    Synaesthesia, hypnosis and consciousness

    For people with synaesthesia, a percept or concept (inducer) triggers another experience (concurrent) which is usually in a different modality. The concurrent is automatic, and in the case of certain types of synaesthesia also consistent, however the relationship between the inducer and concurrent is not fully understood and shall be investigated in this thesis from different perspectives. The first is using hypnosis to suggest synaesthesia-like phenomenological experiences to participants, and measuring behavioural responses to see whether they behave in a similar manner to developmental synaesthetes. Results from hypnotic; 1) grapheme-colour (GC) synaesthesia; 2) motion-sound synaesthesia; suggest that phenomenological experiences similar to developmental synaesthesia can be experienced by highly susceptible participants, but is not associated with the same behaviour as developmental synaesthetes. Developmental GC synaesthetes were tested to determine whether a grapheme presented preconsciously binds with the concurrent colour to the extent that it influences behaviour or evokes the phenomenology of colour. Two techniques were used, gaze-contingent substitution (GCS) and continuous flash suppression (CFS). Using GCS, it was shown that although digits can be primed preconsciously, they don’t bind with their concurrent colour to influence behaviour. Nevertheless, many synaesthetes still experienced colours though they didn’t necessarily match the primed digit. CFS experiments showed that the colour of a grapheme’s concurrent, or whether the grapheme is presented in the correct or incorrect colour for that synaesthete, doesn’t influence the time for conscious perception of a grapheme, even though colour words presented in the correct colour are perceived faster than those in the wrong colour. Phenomenological differences were compared to the behavioural measures using questionnaires modified using factor analysis (the R-RSPA and R-ISEQ). Overall, inducers must be seen consciously for them to bind with the concurrent, and experiencing the phenomenology of synaesthesia is not sufficient to behave like a synaesthete

    From dust to more dust: a paleoceanographic history of the East Asian Monsoon

    At present, the East Asian Monsoon (EAM) influences water availability for nearly one third of the global population. The intensity and position of the EAM has varied considerably since its onset, but disagreement still exists related to the precise latitudinal and intensity shifts of the Westerly Jet and associated storm fronts, which mark the northern extent of the monsoon. Paleoclimate research can assist in improved assessment and prediction of EAM intensity, radiative forcing, and biogeochemical cycles in the Japan Sea and North Pacific, especially under the currently changing climate. My research primarily focuses on using major-, trace- and rare earth elements in sediments from International Ocean Drilling Program Expedition 346 in the Japan (Ulleung Basin) and East China Seas (Okinawa Trough) to track variability in the EAM on millennial time scales. Using geochemical and multivariate statistical techniques (Q-Mode Factor analysis and Constrained Least Squares multiple linear regressions), I differentiated compositionally similar terrigenous aluminosilicate materials (continental crust components, eolian dusts, volcanic ash) from these sediment archives. I successfully constructed a robust record of aluminosilicate provenance, which enables more precise determinations of EAM position and intensity than previously possible. Most of my research focused on the interpretation of aluminosilicate records over several different timescales from three sites from Expedition 346. In tandem with this research, I also refined values of the well-known, and widely used, Standard Reference Material (SRM) Hawaiian Volcano Observatory Basalt (BHVO-2). In the Okinawa trough (Sites U1428/U1429), I identified and tracked the increase in flux of five continental crust materials, loesses, and volcanic ashes during glacial cycles, continental shelf exposure, and the migration of paleo-rivers in the last 400 kyr. Additionally, I constructed a 12 Myr record, which identified and quantified the dust fluxes to Ulleung Basin (Site U1430), and emphasized the importance of the Taklimakan and Gobi Deserts as main sources of dust to the Japan Sea and Pacific through the Cenozoic. Collectively, these aluminosilicate flux reconstructions are first to identify multiple specific Asian source regions through the Cenozoic, and highlight the complexity of accurately reconstructing monsoons and other aspects of paleoclimate from sediment in dynamic environments

