    Assessment of genotype x environment interaction and stability of sesame (Sesamum indicum L.) seed yield under rainfed and supplementary irrigation in central Sudan

    ABSTRACT       Sesame knowledge of genotype x environment interaction (GEI) is advantageous in order to have a cultivar that gives consistently high yield in a broad range of environments and to increase the efficiency of breeding programs and selection of the best genotypes. Fifteen genotypes of sesame (Sesamum  indicum L.) were evaluated during 2011 and 2012 rainy seasons, at Wad Medani, Rahad (under supplementary irrigation) and Gedarif (under rainfed), to assess genotype x environment interaction and stability of seed yield. A randomized complete block design with four replicates was used in each location. The analysis of variance procedure revealed highly significant differences among the 15 sesame genotypes for seed yield over the eight environments. The mean squares of environment, genotype and genotype x environment interaction were highly significant for seed yield. Both statistical stability models, i. e. Eberhart and Russell (1966) as well as the Additive Main Effect and Multiplicative Interaction (AMMI) analysis, indicated that genotypes Elgezouli, Promo, Um Shagra and Kenana-2 were the most high yielding and stable genotypes and can be recommended for both rainfed and irrigated areas of central Sudan.                من ميزات دراسة التفاعل الوراثي- البيئي في محصول السمسم معرفة افضل الاصناف ذات الانتاجية العالية في مدي واسع من البيئات المختلفة مما يزيد من كفاءة برامج التربية المستعملة وانتخاب افضل الاصناف. تم تقويم 15 سلالة من السمسم في الموسمين الزراعيين 2011 و2012م في ثلاثة مواقع هي مدني، الرهد (ري تكميلي) والقضارف (ري مطري)، وذلك لتقويم التفاعل الوراثي والبيئي وثبات درجة انتاجية بذور السمسم. استخدم تصميم القطاعات العشوائية الكاملة بأربع مكررات. اظهرت نتائج تحليل التباين لعدد 15 سلالة من السمسم فروقات معنوية عالية جدا لمعظم الصفات التي درست في كل المواقع والمواسم مع معنوية للتفاعل الوراثي والبيئي لإنتاجية البذور. بناءاً علي نماذج التحليل Eberhart and Russel (1966)              وAdditive Main Effect and Multiplicative Interaction(AMMI) لتحديد ثبات الاداء معاً وجد ان انتاجية بذور الاصناف الجزولي، برومو، ام شجرة وكنانة-2 ثابتة وعالية في كل المواقع والمواسم. وبناءاً علي ذلك يوصي بزراعة هذه الاصناف في المناطق المطرية والمروية في وسط السودان.                   &nbsp

    Immunohistochemical Expression of “c-MET” in Breast Carcinomas

    BACKGROUND: Targeted therapies achieved great success in managing breast cancer, however, triple negative breast cancers (TNBCs) lack the expression of traditional therapeutic targets, and other subtypes develop resistance to current therapies. The c-MET receptor emerged as a potential target with well documented pro-proliferative pro-motility downstream signals and wide network of crosstalk with other effectors. Some reports describe a preferential expression of c-MET in TNBCs and promising results in early anti-c-MET clinical trials. However, the main cause of failure of these trials was attributed to patient selection. AIM: The objectives of the study were to assessment of c-MET in subtypes of breast cancer and its association with other clinicopathological variables that may predict its expression and possibly establish other rationales to refine patient selection for clinical trials. MATERIALS AND METHODS: Retrospective immunohistochemical study assessing c-MET (clone SP44) in 55 cases of breast carcinoma. The expression of c-MET> 5% was considered positive. RESULTS: c-MET was detected in 42% of cases. A statistically significant association of c-MET with extremes of age, advanced prognostic stage, carcinoma with medullary features, and high tumor infiltrating lymphocytes (TILs) was observed for the first time in our study. Grade III, hormone receptor negativity and TNBCs were also significantly associated with c-MET. Only negative progesterone receptor (PR) and high TILs were independently associated with c-MET in a multivariate analysis (p < 0.05). No significant association between c-MET and multifocality, size, node status, anatomic stage, lymphovascular or perineural invasion, and Ki-67 expression. CONCLUSION: PR negativity and high TILs might be useful c-MET predictors and selection tools for clinical trials but further studies are needed to validate the unprecedented findings which may not only aid in patient selection but may also inspire new paradigms in future studies

    Prosopis juliflora leave extracts induce cell death of MCF-7, HepG2, and LS-174T cancer cell lines

    Prosopis juliflora (P. juliflora) is a widespread phreatophytic tree, which belongs to the Fabaceae family. The goal of the present study is to investigate the potential anti-cancer effect of P. juliflora leave extracts and to identify its chemical composition. For this purpose, MCF-7 (breast), HepG2 (liver), and LS-174T (colorectal) cancer cell lines were cultivated and incubated with various concentrations of P. juliflora leave extracts, and its impact on cell viability, proliferation, and cell cycle stages was investigated. P. juliflora leave extracts induced concentration-dependent cytotoxicity against all tested cancer cell lines. The calculated IC50 was 18.17, 33.1 and 41.9 μg/ml for MCF-7, HePG2 and LS-174T, respectively. Detailed analysis revealed that the cytotoxic action of P. juliflora extracts was mainly via necrosis but not apoptosis. Moreover, DNA content flow cytometry analysis showed cell-specific anti-proliferative action and cell cycle stages arrest. In order to identify the anti-cancer constituents of P. juliflora, the ethyl extracts were analyzed by liquid chromatography-mass spectrometry. The major constituents identified in the ethyl extracts of P. juliflora leaves were hydroxymethyl-pyridine, nicotinamide, adenine, and poly-(methyl methacrylate) (PMMA). In conclusion, P. juliflora ethyl acetate extracts have a potential anti-cancer effect against breast adenocarcinoma, hepatocellular carcinoma, and colorectal adenocarcinoma, and is enriched with anti-cancer constituents

