    Trafficking of cholinesterases and neuroligins mutant proteins. An association with autism

    Autism encompasses a wide spectrum of disorders arising during brain development. Recent studies reported that sequence polymorphisms in neuroligin-3 (NLGN3) and neuroligin-4 (NLGN4) genes have been linked to autism spectrum disorders indicating neuroligin genes as candidate targets in brain disorders. We have characterized a single mutation found in two affected brothers that substituted Arg451 to Cys in NL3. Our data show that the exposed Cys causes retention of the protein in the endoplasmic reticulum (ER) when expressed in HEK-293 cells. To examine whether the introduction of a Cys in the C-terminal region of other α/β-hydrolase fold proteins could promote the same cellular phenotype, we made homologous mutations in acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) and found a similar processing deficiency and intracellular retention (De Jaco et al., J Biol Chem. 2006, 281:9667-76). NL3, AChE and BChE mutant proteins are recognized as misfolded in the ER, and degraded via the proteasome pathway. A 2D electrophoresis coupled with mass spectrometry based approach was used to analyze proteins co-immunoprecipitating with NL3 and show differential expression of factors interacting with wild type and mutant NL3. We identified several proteins belonging to distinct ER resident chaperones families, including calnexin, responsible for playing a role in the folding steps of the AChE and NLs. © 2008 Elsevier Ireland Ltd.     Small coupling limit and multiple solutions to the Dirichlet Problem for Yang Mills connections in 4 dimensions - Part I

    In this paper (Part I) and its sequels (Part II and Part III), we analyze the structure of the space of solutions to the epsilon-Dirichlet problem for the Yang-Mills equations on the 4-dimensional disk, for small values of the coupling constant epsilon. These are in one-to-one correspondence with solutions to the Dirichlet problem for the Yang Mills equations, for small boundary data. We prove the existence of multiple solutions, and, in particular, non minimal ones, and establish a Morse Theory for this non-compact variational problem. In part I, we describe the problem, state the main theorems and do the first part of the proof. This consists in transforming the problem into a finite dimensional problem, by seeking solutions that are approximated by the connected sum of a minimal solution with an instanton, plus a correction term due to the boundary. An auxiliary equation is introduced that allows us to solve the problem orthogonally to the tangent space to the space of approximate solutions. In Part II, the finite dimensional problem is solved via the Ljusternik-Schirelman theory, and the existence proofs are completed. In Part III, we prove that the space of gauge equivalence classes of Sobolev connections with prescribed boundary value is a smooth manifold, as well as some technical lemmas used in Part I. The methods employed still work when the 4-dimensional disk is replaced by a more general compact manifold with boundary, and SU(2) is replaced by any compact Lie group

    Supramolecular aggregates containing lipophilic Gd(III) complexes as contrast agents in MRI

    Magnetic resonance imaging (MRI) contrast agents based on paramagnetic gadolinium complexes are widely used in biomedical research and diagnosis. Their application is intended to improve efficacy of MRI providing physiological information along with the impressive anatomical detail already obtained by images without contrast. The classical gadolinium complexes currently used for MRI contrast enhancement are all lowmolecularweightcompounds that rapidly equilibrate between the intra and extravascular spaces after intravenous administration. In order to obtain gadolinium-based agents with different pharmacokinetic properties, supramolecular aggregates such as micelles and liposomes have been recently proposed. Micelles and liposomes, obtained by the aggregation of lipophilic gadolinium complexes are here described, with the aim to correlate their structural and relaxometric properties.We report on the state of the art in the development of supramolecular aggregates obtained by self-assembly of lipophilic gadolinium complexes and aggregates in which lipophilic gadolinium complexes are assembled with surfactants. Moreover aggregates derivatized with bioactive molecules, such as peptides and antibodies, acting as target selective MRI contrast agents are described

    Short-communication: a comparison of the in vitro angiotensin-1-converting enzyme inhibitory capacity of dairy and plant protein supplements

