    Hydrogen Sulfide, a Toxic Gas with Cardiovascular Properties in Uremia: How Harmful Is It?

    Hydrogen sulfide (H2S) is a poisonous gas which can be lethal. However, it is also produced endogenously, thus belonging to the family of gasotransmitters along with nitric oxide and carbon monoxide. H2S is in fact involved in mediating several signaling and cytoprotective functions, for example in the nervous, cardiovascular, and gastrointestinal systems, such as neuronal transmission, blood pressure regulation and insulin release, among others. When increased, it can mediate inflammation and apoptosis, with a role in shock. When decreased, it can be involved in atherosclerosis, hypertension, myocardial infarction, diabetes, sexual dysfunction, and gastric ulcer; it notably interacts with the other gaseous mediators. Cystathionine γ-lyase, cystathionine β-synthase, and 3-mercaptopyruvate sulfurtransferase are the principal enzymes involved in H2S production. We have recently studied H2S metabolism in the plasma of chronic hemodialysis patients and reported that its levels are significantly decreased. The plausible mechanism lies in the transcription inhibition of the cystathionine γ-lyase gene. The finding could be of importance considering that hypertension and high cardiovascular mortality are characteristic in these patients

    The Wide Range of Antibiotic Resistance and Variability of Genotypic Profiles in Escherichia coli from Domestic Animals in Eastern Sicily

    serious public health threat. Escherichia coli, a usual host of intestinal microbiota, is recognized also as etiological agent of numerous infections widespread in both humans and animals. The colibacillosis is one of the most reported zoonoses worldwide, typically treated with antibiotics in the primary stages. This strategy has promoted the onset of antibiotic-resistant serotypes of E. coli, reducing the effectiveness of therapeutic treatments and contributing to antibiotic resistance spread. The current study focused on biodiversity, pathogenicity, and antibiotic resistance profile of 104 E. coli strains isolated from domestic animals in Eastern Sicily. The strains were isolated from sick animals and carcasses of six different animal species and screened for resistance against 16 antibiotic molecules, as recommended by WHO and OIE. The antibiotic resistance patterns highlighted that all strains were multi-resistant, showing resistance to at least three antibiotic classes. The highest incidence of resistance was observed against amoxicillin (100%), tylosin (97%), sulfamethoxazole (98%), and erythromycin (92%), while the lowest for colistin (8%). The pathotype characterization identified two EPEC strains and the study of genetic linkage (PFGE) showed a wide variety of profiles.     Anti-mycobacterial activity of labdane and halimane diterpenes obtained from Plectranthus ornatus Codd

Vários diterpenos das espécies Plectranthus foram referidos com actividade tuberculostática interessante, sendo que o P. omatus Codd. é usado como anti-infeccioso em algumas regiões do Brasil. Em trabalhos anteriores, um diterpeno de esqueleto de halimano outro labdano foram isolados a partir de P. ornatus Codd. Neste trabalho, avaliou-se preliminarmente a sua actividade micobacteriana com uma estirpe não virulenta de Mycobacterium smegmatis. A citotoxicidade dos compostos foi testada medindo a libertação de lactato desidrogenase (LDH) não sendo encontrados efeitos citotóxicos consideráveis até 25 ug/mL. Posteriormente, o método de microdiluição foi utilizado para determinar a concentração mínima inibitória (CMI) de M. smegmatis. A CMI>99% para o esqueleto de halimano foi 100 ug/mL e >100 ug/mL para o esqueleto de labdano. De acordo com o nosso conhecimento, este é o primeiro estudo usando diterpenos de esqueleto de halimano e de labdano isolados a partir de P. omatus em ensaios de citotoxicidade em macrófagos, e num ensaio preliminar sobre a sua atividade anti-micobacteriana. Estudos futuros são sugeridos na estirpe virulenta de M. tugerculosis, particularmente para os diterpenos de esqueleto de halimano.Plectranthus spp. have been reported to have interesting tuberculostatic activity and P. ornatus Codd. has been used in some regions of Brazil as an anti-infective. Previously, diterpenes with halimane and labdane skeletons were isolated in large quantities from P. ornatus Codd. We assessed the anti-mycobacterial activity of these compounds, performing a preliminar assay with the non-virulent strain Mycobacterium smegmatis. The cytotoxicity of the diterpenes with halimane and labdane skeletons was tested with the lactate dehydrogenase assay, where no considerable cytotoxic effects were found up to 25 μg.mL-1. Subsequently, the microdilution method was used to determine the minimum inhibitory concentration (MIC) against M. smegmatis. The MIC that inhibited the growth of the non-virulent mycobacteria by ≥99% was 100 μg.mL-1 for the diterpene with halimane skeleton, whereas for the diterpene with a labdane skeleton was >100 μg.mL-1. To the best of our knowledge, this is the first report on diterpenes with halimane and labdane skeletons isolated from P. ornatus in macrophages cytotoxicity, and in a preliminar assay for anti-mycobacterial activity. Further studies are suggested on M. tuberculosis, particularly for the diterpenes with halimane skeleton

    Differential expression of microRNA501-5p affects the aggressiveness of clear cell renal carcinoma

