    Patriarchal theory reconsidered: torture and gender based violence in Turkey

    Gender theory in general, and patriarchal theory in particular, have been explored in this research to describe the procedures, processes, norms, values and, most importantly, structures that define the subject. Patriarchal theory mostly perceives men as the abusers and women as the abused. However, the nuances and particularities of these oppressive structures have not been explored in detail. In this research, the reader is introduced to the various manifestations of how being privileged and underprivileged is constructed. This research focuses on processes and structures and it mostly explores alternative approaches towards political sociology and its intersection with gender theory. The thesis adopts a multi-level analysis that involves the different manifestations of the ruler-subject binary at the societal and interpersonal levels of analysis. The so-called private and public spheres with their fluid identities have been analyzed after descriptions of internal mechanisms reproducing the social construction of oppression are understood. Similarities between analysis concerned with the household and the public sphere reflects on how binaries such as the masculine and feminine, and the ruler and the subject, reproduce, mirror and reinforce one another. This research, therefore, focuses on structural and systematic ways of reproducing patriarchy as a system that affects the society in an inclusive way. This required and understanding of norms and values that have been analyzed as a reflection of processes that accommodate oppression. The intersection of these processes has led the author to argue that ‘women are to men, what the citizen is for the state, in the context of Turkey’. The feminization of the male political subject has been argued after presenting three chapters that represent my original contribution to knowledge. Through utilizing interviews conducted by other scholars, I initially analyzed male and female statements on domestic violence in Turkey. Second, I analyzed written texts, including official documents, which inform the reader of state officials’ views on gender inequality. Third, I have analyzed the relationship between the state and the citizen through the research I conducted, on police violence during the Gezi Park protests and other interviews include the research conducted with feminist and human rights laywers. The similarities between the manner, processes and values between the male and female (as well as the ruler and the subject) led to a discussion that the male political subject is simultaneously masculine and feminine. The feminization of the male political subject represented an alignment between two spheres that reinforce one another, through mirroring the public and the private. These two systems created a contradiction within the subject often leading him to over-compensate his damage. Therefore, patriarchal relativity was introduced to discuss a perspective on over-compensation amongst subjects and agents that coincide and conflate within vertical patriarchy. Accordingly, new concepts of patriarchy were needed to capture the nuances within a system that defines the subject at macro and micro levels. Throughout this research, the contributions produced by scholars during the past forty years over debates on patriarchal theory have been reproduced to a great extent. This research has utilized a multi-level analysis through comparisons made by references to metaphors. Metaphoric reproduction is a rare approach within patriarchal theory, often different to utilizing a single theoretical framework. The employment of semi-structured and unstructured interviews with additional content analysis substantiates the author’s subjectivity. This subjectivity reflects a feminist understanding of politics, political sociology, philosophy, and gender theory.     DIVERSITY OF MICROFUNGI ON FAGACEAE IN ULUDAG FORESTS

    WOS: 000363091600042Forests ecosystems are sources of oxygen and wood products, also they prevent soil erosion, improve water and air quality, serve as homes for wildlife; and therefore, they preserve and increase biodiversity. Forests can host a diverse community of fungal species with various effects on their host trees. In this research, trees of Fagaceae family of Uludag forests of Bursa province were investigated between the years of 2002 and 2008. By microscopic examination we identified 38 microfungi species in 27 genera belongs to Ascomycota and 1 microfungus species in 1 genus belongs to Basidiomycota. The taxa belong to 15 families: Botryosphaeriaceae, Diaporthaceae, Diatrypaceae, Dothioraceae, Erysiphaceae, Gnomoniaceae, Incertae sedis, Melanconidaceae, Microstromataceae, Nectriaceae, Pseudovalsaceae, Rhytismataceae, Trichosphaeriaceae, Valsaceae and Xylariaceae. The distribution of species by trophic groups revealed a dominance of xylotrophic species. With this study, fungal diversity of Fagaceae family in Uludag forests was identified and included in the mycobiota of Turkey

    Suspect identification based on descriptive facial attributes

    We present a method for using human describable face attributes to perform face identification in criminal inves-tigations. To enable this approach, a set of 46 facial at-tributes were carefully defined with the goal of capturing all describable and persistent facial features. Using crowd sourced labor, a large corpus of face images were manually annotated with the proposed attributes. In turn, we train an automated attribute extraction algorithm to encode target repositories with the attribute information. Attribute extrac-tion is performed using localized face components to im-prove the extraction accuracy. Experiments are conducted to compare the use of attribute feature information, derived from crowd workers, to face sketch information, drawn by expert artists. In addition to removing the dependence on expert artists, the proposed method complements sketch-based face recognition by allowing investigators to imme-diately search face repositories without the time delay that is incurred due to sketch generation. 1