    La menopausia se ha asociado a una elevada incidencia de arteriosclerosis y de enfermedad cardiovascular ya que debido al cambio hormonalfavorece laresistencia insulínica y un perfil lipídico más aterogénico, con aumento de triglicéridos, colesterol LDL pequeño y denso, incremento de ApoB y reducción de c-HDL.Existe abundante evidencia de que la DietaOccidental, que conforma el patrón dietético en Venezuela influye directamente en el perfil de riesgo cardiovascular.Por otra parte, La evidencia ha demostrado que 2 cucharaditas diarias (23g) de aceite de oliva virgen extra pueden reducir el riesgo coronario; mientras que con el aceite de canola se ha sugerido una y media cucharadas (19 g) pero las evidencias de este último son limitadas y no concluyentes, por lo que este trabajo aportaría información necesaria en la selección más acertada del tipo de aceite a utilizar en la dietoterapia de estas pacientes. Para este fin 18 mujeres postmenopáusicasconsumieron durante 28 días 3 tipos de dietas: 1.- Una dieta rica en grasas saturadas y su fórmula calórica total: 15% de proteínas, 47% de carbohidratos (CHO) y 38% como grasas (20% de AGS), 12% de AGM) y 6% de AGPI (omega 3), 2.- Una dieta rica en grasas monoinsaturadas rica en aceite de oliva virgen extra ( 15% de proteínas, 47% de CHO, 38% de grasas (<10% de AGS, 22% de AGM y 6% de AGPI(omega 3)). 3.-Una dieta rica en grasas monoinsaturadas a expensas de aceite de canola (15% de proteínas, 47% de CHO, 38% de grasas(< 10 % de AGS, 20% de AGM y 8 % de AGPI(omega 3) ) Al inicio y al final tras cada período se determinaron niveles plasmáticos de glucosa, insulina, HOMA-IR colesterol total, c-HDL, c-LDL, c-VLDL y TG, retinol, estradiol, FSH, testosterona, progesterona, cortisol, TSH, T3, T4 y DHEA. Al finalizar el período de intervención de la dieta SAT se observaron cambios significativospara las variables: glucemia, HOMA, Colesterol total, LDL-c, Colesterol No HDL y el estradiol. Tanto la dieta OLIVA como la dieta CANOLAindujeron un aumento en los niveles plasmáticos de c-HDL, sin embargo el aceite de oliva tuvo mayor efectividad en la mejoría de la LDL-c. Tras el consumo de la dieta rica en aceite de oliva se mostró una disminución del estradiol. Se concluye que la ingesta crónica de una comida grasa induce un estado hiperlipemia basal independientemente del tipo de grasa; además produce leves modificaciones en el perfil hormonal, pero no influyó en el perfil metabólico, ni en los niveles plasmáticos como antioxidante del retinol. La ingesta crónica del modelo de dieta OLIVA modificó el perfil lipídico comparado con la dieta SAT. La dieta CANOLA indujo un aumento de igual magnitud que la dieta OLIVA de c-HDL

    LIN28 is selectively expressed by primordial and pre-meiotic germ cells in the human fetal ovary

    Germ cell development requires timely transition from primordial germ cell (PGC) self-renewal to meiotic differentiation. This is associated with widespread changes in gene expression, including downregulation of stem cell–associated genes, such as OCT4 and KIT, and upregulation of markers of germ cell differentiation and meiosis, such as VASA, STRA8, and SYCP3. The stem cell–expressed RNA-binding protein Lin28 has recently been demonstrated to be essential for PGC specification in mice, and LIN28 is expressed in human germ cell tumors with phenotypic similarities to human fetal germ cells. We have therefore examined the expression of LIN28 during normal germ cell development in the human fetal ovary, from the PGC stage, through meiosis to the initiation of follicle formation. LIN28 transcript levels were highest when the gonad contained only PGCs, and decreased significantly with increasing gestation, coincident with the onset of germ cell differentiation. Immunohistochemistry revealed LIN28 protein expression to be germ cell–specific at all stages examined. All PGCs expressed LIN28, but at later gestations expression was restricted to a subpopulation of germ cells, which we demonstrate to be primordial and premeiotic germ cells based on immunofluorescent colocalization of LIN28 and OCT4, and absence of overlap with the meiosis marker SYCP3. We also demonstrate the expression of the LIN28 target precursor pri-microRNA transcripts pri-LET7a/f/d and pri-LET-7g in the human fetal ovary, and that expression of these is highest at the PGC stage, mirroring that of LIN28. The spatial and temporal restriction of LIN28 expression and coincident peaks of expression of LIN28 and target pri-microRNAs suggest important roles for this protein in the maintenance of the germline stem cell state and the regulation of microRNA activity in the developing human ovary

    The contribution of OCTN1/2 variants within the IBD5 locus to disease susceptibility and severity in Crohn's disease