    The consumption of supplements based on dairy or plant proteins may be associated with bioactive potential, including angiotensin-1-converting enzyme inhibitory (ACE-1i) activity, which is linked with blood pressure reduction in vivo. To gain insight into this proposed mechanism, the ACE-1i potential of protein-based supplements, including a selection of dairy (n = 10) and plant (n = 5) proteins were in vitro digested. The total digest was filtered and permeate and retentate were obtained. ACE-1i activity was measured as the ability of proteins (pre-digestion, 'gastric', permeate, and retentate) to decrease the hydrolysis of furanacroloyl-Phe-Glu-Glu (FAPGG) substrate for the ACE-1 enzyme. Permeate and retentate of dairy proteins exerted a significantly higher ACE-1i activity (mean of 10 proteins: 27.05 ± 0.2% and 20.7 ± 0.2%, respectively) compared with pre-digestion dairy proteins (16.7 ± 0.3%). Plant protein exhibited high ACE-1i in 'gastric' and retentate fractions (mean of five proteins: 54.9 ± 0.6% and 35.7 ± 0.6%, respectively). The comparison of the in vitro ACE-1i activity of dairy and plant proteins could provide valuable knowledge regarding their specific bioactivities, which could inform their use in the formulation of specific functional supplements that would require testing for blood pressure control in human randomly-controlled studies

    A Cross-Linguistic Study of the Relationship between Grammar & Lexical Development

    The relationship between grammatical and lexical development was compared in 233 English and 233 Italian children aged between 1;6 and 2;6, matched for age, gender, and vocabulary size on the MacArthur Communicative Development Inventories (CDI). Four different measures of Mean Length of Utterance were applied to the three longest utterances reported by parents, and to corrected/expanded versions representing the 'target' for each utterance. Italians had longer MLUs on most measures, but the ratio of actual to target MLUs did not differ between languages. Age and vocabulary both contributed significant variance to MLU, but the contribution of vocabulary was much larger, suggesting that vocabulary size may provide a better basis for crosslinguistic comparisons of grammatical development. The relationship between MLU and vocabulary size was non-linear in English but linear in Italian, suggesting that grammar 'gets off the ground' earlier in a richly inflected language. A possible mechanism to account for this difference is discussed

    Quaoar: New, Longitudinaly Resolved, Spectroscopic Characterization of Its Surface

    (50000) Quaoar, one of the largest Trans-neptunian objects, is comparable in size to Pluto's moon Charon. However, while Charon's surface is rich almost exclusively in H2O ice, Quaoar's surface characterized by ices of CH4, N2, as well as C2H6, a product of irradiation of CH4 (Dalle Ore et al. 2009). Because of its distance from the Sun, Quaoar is expected to have preserved, to a degree, its original composition, however, its relatively small size did not make it a prime candidate for presence of volatile ices in the study by Schaller and Brown (2007). Furthermore, based on the Brown et al. (2011) study (Brown, Schaller, & Fraser, 2011. A Hypothesis for the Color Diversity of the Kuiper Belt. ApJL, 739, L60) its red coloration points to CH3OH as the ice which, when irradiated, might have produced the red material. We present new visible to near-infrared (0.3-2.48 micrometers) spectro-photometric data obtained with the XSHOOTER (Vernet et al. 2011, A&A, 536A, 105 ) instrument at the VLT-ESO facility at four different longitudes on the surface of Quaoar. The data are complemented by previously published photometric observations obtained in the near-infrared (3.6, 4.5 micrometers) with the Spitzer Space Telescope, which provide an extra set of constraints in the model calculation process in spite of the different observing times that preclude establishing the spatial consistency between the two sets. For each of the four spectra we perform spectral modeling of the entire wavelength range -from 0.3 to 4.5 micrometers- by means of a code based on the Shkuratov radiative transfer formulation of the slab model. We obtain spatially resolved compositional information for the surface of Quaoar supporting the presence of CH4 and C2H6, as previously reported, along with evidence for N2 and NH3OH. The albedo at the two Spitzer bands indicates the likely presence of CO and CO2. CH3OH, predicted on the basis of Quaoar's coloration (Brown et al. 2011), is not found at any of the four longitudes, implying that the presence of this ice is a sufficient, but not necessary condition for reddening of TNO surfaces. Other ices, in particular CH4 (Brunetto et al. 2006), have been shown to be plausible precursors for reddening of TNO surfaces

    Modulation of tumor angiogenesis by conditional expression of fibroblast growth factor-2 affects early but not established tumors.

