    Multiphase gas mechanisms in groundwater

    Numerous groundwater springs bubble, yet the flow and transport of gases are not well understood in hydrogeology. An understanding of the processes by which gases enter, migrate, and are liberated from groundwater is required. A quantitative conceptual model of gas migration in groundwater will allow an understanding of what informative aquifer signals may be present in gas data, as well as what information signals may be masked or diminished by phase changes. Through analysis of existing published literature, seven facies of groundwater bubbles were developed to provide a framework for research in these specific categories of gas transport. In order to better understand these multiphase bubbling springs, an instrument was designed and deployed over the discharge of a spring in southern Oklahoma that measured the total gas flux, ebullative and diffusive. By measuring the water discharge from the spring too, a hydropneumograph of gas and water mass flux over time can be produced. In addition to the mass flow rates of the two phases provided by the hydropneumograph, water and gas samples were collected for compositional analysis. By combining the compositional data of exsolved and dissolved gas with the mass flow rates from the hydropneumograph, estimation of the quantity of light noble gases is radically changed (60% for He, 45% for Ne) which provides improvements in the calculation of recharge temperature of 4 to 25% depending on the model selected. These improvements in the understanding of the physical hydrogeology of bubbling springs provide an additional avenue for researchers to explore aquifer dynamics that is largely ignored in the extant literature

    Designer Reagents for Mass Spectrometry-Based Proteomics: Clickable Cross-Linkers for Elucidation of Protein Structures and Interactions

    We present novel homobifunctional amine-reactive clickable cross-linkers (CXLs) for investigation of three-dimensional protein structures and protein–protein interactions (PPIs). CXLs afford consolidated advantages not previously available in a simple cross-linker, including (1) their small size and cationic nature at physiological pH, resulting in good water solubility and cell-permeability, (2) an alkyne group for bio-orthogonal conjugation to affinity tags via the click reaction for enrichment of cross-linked peptides, (3) a nucleophilic displacement reaction involving the 1,2,3-triazole ring formed in the click reaction, yielding a lock-mass reporter ion for only clicked peptides, and (4) higher charge states of cross-linked peptides in the gas-phase for augmented electron transfer dissociation (ETD) yields. Ubiquitin, a lysine-abundant protein, is used as a model system to demonstrate structural studies using CXLs. To validate the sensitivity of our approach, biotin-azide labeling and subsequent enrichment of cross-linked peptides are performed for cross-linked ubiquitin digests mixed with yeast cell lysates. Cross-linked peptides are detected and identified by collision induced dissociation (CID) and ETD with linear quadrupole ion trap (LTQ)-Fourier transform ion cyclotron resonance (FTICR) and LTQ-Orbitrap mass spectrometers. The application of CXLs to more complex systems (e.g., in vivo cross-linking) is illustrated by Western blot detection of Cul1 complexes including known binders, Cand1 and Skp2, in HEK 293 cells, confirming good water solubility and cell-permeability

    Entangled Bessel beams

    Orbital angular momentum (OAM) entanglement is investigated in the Bessel-Gauss (BG) basis. Having a readily adjustable radial scale, BG modes provide a more favourable basis for OAM entanglement over Laguerre-Gaussian (LG) modes. The OAM bandwidth in terms of BG modes can be increased by selection of particular radial modes and leads to a flattening of the spectrum. The flattening of the spectrum allows for higher entanglement. We demonstrate increased entanglement in terms of BG modes by performing a Bell-type experiment and violating the appropriate Clauser Horne Shimony Holt (CHSH) inequality. In addition, we reconstruct the quantum state of BG modes entangled in high-dimensions.Comment: 8 pages, 4 figure

    Face-To-Face With Scorching Wildfire: Potential toxicant Exposure and the Health Risks of Smoke For Wildland Firefighters at the Wildland-Urban interface

    As wildfire risks have elevated due to climate change, the health risks that toxicants from fire smoke pose to wildland firefighters have been exacerbated. Recently, the International Agency for Research on Cancer (IARC) has reclassified wildland firefighters\u27 occupational exposure as carcinogenic to humans (Group 1). Wildfire smoke contributes to an increased risk of cancer and cardiovascular disease, yet wildland firefighters have inadequate respiratory protection. The economic cost of wildland fires has risen concurrently, as illustrated by the appropriation of $45 billion for wildfire management over FYs 2011-2020 by the U.S. Congress. Occupational epidemiological studies of wildland firefighters are crucial for minimizing health risks; however, they must account for the mixture of exposures in wildfire smoke. This review focuses on four aspects of wildland firefighters\u27 health risks at the wildland-urban interface: 1) economic costs and health impact, 2) respiratory protection, 3) multipollutant mixtures, and 4) proactive management of wildfires
