    Grain size effects in rime judgment across literacy development in German

    Phonological similarity effects are biases to judge words as phonologically similar (i.e., rhyming), even if they are not. First found in rime awareness tasks in preliterates, these biases have recently also been found in proficient adult readers. In this study, we evaluated underlying phonological processing in rime judgment longitudinally, across literacy development. To this end, we created a new rime judgment task with two distractor conditions, that varied in size of phonological overlap. The task was administered to a group of 61 German speaking children at four time-points across school entry and to 21 adults. Accuracy and latency responses were recorded. Results showed that children and adults showed phonological similarity effects but the effect decreased gradually over time. However, preliterate children were more sensitive to large compared to small phonological overlap, while the same effect was significantly smaller in literate children and adults. Results suggests that preliterate children are more sensitive to larger grain sizes and become more sensitive to fine-grained units across literacy development.     MAPK phosphatase-1 represents a novel antiinflammatory target of glucocorticoids in the human endothelium

    Glucocorticoids are well-established anti- inflammatory drugs thought to mainly act by inhibition of proinflammatory transcription factors like NF-κB. In recent years, however, transcription factorindependent mechanisms of glucocorticoid action have been proposed, namely the influence on MAPK pathways. Here we identify MAPK phosphatase-1 (MKP-1) as a pivotal mediator of the anti-inflammatory action of glucocorticoids in the human endothelium. We applied dexamethasone (Dex) to TNF-α-activated human endothelial cells and used the adhesion molecule E-selectin as inflammatory read-out parameter. Dex is known to reduce the expression of E-selectin, which is largely regulated by NF-κB. Here, we communicate that Dex at low concentrations (1–100 nM) markedly attenuates E-selectin expression without affecting NF-κB. Importantly, Dex is able to increase the expression of MKP-1, which causes an inactivation of TNF-α-induced p38 MAPK and mediates inhibition of E-selectin expression. In endothelial MKP-1ˉ/ˉ cells differentiated from MKP-1ˉ/ˉ embryonic stem cells and in MKP-1-silenced human endothelial cells, Dex did not inhibit TNF-α-evoked E-selectin expression. Thus, our findings introduce MKP-1 as a novel and crucial mediator of the anti-inflammatory action of glucocorticoids at low concentrations in the human endothelium and highlight MKP-1 as an important and promising antiinflammatory drug target

    High-Resolution Identification of Multiple Salmonella Serovars in a Single Sample by Using CRISPR-SeroSeq

    Salmonella enterica is represented by \u3e2,600 serovars that can differ in routes of transmission, host colonization, and in resistance to antimicrobials. S. enterica is the leading bacterial cause of foodborne illness in the United States, with well-established detection methodology. Current surveillance protocols rely on the characterization of a few colonies to represent an entire sample; thus, minority serovars remain undetected. Salmonella contains two CRISPR loci, CRISPR1 and CRISPR2, and the spacer contents of these can be considered serovar specific. We exploited this property to develop an amplicon-based and multiplexed sequencing approach, CRISPR-SeroSeq (serotyping by sequencing of the CRISPR loci), to identify multiple serovars present in a single sample. Using mixed genomic DNA from two Salmonella serovars, we were able to confidently detect a serovar that constituted 0.01% of the sample. Poultry is a major reservoir of Salmonella spp., including serovars that are frequently associated with human illness, as well as those that are not. Numerous studies have examined the prevalence and diversity of Salmonella spp. in poultry, though these studies were limited to culture-based approaches and therefore only identified abundant serovars. CRISPR-SeroSeq was used to investigate samples from broiler houses and a processing facility. Ninety-one percent of samples harbored multiple serovars, and there was one sample in which four different serovars were detected. In another sample, reads for the minority serovar comprised 0.003% of the total number of Salmonella spacer reads. The most abundant serovars identified were Salmonella enterica serovars Montevideo, Kentucky, Enteritidis, and Typhimurium. CRISPR-SeroSeq also differentiated between multiple strains of some serovars. This high resolution of serovar populations has the potential to be utilized as a powerful tool in the surveillance of Salmonella species

    A meta-analysis on the heritability of vertebrate telomere length

    Telomere dynamics are linked with both cellular and organismal senescence, and life history, individual quality and health. Telomere dynamics, particularly telomere length, have therefore garnered much research interest in evolutionary biology. To examine the evolution of telomere length, it is important to quantify its heritability, the proportion of total variation explained by additive genetic effects. Many studies have quantified telomere length heritability, but estimates are varied, and no general conclusion has been drawn. Additionally, it is unclear whether biological and methodological factors influence telomere length heritability estimates. We present the first meta-analysis of telomere length heritability, using 104 estimates from 43 studies over 18 vertebrate species. We calculated an overall mean heritability and examined how estimates varied by study, phylogeny, species-specific ecology, environmental setting, age at sampling, laboratory methods, statistical methods, sex and repeated measurements. Overall heritability was moderate (44.9%, 95% CI: 25.2-64.7%), and there was considerable heterogeneity in heritability estimates, in particular among studies and estimates. Laboratory method influenced heritability estimates, with in-gel hybridization TRF yielding higher heritabilities than qPCR and Southern blot TRF. There was also an effect from statistical method, with twin-based and SNP-based estimates lower than correlation-based or pedigree-based estimates. Our results highlight an overall heritable basis of telomere length, and we recommend future research on a wider range of taxa, and the use of variance-partitioning methods with relatedness or SNP data over correlation methods to minimize heritability estimation bias

    Hintergrund: Brustschmerz ist eine der am häufigsten präsentierten somatischen Beschwerden in medizinischen Einrichtungen der primären und sekundären Versorgung. Dennoch kann bei bis zu 66 % der Betroffenen keine ursächliche kardiale Pathologie festgestellt werden – man spricht von nicht-kardialen Brustschmerzen, für deren Entstehung und Verlauf verschiedene physiologische, aber auch psychologische Bedingungsfaktoren (z. B. subjektive Krankheitsrepräsentationen) angenommen werden. Die einfache ärztliche Rückmeldung über unauffällige kardiodiagnostische Befunde erweist sich zwar kurzfristig als beruhigend, scheint langfristig jedoch nicht ausreichend, um dysfunktionale Krankheitsrepräsentationen, Ängste und Sorgen der Betroffenen langfristig zu reduzieren.     Airborne radar sounding evidence for deformable sediments and outcropping bedrock beneath Thwaites Glacier, West Antarctica

    The geologic and morphologic records of prior ice sheet configurations show evidence of rapid, back-stepping, meltwater intensive retreats. However, the potential for such a retreat in a contemporary glacier depends on the lithology of the current ice sheet bed, which lies beneath kilometers of ice, making its physical properties difficult to constrain. We use radar sounding and marine bathymetry data to compare the bed configuration of Thwaites Glacier to the bed of paleo-Pine Island Glacier. Using observed and modeled radar scattering, we show that the tributaries and upper trunk of Thwaites Glacier are underlain by ice flow-aligned bedforms consistent with deformable sediment and that the lower trunk is grounded on a region of high bed roughness consistent with outcropping bedrock. This is the same configuration as paleo-Pine Island Glacier during its retreat across the inner continental shelf
