    An organization overview of pedagogical practice in work-integrated education

    Tertiary curriculum design has increasingly emphasized work-integrated learning (WIL) opportunities. This qualitative study provides an overview of a variety of WIL activities at Massey University, New Zealand. Descriptive comments, provided through interviews with fifteen academic supervisors from disciplines ranging from the applied sciences through social sciences to business, education and creative arts, highlight the following six factors to be considered in the resourcing of WIL programs. Themes related to set-up include placement requirements, support, selection, location, and risk management issues. Student preparation involves pre-requisite theoretical knowledge, general career preparation (CV & interview skills) and readiness for practice. With respect to supervision, an on-campus academic mentor and a work-place supervisor are both important to the student. Competencies linked to team work and professional standards include self-confidence, communication and people skills. The teaching pedagogies used include lectures and labs, oral presentations, scenario-based-learning and project work.     Quick assessment of the condition of gearboxes used in helicopters is a safety requirement. One of the most widely used helicopter on-board-mounted condition monitoring system these days is the Health and Usage Monitoring System. It has been specifically designed to monitor the condition of all safety-critical components operating in the helicopter through calculation of so-called condition indicators (CIs) - signal processing routines designed to output a single number that represents the condition of the monitored component. Among number of available parameters, there is a couple of CIs that over the years of testing have earned a reputation of being the most reliable measures of the gear tooth condition. At the same time, however, it has been observed that in some cases, those techniques do not properly indicate the deteriorating condition with the propagation of a gear tooth fault with the period of operation. Hence, three more robust methods have been suggested, which are discussed in this article

    Through hard work and thorough research, Richard Edwards, Jacob Friefeld, and Rebecca Wingo seek to dispel any false notions and set the record straight on what was one of the most influential events in the history of the United States in their book, Homesteading the Plains. Th roughout the work, the authors are able to correct false historical accounts that cast a poor light on the Homestead Act while they provide a wealth of statistical evidence to move toward a new history, as the book’s subtitle suggests. Th e Homestead Act provided opportunity for many ancestors of current Great Plains residents and helped populate the Great Plains along with making the region a major agricultural producer, cementing our roles in both national and international markets. As land was given out, communities were formed and churches and schools were soon to follow, helping to make the region and its people not only economically viable, but also culturally significant. While nothing is without its flaws, the Homestead Act was successful in its original goals, which is conclusively proven throughout the book

    Selection and Validation of Reference Genes for miRNA Expression Studies during Porcine Pregnancy

    MicroRNAs comprise a family of small non-coding RNAs that modulate several developmental and physiological processes including pregnancy. Their ubiquitous presence is confirmed in mammals, worms, flies and plants. Although rapid advances have been made in microRNA research, information on stable reference genes for validation of microRNA expression is still lacking. Real time PCR is a widely used tool to quantify gene transcripts. An appropriate reference gene must be chosen to minimize experimental error in this system. A small difference in miRNA levels between experimental samples can be biologically meaningful as these entities can affect multiple targets in a pathway. This study examined the suitability of six commercially available reference genes (RNU1A, RNU5A, RNU6B, SNORD25, SCARNA17, and SNORA73A) in maternal-fetal tissues from healthy and spontaneously arresting/dying conceptuses from sows were separately analyzed at gestation day 20. Comparisons were also made with non-pregnant endometrial tissues from sows. Spontaneous fetal loss is a prime concern to the commercial pork industry. Our laboratory has previously identified deficits in vasculature development at maternal-fetal interface as one of the major participating causes of fetal loss. Using this well-established model, we have extended our studies to identify suitable microRNA reference genes. A methodical approach to assessing suitability was adopted using standard curve and melting curve analysis, PCR product sequencing, real time PCR expression in a panel of gestational tissues, and geNorm and NormFinder analysis. Our quantitative real time PCR analysis confirmed expression of all 6 reference genes in maternal and fetal tissues. All genes were uniformly expressed in tissues from healthy and spontaneously arresting conceptus attachment sites. Comparisons between tissue types (maternal/fetal/non-pregnant) revealed significant differences for RNU5A, RNU6B, SCARNA17, and SNORA73A expression. Based on our methodical assessment of all 6 reference genes, results suggest that RNU1A is the most stable reference gene for porcine pregnancy studies