    Unusual presentation of eosinophilic fasciitis: two case reports and a review of the literature

    <p>Abstract</p> <p>Introduction</p> <p>Eosinophilic fasciitis is an uncommon disorder with unknown etiology and a poorly understood pathogenesis. We present the cases of two patients with eosinophilic fasciitis with unusual presentation, and describe the clinical characteristics and laboratory findings related to them.</p> <p>Case presentation</p> <p>The first case involves a 29-year-old Turkish man admitted with pain, edema and induration of his right-upper and left-lower limbs. Unilateral edema and stiffness with prominent pretibial edema was noted upon physical examination. A high eosinophil count was found on the peripheral smear. The second case involves a 63-year-old Turkish man who had pain, edema, erythema, and itching on his upper and lower extremities, which developed after strenuous physical activity. He had cervical lymphadenopathy and polyarthritis upon physical examination, and rheumatoid factor and antinuclear antibody upon laboratory examination.</p> <p>Conclusion</p> <p>Eosinophilic fasciitis can present with various symptoms. When patients exhibit eosinophilia, arthralgia and myalgia, eosinophilic fasciitis should be considered as a possible diagnosis.</p

    Expression of miR-21 and its targets (PTEN, PDCD4, TM1) in flat epithelial atypia of the breast in relation to ductal carcinoma in situ and invasive carcinoma

    <p>Abstract</p> <p>Background</p> <p>Flat epithelial atypia (FEA) of the breast is characterised by a few layers of mildly atypical luminal epithelial cells. Genetic changes found in ductal carcinoma in situ (DCIS) and invasive ductal breast cancer (IDC) are also found in FEA, albeit at a lower concentration. So far, miRNA expression changes associated with invasive breast cancer, like miR-21, have not been studied in FEA.</p> <p>Methods</p> <p>We performed miRNA in-situ hybridization (ISH) on 15 cases with simultaneous presence of normal breast tissue, FEA and/or DCIS and 17 additional cases with IDC. Expression of the miR-21 targets PDCD4, TM1 and PTEN was investigated by immunohistochemistry.</p> <p>Results</p> <p>Two out of fifteen cases showed positive staining for miR-21 in normal breast ductal epithelium, seven out of fifteen cases were positive in the FEA component and nine out of twelve cases were positive in the DCIS component. A positive staining of miR-21 was observed in 15 of 17 IDC cases. In 12 cases all three components were present in one tissue block and an increase of miR-21 from normal breast to FEA and to DCIS was observed in five cases. In three cases the FEA component was negative, whereas the DCIS component was positive for miR-21. In three other cases, normal, FEA and DCIS components were negative for miR-21 and in the last case all three components were positive. Overall we observed a gradual increase in percentage of miR-21 positive cases from normal, to FEA, DCIS and IDC. Immunohistochemical staining for PTEN revealed no obvious changes in staining intensities in normal, FEA, DCIS and IDC. Cytoplasmic staining of PDCD4 increased from normal to IDC, whereas, the nuclear staining decreased. TM1 staining decreased from positive in normal breast to negative in most DCIS and IDC cases. In FEA, the staining pattern for TM1 was similar to normal breast tissue.</p> <p>Conclusion</p> <p>Upregulation of miR-21 from normal ductal epithelial cells of the breast to FEA, DCIS and IDC parallels morphologically defined carcinogenesis. No clear relation was observed between the staining pattern of miR-21 and its previously reported target genes.</p

    Introduction: Hand hygiene is one of the most effective infection control measures to prevent the spread of healthcare-associated infections (HCAI). Water, soap, paper towel and hand disinfectant must be available and adequate in terms of effective hand hygiene. The adequacy of hand hygiene products or keeping water-soap and paper towel is still a problem for many developing countries like Turkey. In this multicenter study, we analyzed the adequacy in number and availability of hand hygiene products.Materials and Methods: This study was performed in all intensive care units (ICUs) of 41 hospitals (27 tertiary-care educational, 10 state and four private hospitals) from 22 cities located in seven geographical regions of Turkey. We analyzed water, soap, paper towel and alcohol-based hand disinfectant adequacy on four different days, two of which were in summer during the vacation time (August, 27th and 31st 2016) and two in autumn (October, 12th and 15th 2016).Results: The total number of ICUs and intensive care beds in 41 participating centers were 214 and 2357, respectively. Overall, there was no soap in 3-11% of sinks and no paper towel in 10-18% of sinks while there was no alcohol-based hand disinfectant in 1-4.7% of hand disinfectant units on the observation days. When we compared the number of sinks with soap and/or paper towel on weekdays vs. weekends, there was no significant difference in summer. However, on autumn weekdays, the number of sinks with soap and paper towel was significantly lower on weekend days (p<0.0001, p<0.0001) while the number of hand disinfectant units with alcohol-based disinfectant was significantly higher (p<0.0001).Conclusion: There should be adequate and accessible hand hygiene materials for effective hand hygiene. In this study, we found that soap and paper towels were inadequate on the observation days in 3-11% and 10-18% of units, respectively. Attention should be paid on soap and paper towel supply at weekends as well

