    Nitric Oxide Synthase Activity and its Modulation in the Treatment of Colorectal Cancer

    The American Cancer Society estimates more than 141,000 new cases of and about 50,000 deaths from colorectal cancer every year. Treatment options include surgery, radiation therapy and targeted therapies such as anti-angiogenics. However, no therapies address the key driving factor of colorectal cancer: inflammation. It is well known that chronic inflammatory conditions such as Crohn’s Disease, ulcerative colitis, diabetes, obesity and cigarette smoking all elevate the risk of developing colorectal cancer. One of the hallmarks of chronic inflammation is the elevated levels of reactive oxygen/nitrogen species (ROS/RNS). A primary source of these ROS/RNS is uncoupled Nitric Oxide Synthase (NOS). Under non-inflammatory conditions NOS generates Nitric Oxide. However, in an inflammatory environment, such as the oxidative tumor microenvironment, NOS’s cofactor tetrahydrobiopterin (BH4) is oxidized to dihydrobiopterin (BH2). NOS bound to BH2 is said to be uncoupled and produces superoxide O2-and peroxynitrite (ONOO-). Previous work in our and other’s labs have shown that increased production of ROS/RNS leads to the activation of pro-inflammatory/proliferative molecules such as NFÎșB, Stat3, ÎČ-Catenin and Akt. NOS can be re-coupled by supplementing cells and animals with BH4 or its precursor Sepiapterin (SP). Herein we show that recoupling NOS with SP in HCT116, Caco-2 and HT29 cells, decreased tumor cell proliferation, increased ÎČ-Catenin degradation and decreased Akt activity. We also see increased tumor cell death measured by in vitro clonogenic assay, as well as decreased metabolic uptake in Azoxymethane/Dextran Sodium Sulfate (AOM/DSS) induced colorectal cancer in vivo measured by [18F]-fluorodeoxyglucose ([18F]-FDG) positron emitted topography (PET) imaging. We believe by recoupling NOS both in vivo and in vitro we are modulating Wnt signaling via Akt and GSK-3ÎČ. Lastly, we conducted studies to determine a mechanistic explanation of how tumor cells maintain a decreased BH4:BH2 ratio

    Real time tracking using nature-inspired algorithms

    This thesis investigates the core difficulties in the tracking field of computer vision. The aim is to develop a suitable tuning free optimisation strategy so that a real time tracking could be achieved. The population and multi-solution based approaches have been applied first to analyse the convergence behaviours in the evolutionary test cases. The aim is to identify the core misconceptions in the manner the search characteristics of particles are defined in the literature. A general perception in the scientific community is that the particle based methods are not suitable for the real time applications. This thesis improves the convergence properties of particles by a novel scale free correlation approach. By altering the fundamental definition of a particle and by avoiding the nostalgic operations the tracking was expedited to a rate of 250 FPS. There is a reasonable amount of similarity between the tracking landscapes and the ones generated by three dimensional evolutionary test cases. Several experimental studies are conducted that compares the performances of the novel optimisation to the ones observed with the swarming methods. It is therefore concluded that the modified particle behaviour outclassed the traditional approaches by huge margins in almost every test scenario

    Magnetic resonance imaging (MRI7) is a very powerful imaging technique for the assessment of stroke aetiology (Condition) and brain imaging. Another class of MRI is ultrahigh frequency based MRI using 7 Tesla is now developed by seamen’s for better imaging in humans. This study examines these MRI. Thisarticle highlights an alternative approach, denoted “interval monitoring,” whose aims is related with more timely detection of tumor cancer changes. The conceptual background and the computational realization of the proposed method are outlined, and its application is illustrated by an empirical example from the image-based photo science, cancer registry of America. Monitoring of cancer patient survival is the first step of its cure so across the globe practice routinely employed by many cancer registries, which is an essential component for its cure. However, changes in prognosis over time are disclosed withconsiderable delay, with traditional methods of monitoring cumulative survival. Our study took sequence of MRI images, GMPLS function locate the cancer after filtering and skeletonization. This study saves time and difference for calculation of cancer equation. This study uses statistical technique to get the desired matrix, further its inverse provides us real time mathematical equation which is unique for each patient. Further survivor analysis is employed to achieve the break or death of subject. The Aim of this research is to provide unique mathematical model of a cancer patient, provides real time graph aboutcancer health and survivor function depicts the death of subject respectively

