    Differences in peripartal plasma parameters related to calcium homeostasis of dairy sheep and goats in comparison with cows

    Recently it has been demonstrated that there are differences between sheep and goats in respect to adaptation to a calcium-restricted diet. It was the aim of the present study to evaluate whether species-specific peculiarities also occur when calcium homoeostasis is challenged by lactation. Therefore, we investigated the time courses of plasma parameters related to calcium homoeostasis (calcium, phosphate, calcitriol, the bone resorption marker CrossLaps® and the bone formation marker osteocalcin) during the transition period in multiparous animals of both species and compared the results to data from a former study carried out with dairy cows. As in cows, plasma calcium and the ratio of bone formation to bone resorption decreased at parturition in goats while plasma calcitriol increased. On day 10 post partum the bone parameters of goats reached prepartum values again, which was not the case in cows. Sheep were found to experience a challenge of calcium homoeostasis already 10d before parturition, reflected by a very low ratio of bone formation to bone resorption, which was not accompanied by an increase in plasma calcitriol. Additionally, sheep and goats which had been in milk for 3 months were sampled, dried-off and sampled again 6 weeks later. In dried-off animals there were no detectable differences in parameters of bone metabolism. In conclusion we could show that the contribution of bone mobilisation to the compensation for the enhanced calcium demand due to lactation differs between the three ruminant specie

    Gammaretrovirus-mediated correction of SCID-X1 is associated with skewed vector integration site distribution in vivo

    We treated 10 children with X-linked SCID (SCID-X1) using gammaretrovirus-mediated gene transfer. Those with sufficient follow-up were found to have recovered substantial immunity in the absence of any serious adverse events up to 5 years after treatment. To determine the influence of vector integration on lymphoid reconstitution, we compared retroviral integration sites (RISs) from peripheral blood CD3(+) T lymphocytes of 5 patients taken between 9 and 30 months after transplantation with transduced CD34(+) progenitor cells derived from 1 further patient and I healthy donor. Integration occurred preferentially in gene regions on either side of transcription start sites, was clustered, and correlated with the expression level in CD34(+) progenitors during transduction. In contrast to those in CD34(+) cells, RISs recovered from engrafted CD3(+)T cells were significantly overrepresented within or near genes encoding proteins with kinase or transferase activity or involved in phosphorus metabolism. Although gross patterns of gene expression were unchanged in transduced cells, the divergence of RIS target frequency between transduced progenitor cells and post-thymic T lymphocytes indicates that vector integration influences cell survival, engraftment, or proliferation

    A systematic assessment of water vapor products in the Arctic: from instantaneous measurements to monthly means

    Water vapor is an important component in the water and energy cycle of the Arctic. Especially in light of Arctic amplification, changes in water vapor are of high interest but are difficult to observe due to the data sparsity of the region. The ACLOUD/PASCAL campaigns performed in May/June 2017 in the Arctic North Atlantic sector offers the opportunity to investigate the quality of various satellite and reanalysis products. Compared to reference measurements at R/V Polarstern frozen into the ice (around 82∘ N, 10∘ E) and at Ny-Ålesund, the integrated water vapor (IWV) from Infrared Atmospheric Sounding Interferometer (IASI) L2PPFv6 shows the best performance among all satellite products. Using all radiosonde stations within the region indicates some differences that might relate to different radiosonde types used. Atmospheric river events can cause rapid IWV changes by more than a factor of 2 in the Arctic. Despite the relatively dense sampling by polar-orbiting satellites, daily means can deviate by up to 50 % due to strong spatio-temporal IWV variability. For monthly mean values, this weather-induced variability cancels out, but systematic differences dominate, which particularly appear over different surface types, e.g., ocean and sea ice. In the data-sparse central Arctic north of 84∘ N, strong differences of 30 % in IWV monthly means between satellite products occur in the month of June, which likely result from the difficulties in considering the complex and changing surface characteristics of the melting ice within the retrieval algorithms. There is hope that the detailed surface characterization performed as part of the recently finished Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) will foster the improvement of future retrieval algorithms

    Quality of Life in Male Breast Cancer:Prospective Study of the International Male Breast Cancer Program (EORTC10085/TBCRC029/BIG2-07/NABCG)

    INTRODUCTION: Prospective data about quality of life (QoL) in men with breast cancer (BC) are lacking. A prospective registry (EORTC10085) of men with all BC stages, including a QoL correlative study, was performed as part of the International Male Breast Cancer Program.METHODS: Questionnaires at BC diagnosis included the EORTC QLQ-C30 and BR23 (BC specific module), adapted for men. High functioning and global health/QoL scores indicate high functioning levels/high QoL; high symptom-focused measures scores indicate high symptoms/problems levels. EORTC reference data for healthy men and women with BC were used for comparisons.RESULTS: Of 422 men consenting to participate, 363 were evaluable. Median age was 67 years, and median time between diagnosis and survey was 1.1 months. A total of 114 men (45%) had node-positive early disease, and 28 (8%) had advanced disease. Baseline mean global health status score was 73 (SD: 21), better than in female BC reference data (62, SD: 25). Common symptoms in male BC were fatigue (22, SD: 24), insomnia (21, SD: 28), and pain (16, SD: 23), for which women's mean scores indicated more burdensome symptoms at 33 (SD: 26), 30 (SD: 32), and 29 (SD: 29). Men's mean sexual activity score was 31 (SD: 26), with less sexual activity in older patients or advanced disease.CONCLUSIONS: QoL and symptom burden in male BC patients appears no worse (and possibly better) than that in female patients. Future analyses on impact of treatment on symptoms and QoL over time, may support tailoring of male BC management.</p
