    Accommodating repair actions into gas turbine prognostics

    Elements of gas turbine degradation, such as compressor fouling, are recoverable through maintenance actions like compressor washing. These actions increase the usable engine life and optimise the performance of the gas turbine. However, these maintenance actions are performed by a separate organization to those undertaking fleet management operations, leading to significant uncertainty in the maintenance state of the asset. The uncertainty surrounding maintenance actions impacts prognostic efficacy. In this paper, we adopt Bayesian on-line change point detection to detect the compressor washing events. Then, the event detection information is used as an input to a prognostic algorithm, advising an update to the estimation of remaining useful life. To illustrate the capability of the approach, we demonstrated our on-line Bayesian change detection algorithms on synthetic and real aircraft engine service data, in order to identify the compressor washing events for a gas turbine and thus provide demonstrably improved prognosis

    Grazing winter wheat relieves plant water stress and transiently enhances photosynthesis

    In order to model the impact of grazing on wheat growth, we measured photosynthesis in the field. Grazing may affect photosynthesis as a consequence of changes to leaf water status, nitrogen content per unit leaf area (Na) or photosynthetic enzyme activity. While light-saturated CO2 assimilation rates (Asat) of field-grown wheat were unchanged during grazing, Asat transiently increased by 33-68% compared to ungrazed leaves over a two-four week period after grazing ended. Grazing reduced leaf mass per unit area, increased stomatal conductance and increased intercellular CO2 concentrations (Ci) by 36-38%, 88-169% and 17-20%, respectively. Grazing did not alter Na. Using a photosynthesis model, we demonstrated that the increase in Asat after grazing required an increase in Rubisco activity of up to 53%, whereas the increase in Ci could only increase Asat by up to 13%. Increased Rubisco activity was associated with a partial alleviation of leaf water stress. We observed a 68% increase in leaf water potential of grazed plants which could be attributed to reduced leaf area index and canopy evaporative demand, as well as increased rainfall infiltration into soil. The grazing of rain-fed grain cereals may be tailored to relieve plant water stress and enhance leaf photosynthesis

    Profile of XeominÂź (incobotulinumtoxinA) for the treatment of blepharospasm

    Even though conventional botulinum neurotoxin (BoNT) products have shown successful treatment results in patients with benign blepharospasm (BEB), the main, potential long-term side effect of BoNT use is the development of immunologic resistance due to the production of neutralizing antibody to the neurotoxin after repeated injections. XeominÂź (incobotulinumtoxinA), a unique botulinum neurotoxin type A (BoNT/A) drug free of complexing proteins otherwise contained in all conventional BoNT/A drugs, was recently approved by US Food and Drug Administration for the treatment of cervical dystonia or blepharospasm in adults. The newly approved BoNT/A drug may overcome this limitation of previous conventional products, since it contains pure neurotoxin (150 kDa) through a manufacturing process that separates it from complexing proteins such as hemagglutinins produced by fermentation of Clostridium botulinum. Many studies have also shown that XeominÂź has the same efficacy and safety profile as complexing protein-containing products such as BotoxÂź and is exchangeable with BotoxÂź using a simple 1:1 conversion ratio. XeominÂź represents a new treatment option for the repeated treatment of patients with blepharospasm in that it may reduce antibody-induced therapy failure. But, long-term comparative trials in naĂŻve patients between XeominÂź and conventional BoNT/A drugs are required to confirm the low immunogenicity of XeominÂź

    PinR mediates the generation of reversible population diversity in Streptococcus zooepidemicus

    Opportunistic pathogens must adapt to and survive in a wide range of complex ecosystems. Streptococcus zooepidemicus is an opportunistic pathogen of horses and many other animals, including humans. The assembly of different surface architecture phenotypes from one genotype is likely to be crucial to the successful exploitation of such an opportunistic lifestyle. Construction of a series of mutants revealed that a serine recombinase, PinR, inverts 114 bp of the promoter of SZO_08560, which is bordered by GTAGACTTTA and TAAAGTCTAC inverted repeats. Inversion acts as a switch, controlling the transcription of this sortase-processed protein, which may enhance the attachment of S. zooepidemicus to equine trachea. The genome of a recently sequenced strain of S. zooepidemicus, 2329 (Sz2329), was found to contain a disruptive internal inversion of 7 kb of the FimIV pilus locus, which is bordered by TAGAAA and TTTCTA inverted repeats. This strain lacks pinR and this inversion may have become irreversible following the loss of this recombinase. Active inversion of FimIV was detected in three strains of S. zooepidemicus, 1770 (Sz1770), B260863 (SzB260863) and H050840501 (SzH050840501), all of which encoded pinR. A deletion mutant of Sz1770 that lacked pinR was no longer capable of inverting its internal region of FimIV. The data highlight redundancy in the PinR sequence recognition motif around a short TAGA consensus and suggest that PinR can reversibly influence the wider surface architecture of S. zooepidemicus, providing this organism with a bet-hedging solution to survival in fluctuating environments

However, the significance of estimated evolutionary spectra has not been formally assessed. This paper describes a test for non-stationarity based on an estimate of the evolutionary spectrum and a time series bootstrap procedure. The test is applied to the seasonal time series of sea level pressure at Darwin. No significant non-stationarity is found

    “Model age-based” and “copy when uncertain” biases in children’s social learning of a novel task

    This work was supported by a John Templeton Foundation grant (40128).Theoretical models of social learning predict that individuals can benefit from using strategies that specify when and whom to copy. Here the interaction of two social learning strategies, model age-based biased copying and copy when uncertain, was investigated. Uncertainty was created via a systematic manipulation of demonstration efficacy (completeness) and efficiency (causal relevance of some actions). The participants, 4- to 6-year-old children (N = 140), viewed both an adult model and a child model, each of whom used a different tool on a novel task. They did so in a complete condition, a near-complete condition, a partial demonstration condition, or a no-demonstration condition. Half of the demonstrations in each condition incorporated causally irrelevant actions by the models. Social transmission was assessed by first responses but also through children’s continued fidelity, the hallmark of social traditions. Results revealed a bias to copy the child model both on first response and in continued interactions. Demonstration efficacy and efficiency did not affect choice of model at first response but did influence solution exploration across trials, with demonstrations containing causally irrelevant actions decreasing exploration of alternative methods. These results imply that uncertain environments can result in canalized social learning from specific classes of model.Publisher PDFPeer reviewe
