Adiponectin and Selenium Rich Diet can act as a Complimentary Medicine in the Treatment of Intestinal and Chronic Inflammation Induced Colon Cancer
Colon cancer is the second largest cause of cancer death in United States. Chronic inflammation and obesity predispose patients to colon cancer. Adipose tissue is a source of bioactive substances called adipokines. Adiponectin (APN), an adipokine has anti-inflammatory property and found at lower levels in obese patients. Selenium (Se), a trace mineral and a dietary supplement, is inversely associated with cancer risk and possess anti-inflammatory and anti-carcinogenic properties. The overall purpose of this dissertation is to determine if chronic inflammation leading to colon and intestinal cancer are regulated by APN or Se rich diet or both. The working hypothesis is that APN deficiency will decrease goblet cell mucous production in colon leading to greater chronic inflammation and exacerbate the clinical symptoms and tumor load related to colon cancer. Se rich diet alone or in combination with APN administration will increase goblet cell production and apoptosis of cancer cells leading to reduced clinical symptoms, tumor load and inflammation. The specific aim 1 studied the role of APN deficiency in chronic inflammation induced colon cancer (CICC) and its effect of goblet cell production. Absence of APN increased the severity of CICC and its administration on goblet cell lines decrease their apoptosis with increase Math-1 production and upregulated mucin (Muc-2) secretion through the activation of its receptors APN R1 and R2. Specific aim 2 was designed to study the effect of Se rich diet and APN deficiency in positive modulation of CICC. Our result indicated a sharp decline in the physical manifestations of colon cancer with Se rich diet and higher severity of CICC with APN deficiency providing another proof for the protective role of APN in CICC. Specific aim 3 studied the effect of APN administration or Se rich diet or both on intestinal cancer. We found a protective effect of Se, APN and both in reducing the clinical score and tumor load of intestinal cancer. Several mechanism of action of both Se and APN were studied in all the 3 aims. In conclusion, APN and Se could be used as a complimentary medicine in the treatment of colon and intestinal cancer
Vaccine Approaches for Pandemic COVID-19: An Overview
The coronavirus strain, SARS-CoV-2 causative agent of the pandemic viral pneumonia disease COVID-19, identified in the late 2019. Unfortunately, there is no vaccine or therapeutics available to control the disease. Although, earlier research efforts of SARS pandemic (2003) provides the precious information to the researchers for fast track vaccine development. Research efforts for vaccine development of COVID-19 pandemic are unprecedented in terms of promising time period. The inactivated whole virus-based SARS-CoV-2 vaccine is first one developed and entered into clinical trial. However, later on after announce of complete genome sequence of SARS-CoV-2, vaccine based on recombinant viral vector, nucleic acid and immunogenic fragment also introduced. Previous experience of undesired immunopotentiation in SARS-CoV studies represents, vaccine safety should be the main concern. Researchers believe that next generation vaccine approaches using recombinant technology such as viral vector or subunit protein-based vaccine will be more effective and safer. Novel cost-effective design for bio-volatilization studies in photosynthetic microalgae exposed to arsenic with emphasis on growth and glutathione modulation
A novel laboratory model was designed to study the arsenic (As) biotransformation potential of the microalgae Chlorella vulgaris and Nannochloropsis sp. and the cyanobacterium Anabaena doliolum. The Algae were treated under different concentrations of As(III) to check their growth, toxicity optimization, and volatilization potential. The results revealed that the alga Nannochloropsis sp. was better adopted in term of growth rate and biomass than C. vulgaris and A. doliolum. Algae grown under an As(III) environment can tolerate up to 200 μM As(III) with moderate toxicity impact. Further, the present study revealed the biotransformation capacity of the algae A. doliolum, Nannochloropsis sp., and Chlorella vulgaris. The microalga Nannochloropsis sp. volatilized a large maximum amount of As (4,393 ng), followed by C. vulgaris (4382.75 ng) and A. doliolum (2687.21 ng) after 21 days. The present study showed that As(III) stressed algae-conferred resistance and provided tolerance through high production of glutathione content and As-GSH chemistry inside cells. Thus, the biotransformation potential of algae may contribute to As reduction, biogeochemistry, and detoxification at a large scale
Major phenolic compounds, antioxidant, antimicrobial, and cytotoxic activities of Selinum carvifolia (L.) collected from different altitudes in India
Antibiotic resistance poses a serious threat to public health, raising the number of diseases in the community. Recent research has shown that plant-derived phenolic compounds have strong antimicrobial, antifungal, and cytotoxic properties against a variety of microorganisms and work as great antioxidants in such treatments. The goal of the current work is to evaluate the anticancerous, antibacterial, antifungal, antioxidant, and cytotoxicity activities in the extracts of the different plant parts (leaves, stems, and roots) of S. carvifolia (L.) L. This is a medicinally important plant and has been used for different kinds of diseases and ailments such as hysteria and seizures. The phenolic compounds from the different plant parts were analyzed using HPLC and the following were found to be present: chlorogenic acid, gallic acid, rutin, syringic acid, vanillic acid, cinnamic acid, caffeic acid, and protocatechuic acid. Gallic acid was found to have the highest concentration (13.93 mg/g), while chlorogenic acid (0.25 mg/g) had the lowest. The maximum TPC value, which ranged from 33.79 to 57.95 mg GAE/g dry extract weight, was found in the stem. Root extract with 9.4 mg RE/g had the greatest TFC level. In the leaf and stem extracts, the RSC ranged from 0.747 mg/mL to 0.734 mg/1 mL GE/g dry extract weight, respectively. