    The ERBB-STAT3 axis drives Tasmanian devil facial tumor disease

    The marsupial Tasmanian devil (Sarcophilus harrisii) faces extinction due to transmissible devil facial tumor disease (DFTD). To unveil the molecular underpinnings of this transmissible cancer, we combined pharmacological screens with an integrated systems-biology characterization. Sensitivity to inhibitors of ERBB tyrosine kinases correlated with their overexpression. Proteomic and DNA methylation analyses revealed tumor-specific signatures linked to the evolutionary conserved oncogenic STAT3. ERBB inhibition blocked phosphorylation of STAT3 and arrested cancer cells. Pharmacological blockade of ERBB or STAT3 prevented tumor growth in xenograft models and restored MHC class I expression. This link between the hyperactive ERBB-STAT3 axis and major histocompatibility complex class I-mediated tumor immunosurveillance provides mechanistic insights into horizontal transmissibility and puts forward a dual chemo-immunotherapeutic strategy to save Tasmanian devils from DFTD.     Sleep-related changes in autonomic control in obstructive sleep Apnea: a model-based perspective

    This paper reviews our current understanding of the long-term effects of obstructive sleep apnea (OSA) on cardiovascular autonomic function in humans, focusing directly on the knowledge derived from noninvasive measurements of heart rate, beat-to-beat blood pressure (BP), and respiration during wakefulness and sleep. While heart rate variability (HRV) as a means of autonomic assessment has become ubiquitous, there are serious limitations with the conventional time-domain and spectral methods of analysis. These shortcomings can be overcome with the application of a multivariate mathematical model that incorporates BP, respiration and other external factors as physiological sources of HRV. Using this approach, we have found that: (a) both respiratory-cardiac coupling and baroreflex dynamics are impaired in OSA; (b) continuous positive airway pressure therapy partially restores autonomic function; (c) baroreflex gain, which increases during sleep in normals, remains unchanged or decreases in OSA subjects; and (d) the autonomic changes that accompany transient arousal from NREM sleep in normals are largely absent in patients with OSA

    Chapter 2 makes the case for using systems thinking as a guiding perspective for TEEBAgriFood’s development of a comprehensive Evaluation Framework for the eco-agri-food system. Many dimensions of the eco-agri-food system create complex analytical and policy challenges. Systems thinking allows better understanding and forecasting the outcomes of policy decisions by illuminating how the components of a system are interconnected with one another and how the drivers of change are determined and impacted by feedback loops, delays and non-linear relationships. To establish the building blocks of a theory of change, systems thinking empowers us to move beyond technical analysis and decisiontool toward more integrated approaches that can aid in the forming of a common ground for cultural changes
