Viabilitas Spermatozoa Semen Beku Babi Duroc dalam Extender Beltsville Thawing Solution Menggunakan Krioprotektan Gliserol dan Dimetillacetamida
This study investigated the effect of glycerol and dimethylacetamide (DMA) in beltsville thawing solution (BTS) extender. Semen samples were collected from three boars of the Duroc breed (n=5) and were evaluated microscopically and macroscopically. Semen that have been evaluated and meets the following criteria: has motility characteristics >70%, concentration >300 million/ml and abnormality 20%; was accepted as appropriate samples. Semen samples were divided into four tubes and were diluted with 5 ml of BTS. The diluted semen was left for two hours in the temperature range from 20 to 22°C before was centrifuged for 15 minutes (500G). The pellet of centrifuged semen was taken as much as 1 ml with its supernatant. The BTS glycerol (BTSG), BTS-DMA (BTSD), BTS-Glycerol-DMA (BTSG-D) were added into the pellet. The diluted semen was packed into straw of 0.5 ml and was equilibrated for 2 hours in 4 °C. The equilibrated semen was frost with the steam of nitrogen and stored in liquid nitrogen. The quality of frozen semen was assessed at least 24 hour after freezing process. The results indicate that the sperm motility after thawing in BTS extender BTSD is 52.14±0.4%, ,higher than the other two extenders (P<0.05). Thus, it is conclude that the DMA concentration in BTS extender maintains the quality of frozen semen from the Duroc breed better than other extenders
Raw Semen Characteristics of Three Different Indonesian Local Roosters
Indonesia has agreat variety of roosters, either indigenous type as well as exotic and cross breed. The purpose of this experiment was to study the characteristics of semen from three types of Indonesian local roosters such as Merawang, Kampung and crosses Sentul Kampung with Kedu (SK Kedu). A total of 15 roosters consist of Merawang roosters, Kampung, and SK Kedu roosters were 5 each. The semen was collected 3 times a week by dorso-abdominal and cloaca massage method. The parameters evaluation was macroscopic characteristics consist of volume, color, consistency, and pH. Microscopic evaluation of semen such as a mass movement, sperm motility, live sperm, sperm abnormality and sperm concentration. Results of this experiment showed that semen volume of Merawang (0.40±0.26 mL) was higher (p<0.05) compare to Kampung (0.24±0.12 mL) or SK Kedu (0.16±0.10 mL) but no difference on semen color, consistency and semen pH. There were no difference in the mass movement, sperm motility and live sperm as well as on sperm abnormality among three types of roosters. Sperm concentration of Merawang (4490 million mL-1) was significantly higher than Kampung (3245 million mL-1) and the SK Kedu roosters (3751 million mL-1). KUALITAS SEMEN DOMBA LOKAL DARI FREKUENSI EJAKULASI BERBEDA (The quality of local ram semen from different ejaculation)
One of the factors that support the success of reproduction in sheep is the quality of the semen. This study aims to analyze the effect of ejaculation frequency on the quality of ram semen. This study used four local rams two years old. All rams have collected the semen using an artificial vagina five times a day with three replications. The experimental parameters were macroscopic (volume, pH, consistency, and color) and microscopic (mass movement, motility, viability, concentration, normal sperm morphology, and sperm intact plasma membrane). The data analysis used a randomized block design (RBD) with SPSS version 20. The results showed that the frequency of ejaculation in rams shows color and consistency in the 1st ejaculate, which was creamier and thicker than the 4th and 5th ejaculate. Mass movement, sperm concentration per mL, and total sperm concentration in ejaculate 1st and 2nd were higher than in 5th ejaculate. The potential of artificial insemination (AI) in the 1st and 2nd ejaculate was higher than in the 5th ejaculate. The study concluded that the frequency of ejaculation affects mass movement, sperm concentration, and the potential straw production for AI in local sheep
The use of transrectal massage combined with artificial vagina as semen collection in javan banteng (
Semen collection in wild animals may be challenging for some methods such as electro ejaculator may cause pain and distress to the animals. Transrectal massage has been widely used in domestic and wild animals semen collection. Even though an artificial vagina may provide a condition that mimics the female reproductive tract and known to provide better results in some species, the application may face obstacles in the field. This study was conducted to evaluate the possibility of modified semen collection in banteng bull by combining the transrectal massage method followed by the use of an artificial vagina. The bull was habituated and trained for semen collection in chute under veterinarian supervision in Taman Safari Indonesia, Cisarua, Bogor, West Java. Semen samples from one Javan banteng bull were collected once a week. Banteng bull showed stud and penile protrusion during semen collection. Watery semen with volume ranged from 12 ml were successfully collected, with sperm progressive motility ranged from 25%-40%. Our study also showed sperm abnormalities comprising 18.11% abnormal sperm heads, 28.82% abnormal sperm tails and 0.94% teratoid form. The results demonstrated the possibility of this modified method as an alternative procedure for semen collection in banteng bull which may also be useful for other wild animals in field condition
