    The effects of refinery effluent on the invertebrate intertidal fauna and flora of Little Wick Bay, Milford Haven

    The effects of continuous low level oil pollution from a refinery effluent, on intertidal fauna and flora in Little Wick Bay are investigated. A shore survey of the intertidal species on six transects at varying distances from the effluent discharge point, is carried out and the results compared with past surveys of the same transects. Any differences or similarities in the findings are related to the environmental agencies in operation since the first survey. A detailed investigation of the size classes and abundance of the Limpet Patella vulgata at each of the six transects and a quantitative analysis of Petroleum Oil Pollutants in P. vulgata by Infra red Spectrophotometry is carried out in an attempt to relate findings to the effluent discharge

    Effect of fluid restriction on hydration status in man.

    Hydration status and the effects of hypohydration have been the topic of much public and scientific debate in recent years. While many physiological responses to hypohydration have been studied extensively, the subjective responses to hypohydration have largely been ignored. The present investigation was designed to investigate the physiological responses and subjective feelings resulting from 13, 24 and 37 h of fluid restriction (FR) and to compare these with a euhydration (EU) trial of the same duration in fifteen healthy volunteers. The volunteers were nine men and six women of mean age 30 (sd 12) years and body mass 71.5 (sd 13.4) kg. Urine and blood samples were collected and subjective feelings recorded on a 100 mm verbally anchored questionnaire at intervals throughout the investigation. In the EU trial the subjects maintained their normal diet. Body mass decreased by 2.7 (sd 0.6) % at 37 h in the FR trial and did not change significantly in the EU trial. Food intake in the FR trial (n 10) provided an estimated water intake of 487 (sd 335) ml and urinary losses (n 15) amounted to 1.37 (sd 0.39) litres. This is in comparison with an estimated water intake of 3168 (sd 1167) ml and a urinary loss of 2.76 (sd 1.11) litres in the EU trial. Plasma osmolality and angiotensin II concentrations increased from 0-37 h with FR. Plasma volume decreased linearly throughout the FR trial amounting to a 6.2 (sd 5.1) % reduction by 37 h. Thirst increased from 0-13 h of FR then did not increase further (P>0.05). The subjects reported feelings of headache during the FR trial and also that their ability to concentrate and their alertness were reduced

    Selective and flexible depletion of problematic sequences from RNA-seq libraries at the cDNA stage

    BACKGROUND A major hurdle to transcriptome profiling by deep-sequencing technologies is that abundant transcripts, such as rRNAs, can overwhelm the libraries, severely reducing transcriptome-wide coverage. Methods for depletion of such unwanted sequences typically require treatment of RNA samples prior to library preparation, are costly and not suited to unusual species and applications. Here we describe Probe-Directed Degradation (PDD), an approach that employs hybridisation to DNA oligonucleotides at the single-stranded cDNA library stage and digestion with Duplex-Specific Nuclease (DSN). RESULTS Targeting Saccharomyces cerevisiae rRNA sequences in Illumina HiSeq libraries generated by the split adapter method we show that PDD results in efficient removal of rRNA. The probes generate extended zones of depletion as a function of library insert size and the requirements for DSN cleavage. Using intact total RNA as starting material, probes can be spaced at the minimum anticipated library size minus 20 nucleotides to achieve continuous depletion. No off-target bias is detectable when comparing PDD-treated with untreated libraries. We further provide a bioinformatics tool to design suitable PDD probe sets. CONCLUSION We find that PDD is a rapid procedure that results in effective and specific depletion of unwanted sequences from deep-sequencing libraries. Because PDD acts at the cDNA stage, handling of fragile RNA samples can be minimised and it should further be feasible to remediate existing libraries. Importantly, PDD preserves the original RNA fragment boundaries as is required for nucleotide-resolution footprinting or base-cleavage studies. Finally, as PDD utilises unmodified DNA oligonucleotides it can provide a low-cost option for large-scale projects, or be flexibly customised to suit different depletion targets, sample types and organisms.This work was supported by an Australian Research Council Discovery Grant (DP130101928) and a NHMRC Senior Research Fellowship (514904) awarded to TP.     A market assessment for modern cooking in Malawi

    This paper presents the findings from a study which used household surveys and expert interviews to investigate cooking practices and understand the barriers and opportunities to the growth of the modern cooking sector in Malawi. The findings from expert interviews highlight barriers to electric and LPG cooking around the weakness of existing infrastructure, lack of consumer willingness and ability to pay and resistance to the adoption of modern cooking devices. The greatest opportunity for electric cooking is in urban areas and on mini-grids, while LPG is also most viable in urban areas, however knowledge and infrastructure gaps need to be narrowed to facilitate growth. An analysis of household surveys in urban, peri-urban and rural areas, using data from ā€œindicative cooking diariesā€, demonstrates the diversity of cooking practices in Malawian households by showing what, how, and with what, dishes are cooked. It is demonstrated that there is a latent demand for modern cooking in Malawi. Targeted research is needed to test modern cooking devicesā€™ ability to cook Malawian dishes in ways which are acceptable to Malawian people, in order to effectively accelerate a transition towards modern cooking in Malawi and address the negative health and environmental impacts of biomass cooking

    (13)C or Not (13)C: Selective Synthesis of Asymmetric Carbon-13-Labeled Platinum(II) cis-Acetylides.

    Asymmetric isotopic labeling of parallel and identical electron- or energy-transfer pathways in symmetrical molecular assemblies is an extremely challenging task owing to the inherent lack of isotopic selectivity in conventional synthetic methods. Yet, it would be a highly valuable tool in the study and control of complex light-matter interactions in molecular systems by exclusively and nonintrusively labeling one of otherwise identical reaction pathways, potentially directing charge and energy transport along a chosen path. Here we describe the first selective synthetic route to asymmetrically labeled organometallic compounds, on the example of charge-transfer platinum(II) cis-acetylide complexes. We demonstrate the selective (13)C labeling of one of two acetylide groups. We further show that such isotopic labeling successfully decouples the two Ī½(Cā‰”C) in the mid-IR region, permitting independent spectroscopic monitoring of two otherwise identical electron-transfer pathways, along the (12)Cā‰”(12)C and (13)Cā‰”(13)C coordinates. Quantum-mechanical mixing leads to intriguing complex features in the vibrational spectra of such species, which we successfully model by full-dimensional anharmonically corrected DFT calculations, despite the large size of these systems. The synthetic route developed and demonstrated herein should lead to a great diversity of asymmetric organometallic complexes inaccessible otherwise, opening up a plethora of opportunities to advance the fundamental understanding and control of light-matter interactions in molecular systems
