    Implementation of the child labour policy in South Africa

    A research report submitted to the Faculty of Management, University of the Witwatersrand, in partial fulfilment of the requirements for the degree of Master of Management (in the field of Public Policy) , April 2016Child labour is a social problem that affects several countries worldwide, and South Africa is not an exception. It is the result of adverse economic conditions in the country, where children feel they have to resort to finding employment in order to support their families. There is a general consensus that child labour in South Africa needs to be dealt with, and this is seen via the ratification of the two most important ILO conventions that speak to child labour. The ratification of these conventions led to the development of a child labour policy for South Africa; the Child Labour Programme of Action. However, despite the existence of this policy and its implementation, child labour continues. This report looked at how the child labour policy was implemented, and to verify if the continued occurrence of child labour was due to a disconnection between the policy as it is presented on paper, and those who are supposed to implement the policy on the ground. The report used qualitative methods, specifically documentary analysis and interviews. The documents that were analysed included the Child Labour Programme of Action, as well as its follow up documents. Meanwhile, government officials as well as members of civil society were the interview subjects. The main finding of the research is that the implementers of the policy do not have a full understanding of the policy and what it entails. And because of this they are unable to properly implement the policy to produce an effective solution against child labour. The report concludes with recommendations intended to help the government to get more out of the Child Labour Programme of Action.MT201

    Formulation and evaluation of polymeric micelles for improved oral delivery of tenofovir disoproxil fumarate and zidovudine using poly-lactic-co-glycolic acid nanoparticles

    Magister Pharmaceuticae - MPharmBackground: Tenofovir disoproxil fumarate (TDF) and Zidovudine (AZT) are both nucleotide and nucleoside analogue reverse transcriptase inhibitors (NtRTIs and NRTIs), respectively. They are used for the management and prevention of the Human Immunodeficiency Virus (HIV) infection. These drugs are faced with oral delivery challenges such as low intestinal permeability and extensive first pass liver metabolism for TDF and AZT, respectively. Their use may also be limited by dose-dependent adverse effects, which may result in treatment failure when patients become non-compliant and non-adherent to their prescribed antiretroviral (ARV) regimen. Non-compliance and non-adherence to ARV regimen may lead to drug resistance and a need for change in regimen, which can be very expensive, not only financially but in terms of morbidity and mortality. To solve such issues, a new drug can be formulated, or an existing drug can be modified. The development and formulation of a new drug is time consuming and expensive, especially with no available data and a high probability of failure. Modifying existing drugs is a cheaper, less time-consuming option with lower probability of failure. Such modification can be achieved via non-covalent interactions using various methods such as preparation of nano-particulates with polymeric micelles (a non-covalent interaction). Polymeric micelles offer a variety of polymers to choose from for drug modification purposes. Purpose: The aim of this study was to formulate polymeric nanoparticles of TDF and AZT using different ratios of poly-lactic-co-glycolic acid (PLGA), characterize the formulated nanoparticles (using the following analyses: particle size, zeta potential, encapsulation efficiency, hot stage microscopy, thermogravimetric analysis, differential scanning calorimetry, Fourier transform infrared spectroscopy and scanning electron microscopy), analyze for stability during storage (2-8˚C) and determine the release rate of the active pharmaceutical ingredients in the formulated nanoparticles. Methods: Nanoparticles were prepared using a modified version of the double emulsion (water-in-oil-in-water) solvent evaporation and diffusion method. Two ratios of PLGA (50:50 and 85:15) were used to prepare four formulations (two each of TDF and AZT). Thereafter, the physicochemical and pharmaceutical properties of the formulations were assessed by characterizing the nanoparticles for particle size, zeta potential, polydispersity index, percentage yield, release profile and particle morphology, using the suggested analytical techniques. Results: For TDF-PLGA 85:15, TDF-PLGA 50:50, AZT-PLGA 85:15 and AZT-PLGA 50:50, nanoparticles of 160.4±1.7 nm,154.3±3.1 nm,127.0±2.32 nm and 153.2±4.3 nm, respectively, were recovered after washing. The polydispersity index (PDI) values were ≤0.418±0.004 after washing, indicating that the formulations were monodispersed. The zeta potential of the particles was -5.72±1 mV, -19.1 mV, -12.2±0.6 mV and -15.3±0.5 mV for TDF-PLGA 85:15, TDF-PLGA 50:50, AZT-PLGA 85:15 and AZT-PLGA 50:50 respectively after washing. The highest percentage yield was calculated to be 79.14% and the highest encapsulation efficiency obtained was 73.82% for AZT-PLGA 50:50, while the particle morphology showed spherical nanoparticles with signs of coalescence and aggregation for all formulated nanoparticles. The release profiles were biphasic; that is, an initial burst which indicated the presence of surface API followed by sustained release. Comparing the release profiles of AZT and TDF at pH 1.2 and 7.4, it was indicative that more AZT was released at pH 1.2 while more TDF was released at pH 7.4. On computing the release data further into various mathematical models, the Weibull model was found to be the best fit. The loaded nanoparticles showed an increase in stability after washing; however, they showed signs of gradual decrease in stability after 10 days of storage at 2-8°C. Conclusions: Relatively small, spherical and smooth nanoparticles were formulated. The nanoparticle release profile was indicative of sustained release; however, there was no conclusive indication that 48 hours duration was sufficient to release all encapsulated drug. Further studies with an increased API or polymer ratio in the formulation needs to be performed to determine if the encapsulation efficiency can be improved and in-vivo studies are required for a better understanding of the API release from formulations as well as its absorption in the body

