    Traffic in Towns, the Loss ofg Urban Resilence and the case of Auckland's civic centre

    There are cogent arguments supporting the idea that resilient urbanism requires successful streets. Successful streets in city centres require a balance between efficient traffic movement and spaces for pedestrians on which urban vitality and economies depend. This balance was fractured in the 1940’s with the growth of car ownership, and traffic solutions prioritising vehicle movement. Responding to these issues in 1963, the Buchanan Report, Traffic in Towns advocated building motorways in towns, but in such a way that these circled what were called ‘environmental areas’. Auckland enthusiastically embraced motorway construction from 1955, and proposals to build a new civic centre at this time were seen as an opportunity to improved traffic flow in the inner city. This included the insertion of a new circular street, Mayoral Drive, cutting across the previous small scale grain of blocks and streets. The success of this street 50 years since its construction is assessed using urban design criteria. The conclusion drawn is that apart from two small areas, Mayoral Drive remains a largely unsuccessful street at the heart of Auckland, with a configuration that remains difficult to remediate from both a private and public investment point of view

    An empirical approach for semantic Web services discovery

    Component retrieval/discovery is a well-established research direction in Software Engineering. With the surge of Service-Oriented Architecture (SOA), service discovery has become increasingly crucial. However, the public UDDI Business Registry the primary service discovery mechanism over the Internet has been shut down permanently since 2006. Moreover, keyword-based service discovery is insufficient in coping with complex discovery requirements posed by modern software developers. In this paper, we propose an empirical semantic based Web service discovery approach. It provides an automatic Web service discovery mechanism that can locate relevant Web services based on concepts rather than keywords. The major contribution of this paper is three fold. First we articulate three requirements that software developers often raise during the component/service development and discovery process. Next, we propose the application of Latent Semantic Analysis into the area of Web services discovery. To our best knowledge, little work has been done in this area which leverages concept-based Information Retrieval models in service discovery. Last, we provide a proof-of-concept system prototype that can suffice three specific requirements of semantic service discovery

    The bacterial strain TR3.2T was isolated from aerobic bioreactor-treated soil from a polycyclic aromatic hydrocarbon (PAH)-contaminated site in Salisbury, NC, USA. Strain TR3.2T was identified as a member of ‘Pyrene Group 2’ or ‘PG2’, a previously uncultivated cluster of organisms associated with the degradation of high-molecular-weight PAHs by stable-isotope probing. Based on its 16S rRNA gene sequence, the strain was classified as a member of the class Gammaproteobacteria but possessed only 90.5 % gene identity to its closest described relative, Methylococcus capsulatus strain Bath. Strain TR3.2T grew on the PAHs pyrene, phenanthrene, anthracene, benz[a]anthracene and fluorene, as well as the azaarene carbazole, and could additionally metabolize a limited number of organic acids. Optimal growth occurred aerobically under mesophilic temperature, neutral pH and low salinity conditions. Strain TR3.2T was catalase and oxidase positive. Predominant fatty acids were C17 : 0 cyclo and C16 : 0. Genomic G+C content of the single chromosome was 67.79 mol% as determined by complete genome sequencing. Due to the high sequence divergence from any cultivated species and its unique physiological properties compared to its closest relatives, strain TR3.2T is proposed as a representative of a novel order, family, genus and species within the class Gammaproteobacteria, for which the name Immundisolibacter cernigliae gen. nov., sp. nov. is proposed. The associated order and family are therefore proposed as Immundisolibacteralesord. nov. and Immundisolibacteraceaefam. nov. The type strain of the species is TR3.2T (=ATCC TSD-58T=DSM 103040T)

    Rugosibacter aromaticivorans gen. nov., sp. nov., a bacterium within the family Rhodocyclaceae, isolated from contaminated soil, capable of degrading aromatic compounds