    AbstractRenal cell carcinoma is a common neoplasia of the adult kidney that accounts for about 3% of adult malignancies. Clear cell renal carcinoma is the most frequent subtype of kidney cancer and 20–40% of patients develop metastases. The absence of appropriate biomarkers complicates diagnosis and prognosis of this disease. In this regard, small noncoding RNAs (microRNAs), which are mutated in several neoplastic diseases including kidney carcinoma, may be optimal candidates as biomarkers for diagnosis and prognosis of this kind of cancer. Here we show that patients with clear cell kidney carcinoma that express low levels of miR501-5p exhibited a good prognosis compared with patients with unchanged or high levels of this microRNA. Consistently, in kidney carcinoma cells the downregulation of miR501-5p induced an increased caspase-3 activity, p53 expression as well as decreased mTOR activation, leading to stimulation of the apoptotic pathway. Conversely, miR501-5p upregulation enhanced the activity of mTOR and promoted both cell proliferation and survival. These biological processes occurred through p53 inactivation by proteasome degradation in a mechanism involving MDM2-mediated p53 ubiquitination. Our results support a role for miR501-5p in balancing apoptosis and cell survival in clear cell renal carcinoma. In particular, the downregulation of microRNA501-5p promotes a good prognosis, while its upregulation contributes to a poor prognosis, in particular, if associated with p53 and MDM2 overexpression and mTOR activation. Thus, the expression of miR501-5p is a possible biomarker for the prognosis of clear cell renal carcinoma

    Valorisation of typical products through characterising and promoting actions: morpho-biometric traits, sensory analysis and flavonol content in Cipolla di Giarratana

    The performances of 9 accessions of Cipolla di Giarratana landrace were evaluated at 5 different sites located 2 in the area where this landrace is traditionally grown (South-Eastern Sicily, 550 m asl), 2 in the neighbor areas with similar average altitude, and one on the South East coast of Sicily (sea level). Biometric and morphological traits, flavonol content and sensory profile were all considered in the valorisation of a typical product of a marginal area of Sicily. Data on the actual yield and crop management at farm scale were also obtained involving more than half of the growers actually cultivating Cipolla di Giarratana , and measuring yield, plant density, and bulb weight directly on the farms. The Cipolla di Giarratana landrace is characterised by a large and heavy bulb (535 g), strongly flattened at the poles, with the maximum diameter at the central section, and white in colour. Significant differences in bulb weight and diameter were found among accessions and locations, but there were no interactions between them and one accession reported the highest bulb weight value in all of the locations studied. With regards to bulb weight, farm scale data confirm the results we obtained at the experimental locations. Fifty percent of registered yield at farm scale ranged between 86 and 152 t ha –1 with a median of 119 t ha –1 , and measured plant density revealed that 50% of the farmers planted 20-27 plants per square meter. Analysis of the UV-vis and mass spectra showed the presence of 10 different flavonols with Quercetin as the most represented flavonol. The sensory profile of Cipolla di Giarratana onion showed that this landrace is characterised by a high intensity of sweet, typical fresh flavour and texture perception

    The prevalence of peripheral and central hearing impairment and its relation to cognition in older adults.

    Age-related hearing loss (ARHL) and dementia are two highly prevalent conditions in the adult population. Recent studies have suggested that hearing loss is independently associated with poorer cognitive functioning. The aim of this study was to evaluate the prevalence of ARHL and cognitive impairment in a large sample of subjects older than 65 years and to correlate hearing function with cognitive function. A total of 488 subjects older than 65 years (mean age 72.8 years) participating in the Great Age Study underwent a complete audiological, neurological and neuropsychological evaluation as part of a multidisciplinary assessment. The prevalence of a hearing loss greater than 25 dB HL was 64.1%, of Central Auditory Processing Disorder (CAPD) was 14.3 and 25.3% of the subjects reported a hearing handicap as reported on the Hearing Handicap Inventory for the Elderly Screening Version questionnaire. Multiple logistic regression analysis corrected for gender, age and education duration showed that mild cognitive impairment (MCI) was significantly associated with hearing impairment (CAPD and hearing threshold; odds ratio 1.6, p = 0.05) and that Alzheimer's disease (AD) was significantly associated with CAPD (odds ratio 4.2, p = 0.05). Given that up to 80% of patients affected by MCI convert to AD, adding auditory tests to a screening cognitive battery might have value in the early diagnosis of cognitive decline

    The J2-Immortalized Murine Macrophage Cell Line Displays Phenotypical and Metabolic Features of Primary BMDMs in Their M1 and M2 Polarization State

    Macrophages are immune cells that are important for the development of the defensive front line of the innate immune system. Following signal recognition, macrophages undergo activation toward specific functional states, consisting not only in the acquisition of specific features but also of peculiar metabolic programs associated with each function. For these reasons, macrophages are often isolated from mice to perform cellular assays to study the mechanisms mediating immune cell activation. This requires expensive and time-consuming breeding and housing of mice strains. To overcome this issue, we analyzed an in-house J2-generated immortalized macrophage cell line from BMDMs, both from a functional and metabolic point of view. By assaying the intracellular and extracellular metabolism coupled with the phenotypic features of immortalized versus primary BMDMs, we concluded that classically and alternatively immortalized macrophages display similar phenotypical, metabolic and functional features compared to primary cells polarized in the same way. Our study validates the use of this immortalized cell line as a suitable model with which to evaluate in vitro how perturbations can influence the phenotypical and functional features of murine macrophages