    Identification of an Appropriate Formulation for Domestic Water Ceramic Filters from Soukamna Clay (Cameroon)

    This paper deals with the formulation of ceramic filters having the porosity adapted to domestic potable water treatment. The filters were made from clays and rice husk obtained from the Far North region of Cameroon (Logone Valley). Nine formulations were investigated to choose those that might have the porosity standing between 35 and 50% (the ideal porosity adapted for water treatment) [1]. The nine formulations investigated were as follow: clay:rice husk mixture weight ratio 0.7:0.3; 0.8:0.2 and 0.9:0.1 with the particle size of 100:100 microns. The sintering temperatures of 900°C, 950°C and 1000°C were applied for each of the mixtures. The results showed that only filters with weight ratio 0.7:0.3 sintered at 900°C, 950°C and 1000°C had porosity between 35 and 50% with values of 39.41±0.96; 40.15±1.59; 40.14±1.31 respectively. Mechanical strength, permeability and iron leaching behavior were investigated for these three formulations. The formulation 0.7:0.3 with sintering temperature of 1000°C had the higher permeability and was the more stable for iron leaching so it is the more adapted for water treatment in terms of flow rate and iron leaching behavior, pore size distribution showed that these filters were macroporous and designed for microfiltration with average pore diameter of 0.46µm

    Volcanic Ash from the 1999 Eruption of Mount Cameroon Volcano: Characterization and Implications to Health Hazards

    Volcanic ash from the 1999 eruption of Mount Cameroon volcano has been characterized for its particle size and shape (by scanning electron microscopy, SEM), and mineralogy (by X-ray diffractometry, XRD). Also the total fluorine (F) content of the ash was determined by the selective ion electrode method. The results show that the Mount Cameroon ash particles have a variety of shapes including fibrous, rounded, subrounded, irregular, angular, eleongated and bladed. All the ash samples have a significant proportion (~ 30%) of ash < 4 ƒÊm in size and thisis classified in occupational medicine as 'thoracic' and 'respirable' ash that is considered harmful to health. The XRD patterns show that the ash contains plagioclase feldspar, enstatite, augite and chromite, which, if fine enough may cause irritation of the respiratory tract, but they are relatively insoluble in the alveolar region. The ash lacks free silica, the main mineral in volcanic ash responsible for causing silicosis. The F concentration in the ash ranges from 46 ƒÊg/ g to 189ƒÊg/g. This is high considering that the lethal dose of F is set internationally at ~ 100ƒÊg/g. This study forms the basis for a long term monitoring of volcanic ash risk and possible mitigation measures of the Mount Cameroon volcano.Keywords: volcanic ash, Mt. Tous les echantillons de cendreont une proportion significative de (~ 30 %) de cendre ayant la taille < 4 m et ceci est classifie dans la medecine du travail comme la cendre 'thoracique' et 'respirable' qui est consideree nocif a la sante. Les modeles de XRD montre que la cendre contient le feldspath de plagioclase, l'enstatite, l'augite et la chromite qui, s'ils sont assez fins, peuvent produire l'irritation de la cavite nasale mais sont relativement insolubles dans la region d'aveolaire. La cendre manque de la silice libre, qui est le minerai principal de la cendre volcanique responsable d'engendrer la silicose. La concentration de F dans la cendre s'echelonne de 189 g/g. Ceci est eleve considerant que la dose mortelle de F est place internationalement aux environs de ~ 100 g/g.
