    Insights into the mode of action of a putative zinc transporter CzrB in thermus thermophilus

NMR, X-ray scattering and size exclusion chromatography provide support for dimer formation. Full-length variants of CzrB in the apo and zinc-loaded states were generated by homology modelling with the Zn2+ / H+ antiporter YiiP. The model suggests a way in which zinc binding to the cytoplasmic fragment creates a docking site to which a metallochaperone can bind for delivery and transport of its zinc cargo. Since the cytoplasmic domain may exist in the cell as an independent, soluble protein a proposal is advanced that it functions as a metallochaperone and that it regulates the zinc-transporting activity of the full-length protein. The latter requires that zinc binding becomes uncoupled from the creation of a metallochaperone-docking site on CzrB

    A Multilevel Mhealth intervention Boosts adherence to Hydroxyurea in individuals With Sickle Cell Disease

    Hydroxyurea reduces sickle cell disease (SCD) complications, but medication adherence is low. We tested 2 mobile health (mHealth) interventions targeting determinants of low adherence among patients (InCharge Health) and low prescribing among providers (HU toolbox) in a multi-center, non-randomized trial of individuals with SCD ages 15-45. We compared the percentage of days covered (PDC), labs, healthcare utilization, and self-reported pain over 24 weeks of intervention and 12 weeks post-study with a 24-week preintervention interval. We enrolled 293 patients (51% male; median age 27.5 years, 86.8% HbSS/HbSβ0-thalassemia). The mean change in PDC among 235 evaluable subjects increased (39.7% to 56.0%; P \u3c 0.001) and sustained (39.7% to 51.4%, P \u3c 0.001). Mean HbF increased (10.95% to 12.78%; P = 0.03). Self-reported pain frequency reduced (3.54 to 3.35 events/year; P = 0.041). InCharge Health was used ≥1 day by 199 of 235 participants (84.7% implementation; median usage: 17% study days; IQR: 4.8-45.8%). For individuals with ≥1 baseline admission for pain, admissions per 24 weeks declined from baseline through 24 weeks (1.97 to 1.48 events/patient, P = 0.0045) and weeks 25-36 (1.25 events/patient, P = 0.0015). PDC increased with app use (P \u3c 0.001), with the greatest effect in those with private insurance (P = 0.0078), older subjects (P = 0.033), and those with lower pain interference (P = 0.0012). Of the 89 providers (49 hematologists, 36 advanced care providers, 4 unreported), only 11.2% used HU toolbox ≥1/month on average. This use did not affect change in PDC. Tailoring mHealth solutions to address barriers to hydroxyurea adherence can potentially improve adherence and provide clinical benefits. A definitive randomized study is warranted.     Mating skew in Barbary macaque males: the role of female mating synchrony, female behavior, and male–male coalitions

    A fundamental question of sexual selection theory concerns the causes and consequences of reproductive skew among males. The priority of access (PoA) model (Altmann, Ann NY Acad Sci 102:338–435, 1962) has been the most influential framework in primates living in permanent, mixed-sex groups, but to date it has only been tested with the appropriate data on female synchrony in a handful of species. In this paper, we used mating data from one large semi-free ranging group of Barbary macaques: (1) to provide the first test of the priority-of-access model in this species, using mating data from 11 sexually active females (including six females that were implanted with a hormonal contraceptive but who showed levels of sexual activity comparable to those of naturally cycling females) and (2) to determine the proximate mechanism(s) underlying male mating skew. Our results show that the fit of the observed distribution of matings with sexually attractive females to predictions of the PoA model was poor, with lower-ranking males mating more than expected. While our work confirms that female mating synchrony sets an upper limit to monopolization by high-ranking individuals, other factors are also important. Coalitionary activity was the main tactic used by males to lower mating skew in the study group. Coalitions were expressed in a strongly age-related fashion and allowed subordinate, post-prime males to increase their mating success by targeting more dominant, prime males. Conversely, females, while mating promiscuously with several males during a given mating cycle, were more likely to initiate their consortships with prime males, thus reducing the overall effectiveness of coalitions. We conclude that high-ranking Barbary macaque males have a limited ability to monopolize mating access, leading to a modest mating skew among them

    The James Webb Space Telescope Mission

    Twenty-six years ago a small committee report, building on earlier studies, expounded a compelling and poetic vision for the future of astronomy, calling for an infrared-optimized space telescope with an aperture of at least 4m4m. With the support of their governments in the US, Europe, and Canada, 20,000 people realized that vision as the 6.5m6.5m James Webb Space Telescope. A generation of astronomers will celebrate their accomplishments for the life of the mission, potentially as long as 20 years, and beyond. This report and the scientific discoveries that follow are extended thank-you notes to the 20,000 team members. The telescope is working perfectly, with much better image quality than expected. In this and accompanying papers, we give a brief history, describe the observatory, outline its objectives and current observing program, and discuss the inventions and people who made it possible.     The influence of trait and induced state impulsivity on food intake in normal-weight healthy women

    Previous research has linked overeating and overweight/obesity to impulsivity. To find out whether impulsivity causes overeating and hence overweight and obesity, we attempted to prime the concept of impulsivity in healthy participants. In a within-subjects design one sample participated in two conditions. In both conditions participants did a priming task that either hinted subtly at the concept of impulsivity or that was neutral in content. Each time the priming task was followed by a bogus taste test. Trait impulsivity was measured by means of a behavioural task and self-report. Firstly, we hypothesized that participants would eat more during the taste test after they had been primed with the concept "impulsivity" compared to after the control session. Secondly, we expected that a more impulsive personality would predict a heightened food intake. Thirdly, we expected that impulsivity would predict food intake better than restraint. The results showed that both the self-report measure of impulsivity and the behavioural task predicted food intake. Restraint did not significantly predict food intake. Primed impulsivity did not increase food intake, possibly because the priming effect did not last long enough

    Treatment engagement in an internet-delivered cognitive behavioral program for pediatric chronic pain

    Internet-delivered cognitive-behavioral therapy (iCBT) is a promising treatment for chronic pain among youth, but effect sizes are small, and strategies aimed at enhancing treatment effects are needed. Participants' engagement with the program may be an important factor in determining treatment outcomes. The primary aim of the current study was to examine the relationship between treatment engagement and treatment outcomes. Secondarily, we sought to characterize participant engagement in an iCBT program for adolescents with chronic pain and their parents. Participants included 134 adolescents randomized to the intervention arm of a controlled trial examining iCBT for chronic pain. Overall engagement with the intervention by adolescents and parents was high. Parental engagement (number of modules completed by parents and number of parent logins) predicted adolescent activity limitations change scores at post-treatment. Contrary to our expectation, adolescent treatment engagement was not predictive of treatment outcomes. Results indicate that parental engagement with the program may be an important predictor of treatment outcomes.
