    Recruitment of trimeric eIF2 by phosphatase non-catalytic subunit PPP1R15B

    Regulated protein phosphorylation controls most cellular processes. The protein phosphatase PP1 is the catalytic subunit of many holoenzymes that dephosphorylate serine/threonine residues. How these enzymes recruit their substrates is largely unknown. Here, we integrated diverse approaches to elucidate how the PP1 non-catalytic subunit PPP1R15B (R15B) captures its full trimeric eIF2 substrate. We found that the substrate-recruitment module of R15B is largely disordered with three short helical elements, H1, H2, and H3. H1 and H2 form a clamp that grasps the substrate in a region remote from the phosphorylated residue. A homozygous N423D variant, adjacent to H1, reducing substrate binding and dephosphorylation was discovered in a rare syndrome with microcephaly, developmental delay, and intellectual disability. These findings explain how R15B captures its 125 kDa substrate by binding the far end of the complex relative to the phosphosite to present it for dephosphorylation by PP1, a paradigm of broad relevance.</p

    Gold catalysts supported on ceria doped by rare earth metals for water gas shift reaction: Influence of the preparation method

    Gold catalysts based on ceria, doped by various RE metals (La, Sm, Gd, Yb, Y) were studied. The influence of the preparation methods on structure, properties and catalytic activity in the WGS reaction was investigated. The catalysts' supports were prepared using two different methods: co-precipitation (CP) and mechanochemical activation (MA). The catalysts were tested in a wide temperature interval without and after reactivation. All samples were characterized using a combination of X-ray diffraction (XRD), high resolution transmission electron microscopy (HRTEM), Raman spectroscopy (RS) and X-ray photoelectron spectroscopy (XPS) and TPR. It was found that the catalytic activity of MA catalysts is higher than CP ones. The gold catalysts based on ceria doped by Yb and Sm exhibited the highest activity. After reactivation in air the MA samples almost kept the WGS activity same, while the CP catalysts increased it. The catalysts of a single- and double-phase structure are formed as a result of CP and MA preparation, respectively. There are no big differences in the gold particles size (2-3 nm) depending on dopants and on the preparation techniques. The RS spectra analysis indicates that most probably the oxygen vacancies are adjacent to Me3+ dopant and the ceria structure seems to be better ordered than in the case of alumina as a dopant. There is no distinct correlation between reducibility and WGS activity. The XPS analysis disclose positively charged gold particles in addition to metallic gold within a surface region of fresh samples and only metallic gold on the samples after catalytic processing. There is no simple correlation between the concentration of Ce3+ in the samples and their WGS activity. © 2009 Elsevier B.V. All rights reserved

    Carbon balance assessment by eddy covariance method for agroecosystems with potato plants and oats & vetch mixture on sod-podzolic soils of Russia

    The carbon balance for the agroecosystems with potato plants and oats &amp; vetch mixture on sod-podzolics soils was evaluated using the eddy covariance approach. Absorption of carbon was recorded only during the growing season; maximum values were detected for all crops in July. The number of days during the vegetation period, when the carbon stocked in the fields with potatoes and oats &amp; vetch mixture was about the same and accounted for 53-55 days. During this period, the increase in gross primary production (GPP) is well correlated with the crop yields. The curve of the gross primary productivity is closely linked to the phases of development of plants; for potatoes, this graph differs significantly for all phases. Form of oats &amp; vetch mixture biomass curve shown linear increases. Carbon losses were observed for all the studied agroecosystems: for fields with an oats &amp; vetch mixture they were 254 g C m-2 y-1, while for fields with potato plants they were 307 g C m-2 y-1. Values about 250-300 g C m-2 per year may be considered as estimated values for the total carbon uptake for agroecosystems with potato plants and oats &amp; vetch mixture on sod-podzolic soils. © Published under licence by IOP Publishing Ltd

    Effect of combined oral contraceptives conaining drospirenone + ethinylestradiol on the mammary gland condition in women of reproductive age

    Drospirenone, as a part of combined oral contraceptives, has an antiproliferative effect on hyperplastic processes in the organs of the female reproductive system. Individual selection of COCs taking into account the dose and properties of the gestagenic and estrogenic components allows, with contraceptive and non-contraceptive purposes, to prescribe it to women of reproductive age. Intake of COC containing drospirenone + ethinyl estradiol is not associated with impaired drug tolerance due to complaints of soreness and breast engorgement in the first 3 months. 6 months of COCs containing drospirenone + ethinyl estradiol are significantly associated with a decrease in the frequency of clinical and US-signs of dyshormonal dysplasia of the breast. © 2018 Stavropol State Medical University.