    A bacterial strain designated Ca6T was isolated from polycyclic aromatic hydrocarbon (PAH)-contaminated soil from the site of a former manufactured gas plant in Charlotte, NC, USA, and linked phylogenetically to the family Rhodocyclaceae of the class Betaproteobacteria. Its 16S rRNA gene sequence was highly similar to globally distributed environmental sequences, including those previously designated ‘Pyrene Group 1’ demonstrated to grow on the PAHs phenanthrene and pyrene by stable-isotope probing. The most closely related described relative was Sulfuritalea hydrogenivorans strain sk43HT (93.6 % 16S rRNA gene sequence identity). In addition to a limited number of organic acids, Ca6T was capable of growth on the monoaromatic compounds benzene and toluene, and the azaarene carbazole, as sole sources of carbon and energy. Growth on the PAHs phenanthrene and pyrene was also confirmed. Optimal growth was observed aerobically under mesophilic temperature, neutral pH and low salinity conditions. Major fatty acids present included summed feature 3 (C16 : 1ω7c or C16 : 1ω6c) and C16 : 0. The DNA G+C content of the single chromosome was 55.14 mol% as determined by complete genome sequencing. Due to its distinct genetic and physiological properties, strain Ca6T is proposed as a member of a novel genus and species within the family Rhodocyclaceae, for which the name Rugosibacter aromaticivorans gen. nov., sp. nov. is proposed. The type strain of the species is Ca6T (=ATCC TSD-59T=DSM 103039T)

    Molecular diagnostics of banana Fusarium wilt targeting Secreted-in-Xylem genes

    Fusarium wilt is currently spreading in banana growing regions around the world leading to substantial losses. The disease is caused by the fungus Fusarium oxysporum f. sp. cubense (Foc), which is further classified into distinct races according to the banana varieties that they infect. Cavendish banana is resistant to Foc race 1, to which the popular Gros Michel variety succumbed last century. Cavendish effectively saved the banana industry, and became the most cultivated commercial variety worldwide. However, Foc tropical race 4 (TR4) subsequently emerged in Southeast Asia, causing significant yield losses due to its high level of aggressiveness to Cavendish and other commonly grown varieties. Preventing further spread is crucially important in the absence of effective control methods or resistant market-acceptable banana varieties. Implementation of quarantine and containment measures depends on early detection of the pathogen through reliable diagnostics. In this study, we tested the hypothesis that secreted in xylem (SIX) genes, which currently comprise the only known family of effectors in F. oxysporum, contain polymorphisms to allow the design of molecular diagnostic assays that distinguish races and relevant VCGs of Foc. We present specific and reproducible diagnostic assays based on conventional PCR targeting SIX genes, using as templates DNA extracted from pure Foc cultures. Sets of primers specifically amplify regions of: SIX6 in Foc race 1, SIX1 gene in TR4, SIX8 in subtropical race 4, SIX9/SIX10 in Foc VCG 0121, and SIX13 in Foc VCG 0122. These assays include simplex and duplex PCRs, with additional restriction digestion steps applied to amplification products of genes SIX1 and SIX13. Assay validations were conducted to a high international standard including the use of 250 Fusarium spp. isolates representing 16 distinct Fusarium species, 59 isolates of F. oxysporum, and 21 different vegetative compatibility groups (VCGs). Tested parameters included inter and intraspecific analytical specificity, sensitivity, robustness, repeatability and reproducibility. The resulting suite of assays is able to reliably and accurately detect R1, STR4, TR4 as well as two VCGs (0121 and 0122) causing Fusarium wilt in bananas

    AbstractThese data document the vitality of mature mouse oocytes (Metaphase II (MII)) and early stage embryos (zygotes) following exposure to the genotoxic chemotherapeutic agent, etoposide, in combination with PSC833, a selective inhibitor of permeability glycoprotein. They also illustrate the vitality of parthenogenetically activated and fertilized embryos following incubation with the calcium chelator BAPTA-AM (1,2-Bis(2-aminophenoxy)ethane- N,N,N′,N′-tetraacetic acid tetrakis (acetoxymethyl ester)), cycloheximide (an antibiotic that is capable of inhibiting protein synthesis), and hydrogen peroxide (a potent reactive oxygen species). Finally, they present evidence that permeability glycoprotein is not represented in the proteome of mouse spermatozoa. Our interpretation and discussion of these data feature in the article “Identification of a key role for permeability glycoprotein in enhancing the cellular defense mechanisms of fertilized oocytes” (Martin et al., in press) [1]
