Are Basophils Important Mediators for Helminth-Induced Th2 Immune Responses? A Debate

    Get PDF
Helminth parasites induce Th2 immune responses. Immunological mechanisms leading to Th2 induction are mainly dependent on IL-4. However, early source of IL-4 has not been precisely identified. Noticeably, basophils seem to be important mediators for inducing and maintaining the Th2 response probably because they secrete IL-4 and exert functions similar to APCs. Nevertheless, recent experimental evidence points that DCs could be also significant participants during this process. The involvement of basophils during memory responses is also discussed

Objective: the relationship between depression and adherence to pharmacological treatment of people with type 2 diabetes mellitus of Hospital II Jorge Reátegui Delgado-Piura, 2019. Methodology: the research is basic, observational, cross-sectional analytical type with primary data collection. A sample of 228 patients diagnosed with DM2 was taken, which, after meeting the inclusion and exclusion selection criteria, could be properly classified. Information was collected through the application of a research instrument (data sheet) and Beck's depression inventory. Results: of the 228 patients, 84.6% are depressed and only 15.4% do not have depression. The level of depression most frequently found in patients is severe depression with 39.2% of all patients, followed by patients with moderate depression, who represent 19.4% of patients, and continuing with patients with extreme depression with 13.2% and finally we have 12.8% of the total of patients with Mild Depression. Regarding the control of Basal Glycemia, the multivariate analysis showed that patients who have poor control of it, have a higher risk of having depression than patients with good control of Basal Glycemia, the same happens with respect to control. of glycated hemoglobin, patients with poor control of it, are more likely to have a type of depression. Taking into account the occupational status, the analysis showed that patients who are not working are the most likely to have depression, as is the case with male patients, patients who have problems with their body mass (weight), patients who are without a romantic partner and people who are under 50 years of age. Conclusions: A relationship was found between depression and adherence to drug treatment of patients with type 2 diabetes mellitus of Hospital Jorge Reátegui Delgado II.Tesi

Engineering of specific bacteriophages for early diagnosis of Alzheimer′s disease

Alzheimer’s disease (AD) is the most common neurodegenerative disease affecting a large proportion of the human population worldwide with great impact on social and economic level. At molecular level, AD is characterized by an increased deposition of plaques, which consist of amyloid-beta however, it is not the amyloid-beta in plaques, but amyloid-beta in soluble oligomeric form that impairs synaptic function and memory encoding. The limitations imposed by the blood-brain barrier (BBB) have hindered the development of new diagnostic/therapeutic techniques. Also, AD-treatments that target plaques have proven to be ineffective, therefore it is important to find diagnostic and therapeutic tools that selectively target amyloid-beta in oligomeric form. Peptie ligands that selectively recognize AB-oligomers are available, however they are not able to cross the BBB. To overcome this limitation, the development and application of viruses has become a very interesting tool. Bacteriophages (or phages – virus that only infect bacterial cells) can bypass the BBB and can be genetically and chemically manipulated in order to recognize and target specific biomarkers commonly used for AD diagnostic. The present work describes the development of a bacteriophage-based system that can be capable of diagnose AD at an early stage by shuttling amyloid-beta specific ligands across the BBB. Phages were genetically engineered with two peptide sequences described to selectively recognize amyloid-beta oligomers in order to target and visualize amyloid-beta aggregates in the brain. Future work will be devoted to test this system in AD-mouse models for diagnosis purposes at an early stage of the disease. If successful, this approach will provide the neuroscience community with a promising tool for AD early diagnose

A Reference Meta-model to Understand DNA Variant Interpretation Guidelines

    Get PDF
Determining the role of a DNA variant in patients’ health status – a process known as variant interpretation – is highly critical for precision medicine applications. Variant interpretation involves a complex process where, regrettably, there is still debate on how to combine and weigh diverse available evidence to achieve proper and consistent answers. Indeed, at the time of writing, 22 different variant interpretation guidelines are available to the scientific community, each of them attempting to establish a framework for standardizing the interpretation process. However, these guidelines are qualitative and vague by nature, which hinders their streamlined application and potential automation. Consequently, more precise definitions are needed. Conceptual modeling provides the means to bring clarification within this domain. This paper presents our efforts to define and use a UML meta-model that describes the main concepts involved in the definition of variant interpretation guidelines and the constructs they evaluate. The precise conceptual definition of the guidelines allowed us to identify four common misinterpretation patterns that hamper the correct interpretation process and that can consequently affect classification results. In several proposed examples, the use of the meta-model provides support in identifying the inconsistencies in the observed process; this result paves the way for further proposing reconciliation strategies for the existing guidelines
