Peritumoural, but not intratumoural, lymphatic vessel density and invasion correlate with colorectal carcinoma poor-outcome markers
To evaluate whether lymphatic vessel density (LVD) and lymphatic vessel invasion (LVI) are useful markers of worse outcome in colorectal carcinoma and if LVD and LVI correlate to the classical clinical-pathological parameters, we analysed 120 cases of colorectal carcinomas selected from the files of Division of Pathology, Hospital das Clinicas, São Paulo University, Brazil. Assessment of LVD and LVI was performed by immunohistochemical detection of lymphatic vessels, using the monoclonal antibody D2-40. Higher LVD was found in the intratumoural area, when comparing with normal and peritumoural areas (p < 0.001). However, peritumoural LVD, but not intratumoural, correlated with both colonic-wall-invasion depth (p = 0.037) and liver metastasis (p = 0.012). Remarkably, LVI was found associated with local invasion (p = 0.016), nodal metastasis (p = 0.022) and hepatic metastasis (p < 0.001). Peritumoural LVD and LVI are directly related to histopathological variables indicative of poor outcome such as lymph-node status and liver metastasis
249 TP53 mutation has high prevalence and is correlated with larger and poorly differentiated HCC in Brazilian patients
<p>Abstract</p> <p>Background</p> <p>Ser-249 TP53 mutation (249<sup>Ser</sup>) is a molecular evidence for aflatoxin-related carcinogenesis in Hepatocellular Carcinoma (HCC) and it is frequent in some African and Asian regions, but it is unusual in Western countries. HBV has been claimed to add a synergic effect on genesis of this particular mutation with aflatoxin. The aim of this study was to investigate the frequency of 249<sup>Ser </sup>mutation in HCC from patients in Brazil.</p> <p>Methods</p> <p>We studied 74 HCC formalin fixed paraffin blocks samples of patients whom underwent surgical resection in Brazil. 249<sup>Ser </sup>mutation was analyzed by RFLP and DNA sequencing. HBV DNA presence was determined by Real-Time PCR.</p> <p>Results</p> <p>249<sup>Ser </sup>mutation was found in 21/74 (28%) samples while HBV DNA was detected in 13/74 (16%). 249<sup>Ser </sup>mutation was detected in 21/74 samples by RFLP assay, of which 14 were confirmed by 249<sup>Ser </sup>mutant-specific PCR, and 12 by nucleic acid sequencing. All HCC cases with p53-249ser mutation displayed also wild-type p53 sequences. Poorly differentiated HCC was more likely to have 249<sup>Ser </sup>mutation (OR = 2.415, 95% CI = 1.001 – 5.824, p = 0.05). The mean size of 249<sup>Ser </sup>HCC tumor was 9.4 cm versus 5.5 cm on wild type HCC (p = 0.012). HBV DNA detection was not related to 249<sup>Ser </sup>mutation.</p> <p>Conclusion</p> <p>Our results indicate that 249<sup>Ser </sup>mutation is a HCC important factor of carcinogenesis in Brazil and it is associated to large and poorly differentiated tumors.</p
Genome of Rhodnius prolixus, an insect vector of Chagas disease, reveals unique adaptations to hematophagy and parasite infection
Rhodnius prolixus not only has served as a model organism for the study of insect physiology, but also is a major vector of Chagas disease, an illness that affects approximately seven million people worldwide. We sequenced the genome of R. prolixus, generated assembled sequences covering 95% of the genome ( approximately 702 Mb), including 15,456 putative protein-coding genes, and completed comprehensive genomic analyses of this obligate blood-feeding insect. Although immune-deficiency (IMD)-mediated immune responses were observed, R. prolixus putatively lacks key components of the IMD pathway, suggesting a reorganization of the canonical immune signaling network. Although both Toll and IMD effectors controlled intestinal microbiota, neither affected Trypanosoma cruzi, the causal agent of Chagas disease, implying the existence of evasion or tolerance mechanisms. R. prolixus has experienced an extensive loss of selenoprotein genes, with its repertoire reduced to only two proteins, one of which is a selenocysteine-based glutathione peroxidase, the first found in insects. The genome contained actively transcribed, horizontally transferred genes from Wolbachia sp., which showed evidence of codon use evolution toward the insect use pattern. Comparative protein analyses revealed many lineage-specific expansions and putative gene absences in R. prolixus, including tandem expansions of genes related to chemoreception, feeding, and digestion that possibly contributed to the evolution of a blood-feeding lifestyle. The genome assembly and these associated analyses provide critical information on the physiology and evolution of this important vector species and should be instrumental for the development of innovative disease control methods
Cirrose hepática: aspectos morfológicos relacionados à s suas possÃveis complicações. Um estudo centrado em necropsias
INTRODUÇÃO: A cirrose hepática é muito prevalente em nosso meio, cursando freqüentemente com complicações como insuficiência e carcinoma hepatocelulares e hipertensão portal. OBJETIVOS: Analisar os aspectos clÃnicos e morfológicos das cirroses, relacionando-os com os principais padrões histológicos e suas complicações. MATERIAL E MÉTODO: Entre 35 fÃgados cirróticos provenientes de necropsias realizadas no Serviço de Verificação de Óbitos da Capital (SVOC-SP), estudados em cortes seqüenciais a cada centÃmetro, dois padrões anatomopatológicos predominaram: os associados a hepatites crônicas e os decorrentes de esteato-hepatites. RESULTADOS E DISCUSSÃO: A alta incidência de cirrose em necropsias (7,1%), com freqüente detecção de macronódulos (62,8%) e de carcinoma hepatocelular (CHC) (37,1%), aponta para a relevância dessas questões em saúde pública. A análise histológica demonstrou o encontro de neoductos, agressão biliar e siderose com maior freqüência na hepatite crônica que na esteato-hepatite. CONCLUSÕES: Essa abordagem sistemática nos espécimes necroscópicos permitiu-nos a identificação de elevada freqüência de cirrose em necropsias do SVOC e, em especial, daquelas decorrentes de hepatites crônicas, aspecto de interesse sanitário. A freqüente detecção de CHC, além de constituir modelo para futuros estudos sobre o surgimento dessas neoplasias, demonstra ser uma das principais complicações da cirrose avançada, qualquer que seja sua causa