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay was used to measure in vitro antioxidant activity. In a concentration-dependent way, promising antioxidant activity was reported. Moreover, 3,5-dinitrosalicylic acid (DNSA) and the Folin–Ciocalteu phenol reagent technique were used to determine reducing sugar content and total phenolic content, respectively. Antibacterial activity against eight strains (MIC: 250–1,000 μg/mL) was analyzed, and the stem extract exhibited maximum activity. Antifungal activity was also assessed, and potent activity was reported especially in the extract obtained from the stem. Cytotoxicity was evaluated using an MTT assay in the A549 cell line, where different doses (0.0625, 0.125, 0.25, 0.5, and 1 mg/mL) of leaf, root, and stem extracts were used. Treatment with these extracts reduced the cell viability, indicating that S. carvifolia may possess anticancer potential, which can be of great therapeutic value
Effect of exercise on chemically-induced colitis in adiponectin deficient mice
<p>Abstract</p> <p>Background</p> <p>Inflammatory bowel diseases are associated with increased adiponectin (APN) levels, which may exert pro-inflammatory effects in these individuals. Since habitual exercise may increase APN, the aim of this study was to determine how exercise training affects mice with acute colitis.</p> <p>Methods</p> <p>Male adiponectin knock out (APNKO) and wild type (WT) mice (C57BL/6) were randomly assigned to 4 different groups: 1) Sedentary (SED); 2) Exercise trained (ET); 3) Sedentary with dextran sodium sulfate (DSS) treatment (SED + DSS); and 4) Exercise trained with DSS (ET + DSS). Exercise-trained mice ran at 18 m/min for 60 min, 5d/wk for 4 weeks. Subsequently, the ET + DSS and the SED + DSS mice received 2% DSS in their drinking water for 5 days (d), followed by 5d of regular water.</p> <p>Results</p> <p>The clinical symptoms of acute colitis (diarrhea, stool haemoccult, and weight loss) were unaffected by exercise and there was no difference between the APNKO and WT mice (p > 0.05) except on day 39. However, the clinical symptoms of the DSS-treated APNKO mice were worse than WT mice treated with DSS and had increased susceptibility to intestinal inflammation due to increased local STAT3 activation, higher IL-6, TNF-α, IL-1β and IL-10 levels, and as a result had increased intestinal epithelial cell proliferation (p < 0.05). Exercise training significantly decreased pro-inflammatory cytokines including IL-6, TNF-α and IL-1β (p < 0.05) in the DSS + EX APNKO mice but had no effect on epithelial cell proliferation. Exercise was also found to significantly decrease the phosphorylation expression of STAT3 in both WT and APNKO mice in DSS + EX group when compared to DSS + SED.</p> <p>Conclusions</p> <p>Exercise training may contribute in alleviating the symptoms of acute colitis and APN deficiency may exacerbate the intestinal inflammation in DSS-induced colitis.</p
Interspecific and intraspecific analysis of Selinum spp. collected from Indian Himalayas using DNA barcoding
Abstract Background DNA barcoding is a powerful method for phylogenetic mapping and species identification. However, recent research has come to a consistent conclusion about the universality of DNA barcoding. We used matK and rbcL markers to test the universality of twelve accessions from different locations belonging to two Selinum species, Selinum tenuifolium Wall. C. B. Clarke and Selinum vaginatum C. B. Clarke, keeping in mind their ability to identify species and establish phylogenetic relationships within and between the accessions. Results The success rates of PCR amplification using matK and rbcL were 75.26% ± 3.65% and 57.24% ± 4.42%, and the rate of DNA sequencing was 63.84% ± 4.32% and 50.82% ± 4.36%, respectively, suggesting that success rates of species identification of the two fragments were higher than 41.00% (matK, 41.50% ± 2.81%; rbcL, 42.88% ± 2.59%), proving that these fragments might be used to identify species. The best evolutionary tree with good supporting values was produced utilizing combinations of matK + rbcL markers when phylogenetic relationships were built with random fragment combinations. The twelve accessions of Selinum collected from different locations and their molecular sequences of matK and rbcL markers were blasted with other genera of Apiaceae family, and it was found that Selinum is most closely related to Angelica species of Apiaceae family. Conclusion The present study has grouped twelve accessions of Selinum species using molecular markers into phylogenies, which is first-of-its-kind report that established interrelationships within different species of Apiaceae with respect to Selinum. Dietary Agents and Phytochemicals in the Prevention and Treatment of Experimental Ulcerative Colitis
Inflammatory bowel diseases (IBDs), consisting mainly of ulcerative colitis (UC) and Crohn's disease (CD), are important immune-mediated diseases of the gastrointestinal tract. The etiology of the disease includes environmental and genetic factors. Its management presents a constant challenge for gastroenterologists and conventional surgeon. 5-Amninosalicylates, antibiotics, steroids, and immune modulators have been used to reduce the symptoms and for maintenance of remission. Unfortunately, long-term usage of these agents has been found to lead to severe toxicities, which are deterrent to the users. Pre-clinical studies carried out in the recent past have shown that certain dietary agents, spices, oils, and dietary phytochemicals that are consumed regularly possess beneficial effects in preventing/ameliorating UC. For the first time, this review addresses the use of these dietary agents and spices in the treatment and prevention of IBD and also emphasizes on the mechanisms responsible for their effects
