    I am Bedu: the Changing Bedouin in a Changing World

    Change occurs throughout the world on a daily basis, and as a result, its inhabitants are encouraged to change as well. The population learns to adapt in various ways in order to survive, often times causing traditional practices to be forced aside in order to make way for more modern methods. Today, the Bedouin lifestyle is no longer limited to the classification of a pastoral nomad, located in the desert living in goat-hair tents, and herding camels. While there are several factors that could be taken into consideration in order to unpack what it means to be a Bedouin in a modern world, the development of nation-states within the Middle East and the consequences of these formations have had a significant influence on Bedouin tribal identity and lifestyle. In Jordan, the process of deconstructing Bedouin identity and lifestyle can largely be contributed to detribalization, sedentarization, and nationalization. While these governmental projects have contributed to the decline of nomadic traditional culture, Bedouin have not completely disappeared in Jordan. Rather than ask where have the Bedouin gone? the better question is where are the Bedouin going? particularly in a changing world

    Evaluating the use of smartphone measurement applications for crime scene documentation

    As smartphone technology continues to evolve, it begs the question as to whether or not smartphones could be used by crime scene investigators. Crime scene documentation is a time-consuming task when using a standard tape measure compared to advanced techniques (such as 3D scanners). However, these advanced techniques are expensive and take time to set up. Measurement applications could potentially serve as an affordable and sufficient alternative. Therefore, it’s important to understand how accurate these applications are as well as how well they work for measuring crime scenes and forensic evidence. Eleven measurement applications were downloaded (onto an Apple iPhone XR) and evaluated and then narrowed down to four applications to be used in this study. Several parameters were assessed in all eleven applications. Factors such as tools available, effective maximum distance, ease of use, and accuracy were tested. The applications that best met all criteria were used for this study. First, the applications were used to measure several known dimensions such as a ruler, then they were used to measure room dimensions, outdoor scenes, footwear impressions, and bloodstains. Accuracy was assessed by comparing the results from the applications to the measurements made from a standard tape measure or ruler. Standard deviation of all measurements was calculated and compared against one another. Feasibility was evaluated by recording the amount of time that it took to conduct the measurements and comparing the times to one another. Adaptability to various environments was also factored in when evaluating feasibility. Overall, the applications Tape Measure and MagicPlan were found to be the most accurate and feasible applications to use for crime scene measurements. Moasure was found to be the least efficient and precise application in this study. Measure, Moasure, MagicPlan, and Tape Measure serve as an affordable, fast, and easy presumptive tool for crime scene documentation. In the future, these applications could be used to document crime scenes, and could assist particular agencies with insufficient monetary resources. While the applications selected (Measure, Moasure, MagicPlan, and Tape Measure) proved to be accurate, additional tests should be conducted before being implemented exclusively by investigators. With continual progression in modern technology, substantial improvements are anticipated that encompass these smartphone applications, which will increase reliability as it relates to forensics

    Implementing TK20 Assessment System: A Collaborative Effort

    The purpose of this paper is to share results of a collaborative effort introducing the TK20 assessment system in the Department of Education and Human Development at the College of Brockport, State University of New York. The authors will define and explain the TK 20 assessment system and its relationship to accreditation and standards. We will discuss the process of pilot testing and implementing this assessment system. We will also describe the interdisciplinary efforts for effective collaboration into a college wide assessment system. We will share the impact of such a system on students using a firsthand voice. Readers can take away ideas for similar implementation efforts and assignments

    Self-Reported Head Injury and Risk of Late-Life Impairment and AD Pathology in an AD Center Cohort

    Aims: To evaluate the relationship between self-reported head injury and cognitive impairment, dementia, mortality, and Alzheimer\u27s disease (AD)-type pathological changes. Methods: Clinical and neuropathological data from participants enrolled in a longitudinal study of aging and cognition (n = 649) were analyzed to assess the chronic effects of self-reported head injury. Results: The effect of self-reported head injury on the clinical state depended on the age at assessment: for a 1-year increase in age, the OR for the transition to clinical mild cognitive impairment (MCI) at the next visit for participants with a history of head injury was 1.21 and 1.34 for the transition from MCI to dementia. Without respect to age, head injury increased the odds of mortality (OR = 1.54). Moreover, it increased the odds of a pathological diagnosis of AD for men (OR = 1.47) but not women (OR = 1.18). Men with a head injury had higher mean amyloid plaque counts in the neocortex and entorhinal cortex than men without. Conclusions: Self-reported head injury is associated with earlier onset, increased risk of cognitive impairment and dementia, increased risk of mortality, and AD-type pathological changes

