    Sequence analysis of the non-coding control region of John Cunningham virus isolates from patients with multiple sclerosis treated with natalizumab

    Introduction. The John Cunningham virus (JCPyV) causes a fatal demyelinating disease of the central nervous system known as progressive multifocal leukoencephalopathy (PML). In healthy people, the JCPyV non-coding control region (NCCR) is not rearranged, while NCCRs in immunocompromised patients are characterized by frequent rearrangements and can be associated with PML development. Therefore, patients treated with natalizumab, which decreases the migration of leukocytes and monocytes through the blood-brain barrier to inflammatory foci, are at increased risk of developing PML. The purpose of the study was to analyze NCCR sequences of JCPyV isolates from patients with multiple sclerosis (MS) treated with natalizumab. Materials and methods. A total of 26 blood plasma samples and 8 cerebrospinal fluid samples were analyzed using nested PCR to study the JCPyV NCCR structure in Russian MS patients treated with natalizumab. The NCCRs present in the samples were cloned and sequenced by Sanger sequencing. All the JCPyV NCCR sequences were compared with the archetype sequence and mapped. The NCCR sequences were also examined for presence of putative transcription factor binding sites. Results. A total of 48 NCCR sequences were found. The analysis showed that up to 55% of NCCRs were identified as rearranged NCCRs, while the other were archetype-like NCCRs. All the sequences can be divided into 6 types with one dominant rearrangement pattern. This rearranged NCCR was also found in a patient with the confirmed PML diagnosis and a poor prognosis. All the rearranged NCCRs were characterized by the presence of additional transcription factor binding sites. Conclusion. The study has helped identify previously unknown NCCR patterns typical of MS patients treated with natalizumab in Russia, thus confirming the need for the further research on NCCR rearrangements in MS patients undergoing natalizumab treatment to gain better understanding of the origin of neurovirulent JCPyV variants

    Методом плазмохимического синтеза по схеме плазменной переконденсации в низкотемпературной азотной плазме из механической смеси металлического молибдена и мочевины получен нанокристаллический порошок оксикарбида молибдена со структурой типа NaCl, который был аттестован методами рентгенографии, растровой электронной микроскопии и энргодисперсионного анализа.A nanocrystalline powder of oxycarbide molybdenum with a NaCl structure was obtained by the method of plasma-chemical synthesis according to the plasma-recondensation scheme in a low-temperature nitrogen plasma from the mechanical mixture of metallic molybdenum and urea, which was certified by X-ray diffraction, scanning electron microscopy, and energy-dispersive analysis.Авторы выражают благодарность канд. техн. наук Э.     Методом плазменной переконденсации в низкотемпературной азотной плазме были переработаны механические смеси микрокристаллических порошков TiC–Co и TiN–Co. В ходе рентгенографических и электронно-микроскопических исследований, в том числе с использованием методик EDX-анализа, было установлено, что нанокристаллические композиции имеют радиально-слоевую структуру, состоящую из тугоплавкого ядра и металлической оболочки, содержащей кобальт, молибден или их взаимные твердые растворы.Mechanical mixtures of microcrystalline TiC–Co and TiN–Co powders were processed by plasma recondensation in a low-temperature nitrogen plasma. It was found that during radiographic and electron microscopic studies, including using EDX analysis techniques, nanocrystalline compositions have a radial layer structure consisting of a refractory core and a metallic shell containing cobalt, molybdenum or their mutual solid solutions.Авторы выражают благодарность канд. техн. наук Э.     Экстремальным воздействием на механическую смесь карбида титана с металлическим молибденом в рамках плазмохимического синтеза по схеме плазменной переконденсации в низкотемпературной азотной плазме получены нанокристаллические композиции, содержащие в своем составе сложный титан-молибденовый карбид Ti1-xMoxC. Аттестация нанопорошков производилась методами рентгенофазового анализа и растровой электронной микроскопии с возможностью применения энергодисперсионного анализа для определения химического состава продуктов синтеза.Nanocrystalline compositions containing Ti1-xMoxC complex titaniummolybdenum carbide have been obtained with an extreme effect on the mechanical mixture of titanium carbide with metallic molybdenum within the framework of plasma-chemical synthesis by the plasma recondensation scheme in a low-temperature nitrogen plasma. Certification of nanopowders was carried out using X-ray phase analysis and scanning electron microscopy with the possibility of using energy-dispersive analysis to determine the chemical composition of the synthesis products.Авторы выражают благодарность к.т.н.     В процессе плазменной переконденсации в низкотемпературной азотной плазме нанокристаллических композиий TiC–Co и TiN–Co, обогащенных металлическим молибденом, зафиксировано присутствие метастабильных сложнолегированных по металлической подрешетке нитридов Ti0,7Co0,3N и Mo0,8Co0,2N0,9 гексагональной модификации, обеспечивающих существование «core-shell»-структуры. Методом рентгенографии показано, что указанные соединения находятся в вынужденно ориентированном состоянии, обусловленном высокими степенями деформации кристаллической решетки вследствие значительной экстремальности синтеза.The presence of metastable Ti0,7Co0,3N and Mo0,8Co0,2N0,9 nitride hexagonal modifications, ensuring the existence of a «core-shell» structure, was detected in the process of plasma recondensation in a low-temperature nitrogen plasma of nanocrystalline TiC–Co and TiN–Co, enriched with metallic molybdenum. It was shown by X-ray diffraction that these compounds are in the forced-oriented state, due to high degrees of deformation of the crystal lattice due to the considerable extremality of the synthesis.Авторы выражают благодарность к.т.н.     Amyloid-b peptide on sialyl-LewisX-selectin-mediated membrane tether mechanics at the cerebral endothelial cell surface