Thermo-analyses of polyaniline and its derivatives
In this work, is presented the thermal behavior of polyaniline (PAN!) and its derivatives poly(o-ethoxyaniline) (POEA) and poly(o-methoxyaniline) (PUMA), which were studied by using differential scanning calorimetry (DSC), modulated DSC (TMDSC), respectively, and thermal gravimetric analysis (TGA). The results from diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) and thermal analysis showed the formation of crosslinking isomerization reaction during the heating process. The results showed that the maximum weight loss and the crystallinity degree depend on the type of the aromatic ring substituent group, i.e. hydrogen, ethoxy or methoxy. (C) 2010 Elsevier B.V. High-fat diet: A trigger of non-alcoholic steatohepatitis? Preliminary findings in obese subjects
Objective: We correlated dietary profile and markers of visceral and somatic obesities in nonalcoholic fatty liver disease. Methods: Patients with histologically proven fatty infiltration of the liver (n = 25, 52 +/- 11 y of age, 64% women) underwent abdominal computed tomography, bioelectrical impedance, and anthropometric measurements. Insulin resistance was evaluated (homeostasis model assessment) and dietary intake of macronutrients was estimated by 24-h recall. Main outcome measurements were correlation of carbohydrate and fat ingestion with liver histology. Results: Metabolic syndrome was present in 72% of the population, and increased waist circumference and low high-density lipoprotein cholesterol occurred in 66%. Total body fat (bioimpedance) and dietary intake of lipids were higher in patients with non-alcoholic steatohepatitis (P < 0.05), but not in diabetic subjects who exhibited more steatosis than non-alcoholic steatohepatitis. Waist circumference exhibited a good correlation with homeostasis model assessment, total energy intake, and ingestion of specific fatty acids. Body mass index correlated well with somatic and visceral adiposities. Conclusion: Energy intake and visceral adiposity were predisposing factors for fatty liver disease. Lipid input correlated with non-alcoholic steatohepatitis in the entire group and after stratification for diabetes. These findings suggest that lipid intake may play a greater role in non-alcoholic steatohepatitis than hitherto suspected. (C) 2008 Elsevier Inc. All rights reserved
Post-transplant recurrent hepatitis C: immunohistochemical detection of hepatitis C virus core antigen and possible pathogenic implications
Introduction: The mechanisms by which severe cholestatic hepatitis develops after liver transplantation are not fully understood. Reports on immunohistochemical distribution of hepatitis C virus (HCV) antigens are still scarce, but recently, HCV immunostaining was suggested for early diagnosis of cholestatic forms of recurrent hepatitis C in liver grafts. After purification, Rb246 pab anticore (aa1-68) yielded specific, granular cytoplasmic staining in hepatocytes. Signal amplification through the Envision-Alkaline Phosphatase System avoided endogenous biotin and peroxidase. Aims/Methods: Rb246 was applied to liver samples of explants of 12 transplant recipients, six with the most severe form of post-transplantation recurrence, severe cholestatic hepatitis (group 1) and six with mild recurrence (group 2). We also assessed immuno-reactivity at two time-points post-transplantation (median 4 and 22 months) in both groups. HCV-core Ag was semiquantified from 0 to 3+ in each time point. Serum HCV-RNA was also measured on the different time points by branched DNA. Results: In the early post-transplant time point, one patient had a mild staining (1+), two patients had a moderate staining (2+) and the other three had no staining in group 1, compared with five patients with no staining (0) and one patient with mild staining (1+) in group 2. Late post-transplant liver samples were available in nine patients, and two out of four samples in group 1 showed a mild staining, compared with no staining patients in five patients in group 2. Strikingly, on the explant samples, HCV immunostaining was strongly positive in group 1, and mildly positive in group 2. Two out of five samples showed 3+ staining, and three samples showed 2+ staining in group 1; two out of five samples showed no staining, two samples showed 1+ staining and one sample showed 2+ staining in group 2. Serum HCV-RNA was significantly higher in group 1, on both time-points post-transplantation. HCV-core Ag was not directly associated with serum HCV-RNA on the different time points. Conclusion: These preliminary results suggest that strong HCV immunostaining in the explant is predictive of more severe disease recurrence
Acromegaly: correlation between expression of somatostatin receptor subtypes and response to octreotide-lar treatment
About one-third of acromegalics are resistant to the clinically available somatostatin analogs (SA). The resistance is related to density reduction or different expression of somatostatin receptor subtypes (SSTR). This study analyzes SSTR`s expression in somatotrophinomas, comparing to SA response, hormonal levels, and tumor volume. We analyzed 39 somatotrophinomas; 49% were treated with SA. The most expressed SSTR was SSTR5, SSTR3, SSTR2, SSTR1, and SSTR4, respectively. SSTR1 and SSTR2 had higher expression in patients that had normalized GH and IGF-I. SSTR3 was more expressed in patients with tumor reduction. There was a positive correlation between the percentage of tumor reduction and SSTR1, SSTR2 and SSTR3 expression. Also, a positive correlation between SSTR2 mRNA expression and the immunohistochemical reactivity of SSTR2 was found. Our study confirmed the association between the SA response to GH and IGF-I and the SSTR2. Additionally, this finding was also demonstrated in relation to SSTR1