    Shiga toxin-producing Escherichia coli clonal complex 32, including serotype O145:H28, in the UK and Ireland

    Introduction. Shiga toxin-producing Escherichia coli (STEC) O157:H7 has been the most clinically significant STEC serotype in the UK for over four decades. Over the last 10 years we have observed a decrease in STEC O157:H7 and an increase in non-O157 STEC serotypes, such as O145:H28. Gap Statement. Little is known about the microbiology and epidemiology of STEC belonging to CC32 (including O145:H28) in the UK. The aim of this study was to integrate genomic data with patient information to gain a better understanding of the virulence, disease severity, epidemic risk assessment and population structure of this clinically significant clonal complex. Methodology. Isolates of E. coli belonging to CC32 (n=309) in the archives of public health agencies in the UK and Ireland were whole-genome-sequenced, virulence-profiled and integrated with enhanced surveillance questionnaire (ESQ) data, including exposures and disease severity. Results. Overall, diagnoses of STEC belonging to CC32 (290/309, 94 %) in the UK have increased every year since 2014. Most cases were female (61 %), and the highest proportion of cases belonged to the 0–4 age group (53/211,25 %). The frequency of symptoms of diarrhoea (92 %), abdominal pain (84 %), blood in stool (71 %) and nausea (51 %) was similar to that reported in cases of STEC O157:H7, although cases of STEC CC32 were more frequently admitted to hospital (STEC CC32 48 % vs O157:H7  34 %) and/or developed haemolytic uraemic syndrome (HUS) (STEC CC32 9 % vs O157:H7 4 %). The majority of STEC isolates (268/290, 92 %) had the stx2a/eae virulence gene combination, most commonly associated with progression to STEC HUS. There was evidence of person-to-person transmission and small, temporally related, geographically dispersed outbreaks, characteristic of foodborne outbreaks linked to nationally distributed products. Conclusion. We recommend more widespread use of polymerase chain reaction (PCR) for the detection of all STEC serogroups, the development of consistent strategies for the follow-up testing of PCR-positive faecal specimens, the implementation of more comprehensive and standardized collection of epidemiological data, and routine sharing of sequencing data between public health agencies worldwide

    Inference of Relationships in Population Data Using Identity-by-Descent and Identity-by-State

    It is an assumption of large, population-based datasets that samples are annotated accurately whether they correspond to known relationships or unrelated individuals. These annotations are key for a broad range of genetics applications. While many methods are available to assess relatedness that involve estimates of identity-by-descent (IBD) and/or identity-by-state (IBS) allele-sharing proportions, we developed a novel approach that estimates IBD0, 1, and 2 based on observed IBS within windows. When combined with genome-wide IBS information, it provides an intuitive and practical graphical approach with the capacity to analyze datasets with thousands of samples without prior information about relatedness between individuals or haplotypes. We applied the method to a commonly used Human Variation Panel consisting of 400 nominally unrelated individuals. Surprisingly, we identified identical, parent-child, and full-sibling relationships and reconstructed pedigrees. In two instances non-sibling pairs of individuals in these pedigrees had unexpected IBD2 levels, as well as multiple regions of homozygosity, implying inbreeding. This combined method allowed us to distinguish related individuals from those having atypical heterozygosity rates and determine which individuals were outliers with respect to their designated population. Additionally, it becomes increasingly difficult to identify distant relatedness using genome-wide IBS methods alone. However, our IBD method further identified distant relatedness between individuals within populations, supported by the presence of megabase-scale regions lacking IBS0 across individual chromosomes. We benchmarked our approach against the hidden Markov model of a leading software package (PLINK), showing improved calling of distantly related individuals, and we validated it using a known pedigree from a clinical study. The application of this approach could improve genome-wide association, linkage, heterozygosity, and other population genomics studies that rely on SNP genotype data

