decline of function as well as its role because the majority of the family members are now snared because of the negative influences of the communication  media. The sympathetic as well as the  empathetic atmosphere are used to be the first decoration in the  house and now  is just becoming a nice memory but sadden. The fascination  is too axtreme makes each of the family member of the communication media and finally appear some attitudes as well as behaviours that don,t go with the values of solidarity and the family as well.     Impact of killer-immunoglobulin-like receptor and human leukocyte antigen genotypes on the efficacy of immunotherapy in acute myeloid leukemia

    Interactions between killer-immunoglobulin-like receptors (KIRs) and their HLA class I ligands are instrumental in natural killer (NK) cell regulation and protect normal tissue from NK cell attack. Human KIR haplotypes comprise genes encoding mainly inhibitory receptors (KIR A) or activating and inhibitory receptors (KIR B). A substantial fraction of humans lack ligands for inhibitory KIRs (iKIRs), that is, a 'missing ligand' genotype. KIR B/x and missing ligand genotypes may thus give rise to potentially autoreactive, unlicensed NK cells. Little is known regarding the impact of such genotypes in untransplanted acute myeloid leukemia (AML). For this study, NK cell phenotypes and KIR/HLA genotypes were determined in 81 AML patients who received immunotherapy with histamine dihydrochloride and low-dose IL-2 for relapse prevention (NCT01347996). We observed that presence of unlicensed NK cells impacted favorably on clinical outcome, in particular among patients harboring functional NK cells reflected by high expression of the natural cytotoxicity receptor (NCR) NKp46. Genotype analyses suggested that the clinical benefit of high NCR expression was restricted to patients with a missing ligand genotype and/or a KIR B/x genotype. These data imply that functional NK cells are significant anti-leukemic effector cells in patients with KIR/HLA genotypes that favor NK cell autoreactivity

    Cerebrospinal fluid biomarkers of brain injury, inflammation and synaptic autoimmunity predict long-term neurocognitive outcome in herpes simplex encephalitis

    OBJECTIVES: To investigate the correlation between biomarkers of brain injury and long-term neurocognitive outcome, and the interplay with intrathecal inflammation and neuronal autoimmunity, in patients with herpes simplex encephalitis (HSE). METHODS: A total of 53 adult/adolescent HSE patients were included from a prospective cohort in a randomized placebo-controlled trial investigating the effect of a 3-month follow-up treatment with valaciclovir. Study subjects underwent repeated serum/CSF sampling and brain MRI the first 3 months along with cognitive assessment by Mattis Dementia Rating Scale (MDRS) during 24 months. CSF samples were analyzed for biomarkers of brain injury, inflammation and synaptic autoimmunity. The pre-defined primary analysis was the correlation between peak CSF neurofilament protein (NFL), a biomarker of neuronal damage, and MDRS at 24 months. RESULTS: Impaired cognitive performance significantly correlated with NFL levels (rho = -0.36, p = 0.020). Development of IgG anti-N-methyl-D-aspartate receptor (NDMAR) antibodies was associated with a broad and prolonged proinflammatory CSF response. In a linear regression model, lower MDRS at 24 months was associated with previous development of IgG anti-NMDAR (beta = -0.6249, p = 0.024) and age (z-score beta = -0.2784, p = 0.024), but not CSF NFL, which however significantly correlated with subsequent NMDAR autoimmunization (p = 0.006). CONCLUSIONS: Our findings show that NFL levels are predictive of long-term neurocognitive outcome in HSE, and suggest a causative chain of events where brain tissue damage increases the risk of NMDAR autoimmunisation and subsequent prolongation of CSF inflammation. The data provides guidance for a future intervention study of immunosuppressive therapy administered in the recovery phase of HSE

    Quantitative modelling of landslide hazard, as opposed to landslide susceptibility, as a function of the earthquake trigger is vital in understanding and assessing future potential exposure to landsliding. Logistic regression analysis is a method commonly used to assess susceptibility to landsliding; however, estimating probability of landslide hazard as a result of an earthquake trigger is rarely undertaken. This paper utilises a very detailed landslide inventory map and a comprehensive dataset on peak ground acceleration for the 1994 Mw6.7 Northridge earthquake event to fit a landslide hazard logistic regression model. The model demonstrates a high success rate for estimating probability of landslides as a result of earthquake shaking. Seven earthquake magnitude scenarios were simulated using the Open Source Seismic Hazard Analysis (OpenSHA) application to simulate peak ground acceleration, a covariate of landsliding, for each event. The exposure of assets such as population, housing and roads to high levels of shaking and high probabilities of landsliding was estimated for each scenario. There has been urban development in the Northridge region since 1994, leading to an increase in prospective exposure of assets to the earthquake and landslide hazards in the event of a potential future earthquake. As the earthquake scenario magnitude increases, the impact from earthquake shaking initially increases then quickly levels out, but potential losses from landslides increase at a rapid rate. The modelling approach, as well as the specific model, developed in this paper can be used to estimate landslide probabilities as a result of an earthquake event for any scenario where the peak ground acceleration variable is available

    Pembrolizumab for Treatment-Refractory Metastatic Castration-Resistant Prostate Cancer: Multicohort, Open-Label Phase II KEYNOTE-199 Study

    PURPOSE: Pembrolizumab has previously shown antitumor activity against programmed death ligand 1 (PD-L1)-positive metastatic castration-resistant prostate cancer (mCRPC). Here, we assessed the antitumor activity and safety of pembrolizumab in three parallel cohorts of a larger mCRPC population. METHODS: The phase II KEYNOTE-199 study included three cohorts of patients with mCRPC treated with docetaxel and one or more targeted endocrine therapies. Cohorts 1 and 2 enrolled patients with RECIST-measurable PD-L1-positive and PD-L1-negative disease, respectively. Cohort 3 enrolled patients with bone-predominant disease, regardless of PD-L1 expression. All patients received pembrolizumab 200 mg every 3 weeks for up to 35 cycles. The primary end point was objective response rate per RECIST v1.1 assessed by central review in cohorts 1 and 2. Secondary end points included disease control rate, duration of response, overall survival (OS), and safety. RESULTS: Two hundred fifty-eight patients were enrolled: 133 in cohort 1, 66 in cohort 2, and 59 in cohort 3. Objective response rate was 5% (95% CI, 2% to 11%) in cohort 1 and 3% (95% CI, = 21.8 months) and 10.6 months (range, 4.4 to 16.8 months), respectively. Disease control rate was 10% in cohort 1, 9% in cohort 2, and 22% in cohort 3. Median OS was 9.5 months in cohort 1, 7.9 months in cohort 2, and 14.1 months in cohort 3. Treatment-related adverse events occurred in 60% of patients, were of grade 3 to 5 severity in 15%, and led to discontinuation of treatment in 5%. CONCLUSION: Pembrolizumab monotherapy shows antitumor activity with an acceptable safety profile in a subset of patients with RECIST-measurable and bone-predominant mCRPC previously treated with docetaxel and targeted endocrine therapy. Observed responses seem to be durable, and OS estimates are encouraging
