    208— Describing Genetic Diversity in a Non-Native Ant-Mimicking Spider

    The ant-mimicking spider Myrmarachne formicaria (Salticidae) is a species native to Eurasia and was first identified in North America in 2001. It has since been found in many locations in the Northeast including western New York, western Pennsylvania, northeastern Ohio, and southern Ontario. Little is known about its introduction to North America and how it has dispersed since. By characterizing the mitochondrial genetic diversity of this species, we can learn about its introduction history and dispersal patterns in North America. Sequencing of a 600-bp mitochondrial DNA gene region spanning the 16s rRNA, leucine tRNA, and part of the ND1 gene from 27 specimens collected from 14 localities in New York, Pennsylvania and Ohio yielded no genetic polymorphisms. Comparisons with sequence data available with GenBank for other salticid species (Myrmarachne erythrocephala and Habronattus ustulatus) have shown that within-species divergence in this mitochondrial DNA region is found in other spiders, so M. formicaria’s lack of variation is likely unusual. Our current data are consistent with a single invasion of Myrmarachne formicaria from one source locality, but data from additional loci and samples would help to confirm this conclusion

    This paper provides an account of Antonio Gramsci’s impact on the area of critical pedagogy. It indicates the Gramscian influence on the thinking of major exponents of the field. It foregrounds Gramsci's ideas and then indicates how they have been taken up by a selection of critical pedagogy exponents who were chosen on the strength of their identification and engagement with Gramsci's ideas, some of them even having written entire essays on Gramsci. The essay concludes with a discussion concerning an aspect of Gramsci's concerns, the question of powerful knowledge, which, in the present author's view, provides a formidable challenge to critical pedagogues.peer-reviewe

    Reconstructing the Wilderness: Finding Identity, Culture and Values in Filipino Children’s Literature

    In children’s literature, more specifically in the Philippine setting, the symbolism of wilderness and the elements therein constitutively influence the intrinsic wildness of children. The literary wilderness becomes an exploratory platform where the stages of development that children undergo are better understood – from exploring childhood freedom to encountering a sense of self and culture. Coming from this, the research particularly focuses on the effect of the ‘essential wildness’ on children as facilitated by the images of the wilderness and its elements in Philippine children’s literature. The notion of wilderness (or wildness) ultimately shifts the idea of losing one’s self in danger to a sense of self-discovery and individuation. To concretize and show the effects of one’s encounter with the natural world through environmental children’s literature, this study presents an ecocritical reading of the best short stories of Severino Reyes in his Lola Basyang collection in order to grasp a conception of wilderness within the context and culture of the Filipino identity. This analysis will also attempt to redirect the general understanding of the concept of inner wildness, reconstructing it from an articulation of danger and peril towards a creative exploration of one’s growth and sensibilities

    Successional change in phosphorus stoichiometry explains the inverse relationship between herbivory and lupin density on Mount St. Helens

    Background: The average nitrogen-to-phosphorus ratio (N:P) of insect herbivores is less than that of leaves, suggesting that P may mediate plant-insect interactions more often than appreciated. We investigated whether succession-related heterogeneity in N and P stoichiometry influences herbivore performance on N-fixing lupin (Lupinus lepidus) colonizing primary successional volcanic surfaces, where the abundances of several specialist lepidopteran herbivores are inversely related to lupin density and are known to alter lupin colonization dynamics. We examined larval performance in response to leaf nutritional characteristics using gelechiid and pyralid leaf-tiers, and a noctuid leaf-cutter. Methodology/Principal Findings: We conducted four studies. First, growth of larvae raised on wild-collected leaves responded positively to leaf %P and negatively to leaf carbon (%C), but there was no effect of %N or quinolizidine alkaloids (QAs). Noctuid survival was also positively related to %P. Second, we raised gelechiid larvae on greenhouse-grown lupins with factorial manipulation of competitors and soil N and P. In the presence of competition, larval mass was highest at intermediate leaf N:P and high %P. Third, survival of gelechiid larvae placed on lupins in high-density patches was greater when plant competitors were removed than on controls. Fourth, surveys of field-collected leaves in 2000, 2002, and 2003 indicated that both %P and %N were generally greater in plants from low-density areas. QAs in plants from low-density areas were equal to or higher than QAs in high-density areas. Conclusions/Significance: Our results demonstrate that declines in lupin P content under competitive conditions are associated with decreased larval growth and survival sufficient to cause the observed negative relationship between herbivore abundance and host density. The results support the theoretical finding that declines in stoichiometric resource quality (caused here by succession) have the potential to cause a decrease in consumer abundance despite very dense quantities of the resource. © 2009 Apple et al

    Highly polymorphic microsatellites in the North American snakeweed grasshopper, Hesperotettix viridis

    Microsatellite markers are preferred for fine-scale population genetic studies requiring high resolution. The grasshopper Hesperotettix viridis (Thomas) is an oligophagous species that feeds on composites and often exhibits locally restricted diets. Divergence in host plant use in some localities is seen where co-occurring subpopulations select alternate plant species, as expected with the evolution of host shifts and associated lineage divergence. To characterize the host-associated divergence patterns among populations of H. viridis, we developed markers from two microsatellite-enriched genomic libraries. Here we report the characterization and optimization of seven polymorphic di- and tri-nucleotide microsatellite loci for this species. One hundred and six individuals from 5 populations were tested for polymorphism. The number of alleles varied from 4 to 38 in all the populations. Ho ranged from 0.339 to 0.790. Homozygote excess was observed across loci, perhaps due to inbreeding. This is the first report of microsatellite markers for the subfamily Melanoplinae

    Impact of Scottish vocational qualifications on residential child care : have they fulfilled the promise?

    This article will present findings from a doctoral study exploring the impact of 'SVQ Care: Promoting Independence (level III)' within children's homes. The study focuses on the extent to which SVQs enhance practice and their function within a 'learning society'. A total of 30 staff were selected from seven children's homes in two different local authority social work departments in Scotland. Each member of staff was interviewed on four separate occasions over a period of 9 months. Interviews were structured using a combination of repertory grids and questions. Particular focus was given to the assessment process, the extent to which SVQs enhance practice and the learning experiences of staff. The findings suggest that there are considerable deficiencies both in terms of the SVQ format and the way in which children's homes are structured for the assessment of competence. Rather than address the history of failure within residential care, it appears that SVQs have enabled the status quo to be maintained whilst creating an 'illusion' of change within a learning society

    ELISA in the multiplex era: potentials and pitfalls

    Multiplex immunoassays confer several advantages over widely adopted singleplex immunoassays including increased efficiency at a reduced expense, greater output per sample volume ratios and higher throughput predicating more resolute, detailed diagnostics and facilitating personalised medicine. Nonetheless, to date, relatively few protein multiplex immunoassays have been validated for in vitro diagnostics in clinical/point-of-care settings. This review article will outline the challenges, which must be ameliorated prior to the widespread integration of multiplex immunoassays in clinical settings: (i) biomarker validation; (ii) standardisation of immunoassay design and quality control (calibration and quantification); (iii) availability, stability, specificity and cross-reactivity of reagents; (iv) assay automation and the use of validated algorithms for transformation of raw data into diagnostic results. A compendium of multiplex immunoassays applicable to in vitro diagnostics and a summary of the diagnostic products currently available commercially are included, along with an analysis of the relative states of development for each format (namely planar slide based, suspension and planar/microtitre plate based) with respect to the aforementioned issues