    Increased deposition of amyloid-b peptide (Ab) at the cerebral endothelial cell (CEC) surface has been implicated in enhancement of transmigration of monocytes across the brain blood barrier (BBB) in Alzheimer’s disease (AD). In this study, quantitative immunofluorescence microscopy (QIM) and atomic force microscopy (AFM) with cantilevers biofunctionalized by sialyl-Lewisx (sLex) were employed to investigate Ab-altered mechanics of membrane tethers formed by bonding between sLex and p-selectin at the CEC surface, the initial mechanical step governing the transmigration of monocytes. QIM results indicated the ability for Ab to increase p-selectin expression at the cell surface and promote actin polymerization in both bEND3 cells (immortalized mouse CECs) and human primary CECs. AFM data also showed the ability for Ab to increase cell stiffness and adhesion probability in bEND3 cells. On the contrary, Ab lowered the overall force of membrane tether formation (Fmtf), and produced a bimodal population of Fmtf, suggesting subcellular mechanical alterations in membrane tethering. The lower Fmtf population was similar to the results obtained from cells treated with an F-actin-disrupting drug, latrunculin A. Indeed, AFM results also showed that both Ab and latrunculin A decreased membrane stiffness, suggesting a lower membrane-cytoskeleton adhesion, a factor resulting in lower Fmtf. In addition, these cerebral endothelial alterations induced by Ab were abrogated by lovastatin, consistent with its anti-inflammatory effects. In sum, these results demonstrated the ability for Ab to enhance p-selectin expression at the CEC surface and induce cytoskeleton reorganization, which in turn, resulted in changes in membrane-cytoskeleton adhesion and membrane tethering, mechanical factors important in transmigration of monocytes through the BBB.This work was supported by Alzheimer Association Grant NIRG-06-24448; NIH Grant 1P01 AG18357, R21NS052385, 5R21AG032579 and in part by 1P01HL095486 and AHA 0835676N; ‘‘Bolashak’’ scholarship and Ministry of Education and Science of the Republic of Kazakhstan 1029/GF2.
