    A Qualitative Examination of Graduate Advising Relationships:The Advisee Perspective

    Sixteen 3rd-year counseling psychology doctoral students were interviewed about their relationships with their graduate advisors. Of those students, 10 were satisfied and 6 were unsatisfied with their advising relationships. Satisfied and unsatisfied students differed on several aspects of the advising relationship, including (a) the ability to choose their advisors, (b) the frequency of meetings with their advisors, (c) the benefits and costs associated with their advising relationships, and (d) how conflict was dealt with in the advising relationship. Furthermore, all of the satisfied students reported that their advising relationships became more positive over time, whereas many of the unsatisfied students reported that their advising relationships got worse (e.g., became more distant) over time

    Effects of Fructose-Derived Advanced Glycation End Products on Acetylation of Histones in the Brain

    Objective: The objective of this study was to determine the effects of fructose and their advanced glycation end products (fru-AGES) on histone acetylation in microglia, the immune cells of the brain. Significance: Fru-AGES primarily form as a result of non-enzymatic reactions between fructose and proteins. One result is inflammation in the brain, which can be directly correlated to increased microglia activity. Microglial activity has been shown to be associated with the acetylation of histones, resulting in a change in transcription of inflammatory genes. Elucidation of a direct link between fructose, fru-AGES and histone acetylation would increase understanding the pathophysiology of inflammatory disorders such as Alzheimer’s disease. Experimental Procedures: An immortalized rat microglial cell line was treated in vitro with control media, fru-AGES or fructose. Histone acetylation was analyzed indirectly through activity of histone deacetlyase (HDAC) using the HDAC Glo I/II Assay (Promega). Chemiluminescent product formation was measuring with a spectrophotometer. Results Obtained: Both treatments with fructose and fru-AGES showed an increase in HDAC activity compared to control by up to 35% and 20%, respectively, correlating to a decrease in global histone acetylation. This is contradictory to initial expectations, as a decrease in acetylation could result in a decrease in transcription of genes . Despite causing an initial inflammatory response, fructose and fru-AGES appear to suppress overall gene transcription. Conclusion: Previous data show that exposure of microglia to fructose and fru-AGES results in a pro-inflammatory activated state. However, at the level of gene transcription, microglia may be desensitized and less able to respond in the long term

    SGAS 143845.1+145407: A Big, Cool Starburst at Redshift 0.816

    We present the discovery and a detailed multi-wavelength study of a strongly-lensed luminous infrared galaxy at z=0.816. Unlike most known lensed galaxies discovered at optical or near-infrared wavelengths this lensed source is red, r-Ks = 3.9 [AB], which the data presented here demonstrate is due to ongoing dusty star formation. The overall lensing magnification (a factor of 17) facilitates observations from the blue optical through to 500micron, fully capturing both the stellar photospheric emission as well as the re-processed thermal dust emission. We also present optical and near-IR spectroscopy. These extensive data show that this lensed galaxy is in many ways typical of IR-detected sources at z~1, with both a total luminosity and size in accordance with other (albeit much less detailed) measurements in samples of galaxies observed in deep fields with the Spitzer telescope. Its far-infrared spectral energy distribution is well-fit by local templates that are an order of magnitude less luminous than the lensed galaxy; local templates of comparable luminosity are too hot to fit. Its size (D~7kpc) is much larger than local luminous infrared galaxies, but in line with sizes observed for such galaxies at z~1. The star formation appears uniform across this spatial scale. In this source, the luminosity of which is typical of sources that dominate the cosmic infrared background, we find that star formation is spatially extended and well organised, quite unlike the compact merger-driven starbursts which are typical for sources of this luminosity at z~0.Comment: 18 pages, 10 figure

    An enhanced CRISPR repressor for targeted mammalian gene regulation.

    The RNA-guided endonuclease Cas9 can be converted into a programmable transcriptional repressor, but inefficiencies in target-gene silencing have limited its utility. Here we describe an improved Cas9 repressor based on the C-terminal fusion of a rationally designed bipartite repressor domain, KRAB-MeCP2, to nuclease-dead Cas9. We demonstrate the system's superiority in silencing coding and noncoding genes, simultaneously repressing a series of target genes, improving the results of single and dual guide RNA library screens, and enabling new architectures of synthetic genetic circuits