    Background and Aims: Recent data suggest that polymorphisms in the organic cation transporter (OCTN) genes OCTN1 (SLC22A4) and OCTN2 (SLC22A5) represent disease-causing mutations within the IBD5 locus (chromosome 5q31). We investigated associations with disease susceptibility, phenotype, and evidence for epistasis with CARD15 in 679 patients with Crohn’s disease (CD) or ulcerative colitis (UC). Methods: A total of 374 patients with CD, 305 patients with UC, and 294 healthy controls (HCs) were studied. Genotyping for single nucleotide polymorphisms IGR2096, IGR2198, and IGR2230, OCTN1 variant (SLC22A4 1672C→T), and OCTN2 variant (SLC22A5 −207G→C) was performed using the TaqMan system. Results: The IBD5 OCTN1 and OCTN2 polymorphisms were in strong linkage disequilibrium (D′, >0.959). IGR2198 variant allele frequency (49.1% vs 40.8%; P = .0046) and homozygosity (21% vs 14.8%; P = .044) were associated with CD versus HCs. Variant allelic frequency of OCTN1 (53.6% vs 43%; P = .0008) and OCTN2 (56.1% vs 48.4%; P = .0092) polymorphisms and homozygosity for the OCTN1/2-TC haplotype (28.4% vs 16%; P = .0042) were associated with CD versus HCs. IGR2198 homozygosity and TC homozygosity were associated with stricturing/penetrating disease at follow-up (P = .011 and P = .011, respectively) and disease progression (P = .038 and P = .049, respectively) on univariate analysis and with need for surgery on multivariate analysis (P = .016 and P = .004, respectively). In the absence of the IBD5 risk haplotype, no association of OCTN1/2 variants with CD was detected. No associations were seen with UC. Conclusions: The IBD5 locus influences susceptibility, progression, and need for surgery in CD. However, the contribution of OCTN1/2 variants is not independent of the IBD5 haplotype; a causative role for these genes remains plausible but is not yet proven. Further genetic, functional, and expression data are now required. </p

    Oxidative stress in relation to diet and physical activity among premenopausal women

    Abstract Higher levels of oxidative stress, as measured by F 2 -isoprostanes, have been associated with chronic diseases such as CVD and some cancers. Improvements in diet and physical activity may help reduce oxidative stress; however, previous studies regarding associations between lifestyle factors and F 2 -isoprostane concentrations have been inconsistent. The aim of this cross-sectional study was to investigate whether physical activity and intakes of fruits/vegetables, antioxidant nutrients, dietary fat subgroups and alcohol are associated with concentrations of F 2 -isoprostane and the major F 2 -isoprostane metabolite. Urinary F 2 -isoprostane and its metabolite were measured in urine samples collected at enrolment from 912 premenopausal women (aged 35–54 years) participating in the Sister Study. Physical activity, alcohol consumption and dietary intakes were self-reported via questionnaires. With adjustment for potential confounders, the geometric means of F 2 -isoprostane and its metabolite were calculated according to quartiles of dietary intakes, alcohol consumption and physical activity, and linear regression models were used to evaluate trends. Significant inverse associations were found between F 2 -isoprostane and/or its metabolite and physical activity, vegetables, fruits, vitamin C, α -carotene, vitamin E, β -carotene, vitamin A, Se, lutein+zeaxanthin and long-chain n -3 fatty acids. Although trans fats were positively associated with both F 2 -isoprostane and its metabolite, other dietary fat subgroups including SFA, n -6 fatty acids, n -3 fatty acids, MUFA, PUFA, short-chain n -3 fatty acids, long-chain n -3 fatty acids and total fat were not associated with either F 2 -isoprostane or its metabolite. Our findings suggest that lower intake of antioxidant nutrients and higher intake of trans fats may be associated with greater oxidative stress among premenopausal women

    Principle component analyses of questionnaires measuring individual differences in synaesthetic phenomenology

    a b s t r a c t Questionnaires have been developed for categorising grapheme-colour synaesthetes into two sub-types based on phenomenology: associators and projectors. The general approach has been to assume a priori the existence of two sub-types on a single dimension (with endpoints as projector and associator) rather than explore, in a data-driven fashion, other possible models. We collected responses from 175 grapheme-colour synaesthetes on two questionnaires, the Illustrated Synaesthetic Experience Questionnaire (Skelton, Ludwig, &amp; Mohr, 2009) and Rouw and Scholte&apos;s (2007) Projector-Associator Questionnaire. After Principle Component Analysis both questionnaires were comprised of two factors which coincide with the projector/associator distinction. This suggests that projectors and associators are not opposites of each other, but separate dimensions of experience (e.g. some synaesthetes claim to be both, others claim to be neither). The revised questionnaires provide a useful tool for researchers and insights into the phenomenology of synaesthesia
