Efficacy of Dexrazoxane in Preventing Anthracycline Cardiotoxicity in Breast Cancer
Abstract Objectives The authors performed a systematic review and meta-analysis of randomized and nonrandomized trials on the efficacy of dexrazoxane in patients with breast cancer who were treated with anthracyclines with or without trastuzumab. Background Breast cancer treatment with anthracyclines and trastuzumab is associated with an increased risk of cardiotoxicity. Among the various strategies to reduce the risk of cardiotoxicity, dexrazoxane is an option for primary prevention, but it is seldom used in clinical practice. Methods Online databases were searched from January 1990 up to March 1, 2019, for clinical trials on the use of dexrazoxane for the prevention of cardiotoxicity in patients with breast cancer receiving anthracyclines with or without trastuzumab. Risk ratios (RRs) with 95% confidence intervals (CIs) were calculated using a random-effects model meta-analysis. Results Seven randomized trials and 2 retrospective trials with a total of 2,177 patients were included. Dexrazoxane reduced the risk of clinical heart failure (RR: 0.19; 95% CI: 0.09 to 0.40; p Conclusions Dexrazoxane reduced the risk of clinical heart failure and cardiac events in patients with breast cancer undergoing anthracycline chemotherapy with or without trastuzumab and did not significantly impact cancer outcomes. However, the quality of available evidence is low, and further randomized trials are warranted before the systematic implementation of this therapy in clinical practice
Dispersion and collapse in stochastic velocity fields on a cylinder
The dynamics of fluid particles on cylindrical manifolds is investigated. The
velocity field is obtained by generalizing the isotropic Kraichnan ensemble,
and is therefore Gaussian and decorrelated in time. The degree of
compressibility is such that when the radius of the cylinder tends to infinity
the fluid particles separate in an explosive way. Nevertheless, when the radius
is finite the transition probability of the two-particle separation converges
to an invariant measure. This behavior is due to the large-scale
compressibility generated by the compactification of one dimension of the
space
Inhibitory effect against polymerase and ribonuclease activities of HIV-reverse transcriptase of the aqueous leaf extract of Terminalia triflora
Dichloromethane, methanol and aqueous extracts from the leaves of Terminalia triflora were investigated for their inhibitory effect on polymerase and ribonuclease activities of HIV reverse transcriptase.The most potent activity was found in the aqueous extract, which inhibited both polymerase and ribonuclease activities of the enzyme with an IC50 of 1.6 micro g/mL and 1.8 micro g/mL respectively. The antiinfective activity of the extract was demonstrated in HLT4LacZ-IIIB cell culture with an IC50 of 1.0 micro g/mL. The extract was submitted to a purification process by extractive and chromatographic methods. The activity remained in the hydrophillic fraction. Tannins present in this active purified fraction, as determined by TLC and HPLC methods, could account for the anti HIV-RT activity found in the aqueous extract
CP violation in supersymmetric model with non-degenerate A-terms
We study the CP phases of the soft supersymmetry breaking terms in
string-inspired models with non-universal trilinear couplings. We show that
such non-universality plays an important role on all CP violating processes. In
particular these new supersymmetric sources of CP violation may significantly
contribute to the observed CP phenomena in kaon physics while respecting the
severe bound on the electric dipole moment of the neutron.Comment: 14 pages, Late
Argentine plant extracts active against polymerase and ribonuclease H activities of HIV-1 reverse transcriptase
Lipophilic and hydrophilic extracts of four Argentine plants (Gamochaeta simplicaulis Cabr. 1, Achyrocline flaccida Wein. D. C. 2, Eupatorium buniifolium H. et A. 3, and Phyllanthus sellowianus Muell. Arg. 4) were examined in vitro for their ability to inhibit the polymerase and ribonuclease H (RNase H) activities of human immunodeficiency virus type 1 (HIV-1) reverse transcriptase (RT) (wild and Y181C mutant types). The active extracts were also examined as inhibitors of viral replication in HLT4LacZ-1IIIB cell cultures, evaluating their cytotoxicity in parallel. Infusions 2I and 4I, among the crude extracts, showed the highest activity. These extracts were refractioned into four fractions; 2I4 and 4I4 were active as inhibitors of DNA-polymerase (wild and Y181C types) and RNase H activities. These fractions were potent as inhibitors of viral replication and were not cytotoxic. Refractionation of 2I4 yielded five new fractions, two of which, 2I4-4 and 2I4-5, showed notable activity. Refractionation of 4I4 yielded for new fractions; of these, 4I4-3 and 4I4-4 were active. The marked biological activity found in the infusion of A. flaccida and P. sellowianus makes them sufficiently attractive to be considered in the combined chemotherapy of the disease
The Formation of Cosmic Structures in a Light Gravitino Dominated Universe
We analyse the formation of cosmic structures in models where the dark matter
is dominated by light gravitinos with mass of eV -- 1 keV, as predicted
by gauge-mediated supersymmetry (SUSY) breaking models. After evaluating the
number of degrees of freedom at the gravitinos decoupling (), we compute
the transfer function for matter fluctuations and show that gravitinos behave
like warm dark matter (WDM) with free-streaming scale comparable to the galaxy
mass scale. We consider different low-density variants of the WDM model, both
with and without cosmological constant, and compare the predictions on the
abundances of neutral hydrogen within high-redshift damped Ly-- systems
and on the number density of local galaxy clusters with the corresponding
observational constraints. We find that none of the models satisfies both
constraints at the same time, unless a rather small value (\mincir
0.4) and a rather large Hubble parameter (\magcir 0.9) is assumed.
Furthermore, in a model with warm + hot dark matter, with hot component
provided by massive neutrinos, the strong suppression of fluctuation on scales
of \sim 1\hm precludes the formation of high-redshift objects, when the
low-- cluster abundance is required. We conclude that all different variants
of a light gravitino DM dominated model show strong difficulties for what
concerns cosmic structure formation.
This gives a severe cosmological constraint on the gauge-mediated SUSY