    Wogonin and related natural flavones are inhibitors of CDK9 that induce apoptosis in cancer cells by transcriptional suppression of Mcl-1

    The wogonin-containing herb Scutellaria baicalensis has successfully been used for curing various diseases in traditional Chinese medicine. Wogonin has been shown to induce apoptosis in different cancer cells and to suppress growth of human cancer xenografts in vivo. However, its direct targets remain unknown. In this study, we demonstrate for the first time that wogonin and structurally related natural flavones, for example, apigenin, chrysin and luteolin, are inhibitors of cyclin-dependent kinase 9 (CDK9) and block phosphorylation of the carboxy-terminal domain of RNA polymerase II at Ser2. This effect leads to reduced RNA synthesis and subsequently rapid downregulation of the short-lived anti-apoptotic protein myeloid cell leukemia 1 (Mcl-1) resulting in apoptosis induction in cancer cells. We show that genetic inhibition of Mcl-1 or CDK9 expression by siRNA is sufficient to mimic flavone-induced apoptosis. Pull-down and in silico docking studies demonstrate that wogonin directly binds to CDK9, presumably to the ATP-binding pocket. In contrast, wogonin does not inhibit CDK2, CDK4 and CDK6 at doses that inhibit CDK9 activity. Furthermore, we show that wogonin preferentially inhibits CDK9 in malignant compared with normal lymphocytes. Thus, our study reveals a new mechanism of anti-cancer action of natural flavones and supports CDK9 as a therapeutic target in oncology

    Epidermal growth factor regulates Mcl-1 expression through the MAPK-Elk-1 signalling pathway contributing to cell survival in breast cancer

    Myeloid cell leukaemia-1 (Mcl-1) is an anti-apoptotic member of the Bcl-2 family that is elevated in a variety of tumour types including breast cancer. In breast tumours, increased Mcl-1 expression correlates with high tumour grade and poor patient survival. We have previously demonstrated that Her-2 levels correspond to increased Mcl-1 expression in breast tumours. Epidermal growth factor (EGF) receptor signalling is frequently deregulated in breast cancer and leads to increased proliferation and survival. Herein, we determined the critical downstream signals responsible for the EGF mediated increase of Mcl-1 and their role in cell survival. We found that both Mcl-1 mRNA and protein levels are rapidly induced upon stimulation with EGF. Promoter analysis revealed that an Elk-1 transcription factor-binding site is critical for EGF activation of the Mcl-1 promoter. Furthermore, we found that knockdown of Elk-1or inhibition of the Erk signalling pathway was sufficient to block EGF upregulation of Mcl-1 and EGF mediated cell survival. Using chromatin immunoprecipitation and biotin labelled probes of the Mcl-1 promoter, we found that Elk-1 and serum response factor are bound to the promoter after EGF stimulation. To determine whether Mcl-1 confers a survival advantage, we found that knockdown of Mcl-1 expression increased apoptosis whereas overexpression of Mcl-1 inhibited drug induced cell death. In human breast tumours, we found a correlation between phosphorylated Elk-1 and Mcl-1 protein levels. These results indicate that the EGF induced activation of Elk-1 is an important mediator of Mcl-1 expression and cell survival and therefore a potential therapeutic target in breast cancer

    Cervical epithelial damage promotes Ureaplasma parvum ascending infection, intrauterine inflammation and preterm birth induction in mice

    Around 40% of preterm births are attributed to ascending intrauterine infection, and Ureaplasma parvum (UP) is commonly isolated in these cases. Here we present a mouse model of ascending UP infection that resembles human disease, using vaginal inoculation combined with mild cervical injury induced by a common spermicide (Nonoxynol-9, as a surrogate for any mechanism of cervical epithelial damage). We measure bacterial load in a non-invasive manner using a luciferase-expressing UP strain, and post-mortem by qPCR and bacterial titration. Cervical exposure to Nonoxynol-9, 24 h pre-inoculation, facilitates intrauterine UP infection, upregulates pro-inflammatory cytokines, and increases preterm birth rates from 13 to 28%. Our results highlight the crucial role of the cervical epithelium as a barrier against ascending infection. In addition, we expect the mouse model will facilitate further research on the potential links between UP infection and preterm birth