    Morphologic and functional correlates of synaptic pathology in the cathepsin D knockout mouse model of congenital neuronal ceroid lipofuscinosis

    Mutations in the cathepsin D (CTSD) gene cause an aggressive neurodegenerative disease (congenital neuronal ceroid lipofuscinosis) that leads to early death. Recent evidence suggests that presynaptic abnormalities play a major role in the pathogenesis of CTSD deficiencies. To identify the early events that lead to synaptic alterations, we investigated synaptic ultrastructure and function in pre-symptomatic CTSD knock-out (Ctsd(−/−)) mice. Electron microscopy revealed that there were significantly greater numbers of readily releasable synaptic vesicles present in Ctsd(−/−) mice than in wild-type control mice as early as postnatal day 16. The size of this synaptic vesicle pool continued to increase with disease progression in the hippocampus and thalamus of the Ctsd(−/−) mice. Electrophysiology revealed a markedly decreased frequency of miniature excitatory postsynaptic currents (EPSCs) with no effect on pair-pulse modulation of the evoked EPSPs in the hippocampus of Ctsd(−/−) mice. The reduced miniature EPSC frequency was observed before the appearance of epilepsy or any morphological sign of synaptic degeneration. Taken together, the data indicate that CTSD is required for normal synaptic function, and that a failure in synaptic trafficking or recycling may be an early and important pathological mechanism in Ctsd(−/−) mice; these presynaptic abnormalities may initiate synaptic degeneration in advance of subsequent neuronal loss

    Determination of the absorption length of CO2, Nd:YAG and high power diode laser radiation for a selected grouting material

    The laser beam absorption lengths of CO2, Nd:YAG and a high power diode laser (HPDL) radiation for a newly developed SiO2/Al2O3-based tile grout have been determined through the application of Beer-Lambert’s law. The findings revealed marked differences in the absorption lengths despite the material having similar beam absorption coefficients for the lasers. The absorption lengths for the SiO2/Al2O3-based tile grout for CO2, Nd:YAG and HPDL radiation were calculated as being 23211 m, 1934 m and 1838 m respectively. Moreover, this method of laser beam absorption length determination, which has hitherto been used predominantly with lasers operated in the pulsed mode, is shown to be valid for use with lasers operated in the continuous wave (CW) mode, depending upon the material being treated

    Exploratory Pollen Analysis of the Ditch of the 1665 Turf Fort, Jamestown, Virginia

    Pollen analysis of subsoil, slopewash, episodic fill, plowzone, and archaeological backdirt deposits in a core from a ditch associated with the 1665 Turf (earthwork) Fort at Jamestown, Virginia, record bare, slightly weedy local conditions around 17th-century artisan dwellings on the Jamestown waterfront and register the Virginia forest in the background before construction of the fort. Goosefoot dominated the earthwork slope; close relatives of the goldenrods were initially the most prominent plants in the open-ditch period. Pollen percolation rates adjusted for plowing and applied to ragweed-type (Ambrosia-type) percentages suggest that cultivation over the ditch began ca. 1729. Cultural matrix depostition, slopewash, and pollen percolation were crticial to the preservation of this record, and serve to emphasize the importance of evaluating pollen record formation processes in cultural landscape studies

    The structure of the title compound, tripotassium trans-tetra­cyanidodioxidotechnetate(V), is isotypic with its Re analogue. The [TcO2(CN)4]3− trans-tetra­cyanido­dioxido­technetate anion has a slightly distorted octa­hedral configuration. The Tc atom is located on a center of inversion and is bound to two O atoms in axial and to four cyanide ligands in equatorial positions. The Tc—O distance is consistent with a double-bond character. The two potassium cations, one located on a center of inversion and one in a general position, reside in octa­hedral or tetra­hedral environments, respectively. K⋯O and K⋯N inter­actions occur in the 2.7877 (19)–2.8598 (15) Å range