    Fibroblast growth factor-2 (FGF2) is a pleiotropic heparin-binding growth factor endowed with a potent angiogenic activity in vitro and in vivo. To investigate the impact of the modulation of FGF2 expression on the neovascularization at different stages of tumor growth, we generated stable transfectants (Tet-FGF2) from the human endometrial adenocarcinoma HEC-1-B cell line in which FGF2 expression is under the control of the tetracycline-responsive promoter (Tet-off system). After transfection, independent clones were obtained in which FGF2 mRNA and protein were up-regulated compared with parental cells. Also, the conditioned medium of Tet-FGF2 transfectants caused proliferation, urokinase-type plasminogen activator up-regulation, migration, and sprouting of cultured endothelial cells. A 3-day treatment of Tet-FGF2 cell cultures with tetracycline abolished FGF2 overexpression and the biological activity of the conditioned medium without affecting their proliferative capacity. Tet-FGF2 cells formed tumors when nude mice received s.c. injections. The administration of 2.0 mg/ml tetracycline in the drinking water before cell transplantation, continued throughout the whole experiment, inhibited FGF2 expression in Tet-FGF2 tumor lesions. This was paralleled by a significant decrease in the rate of tumor growth and vascularization to values similar to those observed in lesions generated by parental HEC-1-B cells. Tetracycline administration 20 days after tumor cell implant, although equally effective in reducing FGF2 expression and inhibiting tumor vascularity, only minimally impaired the growth of established Tet-FGF2 tumors. The results indicate that FGF2 expression deeply affects the initial tumor growth and neovascularization of HEC-1-B human endometrial adenocarcinoma in nude mice. On the contrary, the growth of established tumors appears to be independent of the inhibition of FGF2 expression and decreased vascular density. The possibility that a significant reduction of angiogenesis may not affect the progression of large tumors points to the use of antiangiogenic therapy in early tumor stage

    Pterodactyl: Trade Study for an Integrated Control System Design of a Mechanically Deployable Entry Vehicle

    This paper presents the trade study method used to evaluate and downselect from a set of guidance and control (G&C) system designs for a mechanically Deployable Entry Vehicle (DEV). The Pterodactyl project was prompted by the challenge to develop an effective G&C system for a vehicle without a backshell, which is the case for DEVs. For the DEV, the project assumed a specific aeroshell geometry pertaining to an Adaptable, Deployable Entry and Placement Technology (ADEPT) vehicle, which was successfully developed by NASAs Space Technology Mission Directorate (STMD) prior to this study. The Pterodactyl project designed three different entry G&C systems for precision targeting. This paper details the Figures of Merit (FOMs) and metrics used during the course of the projects G&C system assessment. The relative importance of the FOMs was determined from the Analytic Hierarchy Process (AHP), which was used to develop weights that were combined with quantitative design metrics and engineering judgement to rank the G&C systems against one another. This systematic method takes into consideration the projects input while simultaneously reducing unintentional judgement bias and ultimately was used to select a single G&C design for the project to pursue in the next design phase

    On Charge-3 Cyclic Monopoles

    We determine the spectral curve of charge 3 BPS su(2) monopoles with C_3 cyclic symmetry. The symmetry means that the genus 4 spectral curve covers a (Toda) spectral curve of genus 2. A well adapted homology basis is presented enabling the theta functions and monopole data of the genus 4 curve to be given in terms of genus 2 data. The Richelot correspondence, a generalization of the arithmetic mean, is used to solve for this genus 2 curve. Results of other approaches are compared.Comment: 34 pages, 16 figures.