    ABSTRACT: The Sankhaholi (Evolvulus alsinoides Linn.) of family  Convolvulaceae which is commonly known as sankhpushpi, in traditional system of medicine including Unani Medicine. It is a perennial herb with a small woody branched root stock which contains alkaloids: shankhapushpine and evolvine. Fresh plant of sankhholi contains volatile oil. It also contains a yellow neutral fat, an organic acid and saline substances. Therapeutic uses of Sankhaholi Evolvulus alsinoides Linn, mentioned in the Unani Medicine are Alexiteric (Mufarreh), Cardiac  tonic  (Muqawwi-e Qalb), Brain tonic  (Muqawwi-e Dimag), Digestive (Hazim)  Musaffi-e- Khoon (Blood purifier),General tonic (Muqawwi-e-am), Diuretic (Mudirr-e-Baul), Anti Inflammatory (Muhallil-e-waram), Hypoglycemic (Dafa-e-Ziabitus), Antihypertensive (Dafye Imtella). It is also used in headache, asthma, hyperlipidemia etc. The present article reviews the pharmacological actions and therapeutic uses of Sankhaholi ( Evolvulus alsinoides Linn.) present in Unani literature supported with the available clinical and animal studies.Keywords: Sankhaholi Evolvulus alsinoides Linn.     Majoon-E-Piyaz: A Potent Unani Formulation for Premature Ejaculation

    SurÊ»at-e-Inzāl (Premature Ejaculation) is the most prevalent male sexual dysfunction affecting 25 - 40% global population of men. It is a universal disorder and is independent of age, social or marital status. It has a significant impact on both- patients and their partners, causing distress, anxiety and relationship difficulties affecting the quality of life. Several aetiologies have been proposed by various researchers which are not evidence-based but speculative. Accordingly the International Society for Sexual Medicine (ISSM) issued treatment guidelines for Premature Ejaculation (PE) recommending Serotonergic Antidepressants (SSRIs) and Local Anaesthetics (LA) for its management in modern medicine. However, these treatments were not actually developed for PE, and have limitations associated with their off-label use. Furthermore, nearly all the recommended drugs have a wider spectrum of adverse effects and serious drug interactions which sometimes could be fatal. On the other hand, centuries old Unani medicine offers a complete line of treatment for SurÊ»at-e-Inzāl based on traditional knowledge and experience. Unani physicians devised a large number of poly-herbal recipes which are still in vogue.  Majoon-e-Piyaz (MP) is one of the compound Unani formulations which are in use for the treatment of premature ejaculation since centuries.     Assessment of Wild Edible Fruits Consumed through the Tribal People of Chittagong Hill Tracts (CHTs), Bangladesh

    The present study was conducted with the ethno-botanical survey, documentation and future potentialities of the wild edible fruits used through the tribal people of Chittagong Hill Tracts (CHTs). The wild edible fruits play an important role in meeting the food and nutritionneeds of the tribal people living in the CHTs. This paper deals with documentation of 49 types of wild edible fruits plant speciesbelonging to 25 families used by the tribal people of CHTs. Their botanical names, families, local name, habit and fruit ripening time are discussed. Suggestions have been made to improve the quality (variety) of wild fruits through domestication, hybridization, grafting and also for their chemical analysis for improved food values and economic properties. Attempts should be taken to protect the wild edibles fruit species in the vanishing from natural forests. Also the general public should be encouraged to cultivate and use these wild fruits in addition to the conventional fruits by improving the varieties and nutritional value of the wild fruits through research