    Epidemiology and genomic analysis of Shiga toxin-producing Escherichia coli clonal complex 165 in the UK

    Introduction. Shiga toxin-producing Escherichia coli (STEC) is a zoonotic, foodborne gastrointestinal pathogen that has the potential to cause severe clinical outcomes, including haemolytic uraemic syndrome (HUS). STEC-HUS is the leading cause of renal failure in children and can be fatal. Over the last decade, STEC clonal complex 165 (CC165) has emerged as a cause of STEC-HUS. Gap Statement. There is a need to understand the pathogenicity and prevalence of this emerging STEC clonal complex in the UK, to facilitate early diagnosis, improve clinical management, and prevent and control outbreaks. Aim. The aim of this study was to characterize CC165 through identification of virulence factors (VFs) and antimicrobial resistance (AMR) determinants in the genome and to integrate the genome data with the available epidemiological data to better understand the incidence and pathogenicity of this clonal complex in the UK. Methodology. All isolates belonging to CC165 in the archives at the UK public health agencies were sequenced and serotyped, and the virulence gene and AMR profiles were derived from the genome using PHE bioinformatics pipelines and the Centre for Genomic Epidemiology virulence database. Results. There were 48 CC165 isolates, of which 43 were STEC, four were enteropathogenic E. coli (EPEC) and one E. coli. STEC serotypes were predominately O80:H2 (n=28), and other serotypes included O45:H2 (n=9), O55:H9 (n=4), O132:H2 (n=1) and O180:H2 (n=1). All but one STEC isolate had Shiga toxin (stx) subtype stx2a or stx2d and 47/48 isolates had the eae gene encoding intimin involved in the intimate attachment of the bacteria to the human gut mucosa. We detected extra-intestinal virulence genes including those associated with iron acquisition (iro) and serum resistance (iss), indicating that this pathogen has the potential to translocate to extra-intestinal sites. Unlike other STEC clonal complexes, a high proportion of isolates (93%, 40/43) were multidrug-resistant, including resistance to aminoglycosides, beta-lactams, chloramphenicol, sulphonamides, tetracyclines and trimethoprim. Conclusion. The clinical significance of this clonal complex should not be underestimated. Exhibiting high levels of AMR and a combination of STEC and extra-intestinal pathogenic E. coli (ExPEC) virulence profiles, this clonal complex is an emerging threat to public health

    Liver injury and fibrosis induced by dietary challenge in the ossabaw miniature Swine

    BACKGROUND: Ossabaw miniature swine when fed a diet high in fructose, saturated fat and cholesterol (NASH diet) develop metabolic syndrome and nonalcoholic steatohepatitis (NASH) characterized by liver injury and fibrosis. This study was conducted to further characterize the development of NASH in this large animal model. METHODS: Ossabaw swine were fed standard chow (control group; n = 6) or NASH diet (n = 6) for 24 weeks. Blood and liver tissue were collected and liver histology were characterized at 0, 8, 16 and 24 weeks of dietary intervention. Hepatic apoptosis and lipid levels were assessed at week 24. RESULTS: The NASH diet group developed metabolic syndrome and progressive histologic features of NASH including: (a) hepatocyte ballooning at 8 weeks which progressed to extensive ballooning (>90% hepatocytes), (b) hepatic fibrosis at week 16, which progressed to moderate fibrosis, and (c) Kupffer cell accumulation with vacuolization at 8 weeks which progressed through week 24. The NASH diet group showed increased hepatocyte apoptosis that correlated with hepatic total and free cholesterol and free fatty acids, but not esterified cholesterol or triglycerides. CONCLUSIONS: This report further characterizes the progression of diet-induced NASH in the Ossabaw swine model. In Ossabaw swine fed the NASH diet: (a) hepatocyte injury and fibrosis can occur without macrovesicular steatosis or excess triglyceride accumulation; (b) hepatocyte ballooning generally precedes the development of fibrosis; (c) there is increased hepatocyte apoptosis, and it is correlated more significantly with hepatic free cholesterol than hepatic free fatty acids and had no correlation with hepatic triglycerides