    Optimized D-α-tocopherol polyethylene glycol succinate/phospholipid self-assembled mixed micelles: A promising lipid-based nanoplatform for augmenting the antifungal activity of fluconazole

    Fluconazole (FLZ) is the most widely used antifungal agent for treating cutaneous candidiasis. Although oral FLZ has been proved to be effective, the incidence of side effects necessitates the development of an effective formulation that could surpass the pitfalls associated with systemic availability. Accordingly, this research aimed at developing a self-assembled mixed micelles topical delivery system to enhance the topical delivery of the drug. Self-assembled mixed micelles were developed using D-α-tocopheryl polyethylene glycol 1000 succinate and phospholipids and optimized using Box-Behnken design. The optimized formulation with minimized size was then tested in vivo for the antifungal activity against C. albicans in immunocompromised mice. Treatment with the optimized formulation led to decreased peripheral erythema as well as lesions due to fungal infection in comparison to raw FLZ loaded gel. Therefore, the developed formulation was found to be a promising vehicle for the treatment of cutaneous candidiasis

    Proučavanje relativističke tvorbe hadrona prema naprijed i unatrag u sudarima 3he i 4he s jezgrama u emulziji na energiji ubrzivača u dubni

    The experimental results on 3He- and 4He-emulsion interactions accompanied by relativistic (shower) hadrons flying into the backward (θlab ≤ 90◦ ) hemisphere at 4.5 AGeV/c are presented and analyzed. The dependence of the probabilities of these interactions on the different target sizes, impact parameter and projectile spectator charges is studied. An investigation of average values and multiplicity distributions of these hadrons for the interactions with light and heavy emulsion nuclei has been carried out. In addition, the correlations between the multiplicities of different types of the emitted particles are studied. The data show that backward shower particles are a sensitive target parameter. The values of impact parameters can be used as good indicators for selecting events which occured with light or heavy emulsion nuclei. A comparison with the modified cascade model shows a good performance in describing the data produced in the region having less cascading (i.e. interactions with light nuclei). As for the interactions with heavy nuclei, the model overestimates the experimental data.Predstavljamo eksperimentalne rezultate i analize mjerenja međudjelovanja 3He i 4He u emulziji na 4.5 AGeV/c, u kojima se opažaju relativistički pljuskovi hadrona koji lete unatrag (θlab ≤ 90◦ ). Proučavamo ovisnost vjerojatnosti tih međudjelovanja o veličini jezgre mete, sudarnom parametru i naboju projektila-promatrača. Istražili smo prosječne vrijednosti i raspodjele višestrukosti hadrona za lake i teške jezgre u emulziji. Nadalje, proučavali smo korelacije višestrukosti različitih izlaznih čestica. Podaci pokazuju da su pljuskovi čestica unatrag osjetljiv parametar jezgri mete. Vrijednosti parametara mogu poslužiti kao dobar pokazatelj sudara s lakim odnosno teškim jezgrama. Usporedba s promijenjenim kaskadnim modelom pokazuje dobro slaganje s podacima u kojima je manje kaskada (tj. Manje sudara s lakim jezgrama). Rezultati tog modela za teške jezgre veći su od eksperimentalnih podataka

    Urocortin Neuropeptide Levels Are Impaired in the PBMCs of Overweight Children

    The corticotropin-releasing hormone (CRH) and urocortins (UCNs) have been implicated in energy homeostasis and the cellular stress response. However, the expression of these neuropeptides in children remains unclear. Therefore, we determined the impact of obesity on their expression in 40 children who were normal weight, overweight, and had obesity. Peripheral blood mononuclear cells (PBMCs) and plasma were used to assess the expression of neuropeptides. THP1 cells were treated with 25 mM glucose and 200 µM palmitate, and gene expression was measured by real-time polymerase chain reaction (RT-PCR). Transcript levels of neuropeptides were decreased in PBMCs from children with increased body mass index as indicated by a significant decrease in UCN1, UCN3, and CRH mRNA in overweight and obese children. UCN3 mRNA expression was strongly correlated with UCN1, UCN2, and CRH. Exposure of THP1 cells to palmitate or a combination of high glucose and palmitate for 24 h increased CRH, UCN2, and UCN3 mRNA expression with concomitant increased levels of inflammatory and endoplasmic reticulum stress markers, suggesting a crosstalk between these neuropeptides and the cellular stress response. The differential impairment of the transcript levels of CRH and UCNs in PBMCs from overweight and obese children highlights their involvement in obesity-related metabolic and cellular stress
