    Antitumour Activity of Dehydroxymethylepoxyquinomycin (DHMEQ): a Literature Review

    Carcinogenesis research uncovers new pathogenesis links as vulnerable targets of effective antitumour therapies. Among the key mediators of immune response, cell proliferation, cell apoptosis and inflammation is transcription factor NF-ΞΊB. Misregulation of an NF-ΞΊB-dependent pathway is found in solid and haematopoietic tumour cells. One of the best known NF-ΞΊB functions is expression regulation of genes involved in the apoptosis inhibition or activation and survival in both intact and malignant cells. The NF-ΞΊB-mediated pathways’ involvement in carcinogenesis, angiogenesis and tumour resistance to chemo- and radiotherapies makes this factor a promising target for drug anti-cancer interventions. This review summarises evidence on the antitumour and anti-inflammatory activity of a high-potent and specific low molecular-weight NF-ΞΊB inhibitor, dehydroxymethylhepoxyquinomycin (DHMEQ), as a candidate therapeutic agent in treatment for variant malignancies

    Investigation of Raman Spectra and Ionic Conductivity of Composites Based on NaClO4 and KClO4 Salts Obtained by Mechanoactivation

Published online: 18 March 2024.The work is aimed at studying the effect of mechanical activation on the structure and electrical conductivity of the NaClO4 – and KClO4 – based composites. Based on the result of the analysis of the DSC curves of the (1–x) NaClO4 – x Al2O3 and (1–x) KClO4 – x Al2O3 composites measured while heating and cooling the samples, it was established that the enthalpy of phase transitions in them decreased with an increase in the concentration of the nanosized dopant. A complication of all active vibrational contours corresponding to internal vibrations of the molecular anion in the composites with increasing Al2O3 concentration and a shift of the band of the fully symmetric stretching vibration v1 (A) to a low-frequency region was revealed by Raman spectroscopy. Based on electrochemical impedance spectroscopy data, it was determined that for the 0.4NaClO4 – 0.6Al2O3 system subjected to mechanoactivation, the values of specific ionic conductivity increased by two orders of magnitude as compared to that of pure NaClO4, while for the 0.4KClO4 – 0.6Al2O3 system, the values of specific ionic conductivity increased by three orders of magnitude as compared to the initial salt at T = 320 Β°C.This study was carried out using the instruments of the Analytical Center of Collective Use of the Dagestan Federal Research Center of the Russian Academy of Sciences

    Introduction. Ovarian cancer (OC) is recognized to be a pressing problem of modern oncology. Cytoreductive surgery and combined therapy based on platinum and taxanes play an important role in OC treatment. The response rate to first-line therapy accounts for about 80–90%. However, most patients relapse and develop resistance to therapy. Thus, the search for new effective drugs and new combinations for OC treatment is an urgent task of modern oncology.Aim. To evaluate in vivo the antitumor activity of dehydroxymethylepoxyquinomycin (DHMEQ) and cisplatin combination in an ovarian cancer model.Materials and methods. An experimental model of disseminated OC in rats was used to evaluate antitumor activity. A strain of ovarian tumor (OT) was transplanted into 200 female Wistar rats. The drugs were administered intraperitoneally. The β€œmedian life expectancy” was taken as a benchmark for the quality evaluation of experimental treatment.Results. It was found that DHMEQ and cisplatin combination increased the survival rate by 387% (p = 0.005, log-rank test) compared to the control group and by 91% compared to the group of animals treated with cisplatin (p = 0.003, log-rank test) in mono mode. More than 50% of the animals in the DHMEQ + cisplatin group remained alive on day 73 of the experiment. No animals remained alive in the cisplatin group, and only one rat remained in the DHMEQ group.Discussion. Thus, the obtained data demonstrate a potentiating antitumor effect of the DHMEQ + cisplatin combination by 387% compared to the control group.Conclusion. The results of the experiments demonstrated a potentiating antitumor effect of DHMEQ in combination with cisplatin. DHMEQ in combination with cisplatin manifests high efficacy in an in vivo model of ovarian cancer.Π’Π²Π΅Π΄Π΅Π½ΠΈΠ΅.     The transcription factor NF-kB plays a key role in various physiological processes such as immune response, cell proliferation, cell apoptosis and inflammation. Controlling the transcriptional activity of a number of genes responsible for different ways of inflammatory processes and of malignant, NF-kB is involved in a wide range of diseases. Special attention deserves part signalnych paths controlled NF-kB, in carcinogenesis, angiogenesis, including tumor and stability to chemotherapy and radiotherapy.     Morphological changes of the gastric mucosa in patients with duodenogasric reflux: relationship with acidity and helicobacter pylori

    Purpose: to reveal the peculiarities of the duodenogastric reflux (DGR) impact on acidity and morphological features of the gastric mucosa in patients with "primary" reflux gastritis, as well as in cases of reflux gastritis in combination with H. pylori. Methods: 66 patients with chronic reflux gastritis were examined, 15 people formed a control group. All patients had gastric endoscopy with determination of the gastric acidity and concentration of bile acids. Biopsy material for histological examination was obtained from 55 patients, in 12 cases of suspected intestinal metaplasia additional histochemical (PAS and alcian blue) and immunohistochemical (cytokeratin 20, Villin, carcinoembriogenic antigen, MUC2, CD45) assays were performed. Results: the level of acidity in patients with DGR was higher (pH = 3.55 Β± 2.3) compared to the control group (pH = 6.85 Β± 1.34). Cases of DGR differed from the control group regarding the mononuclear infiltration (p=0,001), foveolar hyperplasia (p=0,001) in both antral and fundic parts of the stomach, mucosal edema (p=0,022) and atrophy (p=0,02) at the level of the antrum, and intestinal metaplasia (p=0,022) at the level of the body. In the presence of H. pylori infection, in addition to the abovementioned signs, infiltration by cells of acute inflammation (p = 0.005) was detected, which indicates an increase in the damaging effect of H. pylori infection in DGR. Conclusion: DGR may lead to "oxidation" of gastric contents. The effect of DGR leads to intestinal metaplasia of the body and atrophy of the antral part of stomach, chronic inflammation and foveolar hyperplasia.
