    Relaxant Activity of the Methanol Extract of Acanthus Montanus (Nees) T Anderson (Acanthaceae) on Isolated Guinea Pig Trachea

    Purpose: To evaluate the effects of the methanol extract of Acanthus montanus on guinea pig trachealis muscle.Methods: Guinea pig tracheae were set up in 10 ml organ baths. The effect of the methanol extract of Acanthus montanus (0.5 - 8 mg/ml) on the spontaneous tone of the trachea and carbacholprecontracted trachea in the absence and presence of propranolol, glibenclamide and procaine wasstudied.Results: The extract (0.5 ÂĄV 8.0 mg/ml) produced a concentration-dependent relaxation of the intrinsic tone in tracheal preparations which was completely blocked by propranolol (3 x 10-7 M), glibenclamide(10-7 M) and procaine (10-3 M). The extract (0.5 - 8 mg/ml) produced a concentration-dependent relaxation of carbachol (10-5M)-precontracted trachea. This effect of the extract was partially blocked by propranolol (3 x 10-7M), procaine (10-3M) and glibenclamide (10-7M) with a progressive increase in the median effective concentration (EC50) values as follows: control, 0.66mg/ml > propranolol, 1.42mg/ml > glibenclamide, 1.54mg/ml > procaine, 2.04 mg/ml.Conclusion: The results obtained suggest that the extract produces a non-specific smooth muscle relaxant effect mediated via Æ’Ă’-adrenergic receptor mechanism or potassium channels.Keywords: Acanthus montanus, Trachea, Carbachol, Potassium channels, Propranolol, Glibenclamide, Procaine

    Significance of IgG-Avidity in Antenatal Rubella Diagnosis.

    OBJECTIVE: A descriptive study was carried out to determine the significance of IgG-affinity in the serological diagnosis of rubella infections in pregnancy. MATERIALS AND METHODS: A total of 92 pregnant women who had never received antirubella vaccines were recruited by simple random selection and did not exceed 24 weeks of gestation were recruited from the antenatal clinics of the University of Ilorin Teaching Hospital. Rubella virus-specific IgG, IgG-affinity and IgM were tested, using the Indirect ELISA methods. RESULTS: IgG-Affinity tests showed that 2 (2.2%) out of the 92 pregnant women, who were in their first and second trimester pregnancies respectively, had primary Rubella infections, while 1 (1.1%) primigravidae had a re-infection with rubella virus. It was also discovered that out of the 13 multigravid subjects that reported to have lost previous pregnancies, 2 (15.4%) cases may have been due to rubella infections that occurred during organogenesis. CONCLUSION: Although the isolation of the whole virus or the viral nucleic material is the best basis for diagnosis, IgG-affinity is a proven supplementary serological diagnosis, to distinguish reinfection or viral persistence from primary exposure for prompt and accurate diagnosis. This is necessary for proper counselling of pregnant women especially in low economies where molecular diagnosis may not be affordable

    ASSESSMENT OF WORK-SPACE AND WORK-METHOD DESIGNS IN NIGERIA AUTOMOBILE SERVICE AND REPAIR INDUSTRY

    Nigeria automobile service industry is labour intensive with high level of work-related injuries among its workforce. This research assessed work-space (WsD) and work-method designs (WmD), level of compliance with recommended standards (RSs) and effects on workers’ wellbeing. Clearances for services in 55 supine positions and 45 stand-up workbenches’ (SUWb) heights were measured and compared with RSs. Questionnaires were completed among 252 workers to measure prevalence of work-related injuries. Descriptive statistical procedure and Chi-Square tests were conducted using SPSS. 94.4% of workers suffered from low back, 43.5% neck and 40% knees pains among others. Averagely, 43.33±7.3cm and 67±0.15cm were measured for supine position clearance and  SUWb heights with 15.03±14.3 and 26.55±16.23 percentage deviations respectively from the minimum RSs. Automotive mechanics may be prone to back, hand and/or wrist related problems and Cumulative Trauma Disorders.     The Novel Antihyperglycaemic Action of Hunteria umbellate Seed Fractions Mediated Via Intestinal Glucose Uptake Inhibition

    The present study evaluated the antihyperglycaemic effect and mechanism of action of fractions of the aqueous seed extract of Hunteria umbellata (K. Schum.) Hallier f. (HU) in normal and alloxan-induced hyperglycaemic rats. HU was partitioned in chloroform, acetyl acetate and butan-1-ol to give chloroform fraction (HUc), ethyl acetate fraction (HUe), butanol fraction (HUb) and the “residue” (HUm), respectively. 200 mg/kg of each of these fraction dissolved in 5% Tween 20 in distilled water was investigated for its acute oral hypoglycaemic effects in normal rats over 6 hours while its repeated dose antihyperglycaemic effect was evaluated in alloxan-induced hyperglycaemic rats over 5 days. In addition, 50 mg/kg of the crude alkaloid fraction (HUAf) extracted from HU was evaluated for its possible antihyperglycaemic activity in alloxaninduced hyperglycaemic rats using oral glucose tolerance test (OGTT) over 6 hours. Using the solvent system, distilled water-butanol-ammonium hydroxide (2:15:1, v/v/v), HUb was chromatographed and stained with Dragendorff’s reagent for confirmatory qualitative analysis for alkaloids. Results showed that oral pre-treatment with 200 mg/kg of HUe, HUb and HUm resulted in a significant (p<0.05, p<0.001) time dependent hypoglycaemic effect, with the butan-1-ol fraction HU causing the most significant (p<0.001) hypoglycaemic effect. In the alloxan-induced hyperglycaemic rats,  repeated oral treatment with 200 mg/kg of same HU fractions for 5 days resulted in significant (p<0.05) decreases in the fasting blood glucose concentrations with the most significant (p<0.01) antihyperglycaemic effect also recorded for HUb. Similarly, oral pretreatment with 50 mg/kg of HUAf significantly (p<0.05, p<0.01 and p<0.001) attenuated an increase in the post-absorptive glucose concentration at 1st -6th h in the alloxan-induced hyperglycaemic OGTT model. In addition, alkaloid was present in most of the separated spots on the TLC plate. In conclusion, results of this study showed that HU contains a relative high amount of alkaloids which could have accounted for the antihyperglycaemic action of HU that was mediated via intestinal glucose uptake inhibition.Key words: Hunteria umbellata aqueous seed extract, Alkaloid fraction, Intestinal glucose uptake inhibition, Normal and alloxan-induced hyperglycaemic rat

    Prescribing Pattern of Non-Steroidal Ant­‐inflammatory Drugs at the Outpatient Pharmacy Department of Lagos University Teaching Hospital, Nigeria

    Background: Non-steroidal Anti- inflammatory Drugs (NSAID), are among the most widely used and misused of all drugs. Though they provide symptomatic relief from pain and swelling in chronic joint diseases, they may cause renal impairment, especially in combination with other nephrotoxic agents.Objectives: This study aimed to investigate the prescription pattern of NSAID in the Out-patient Pharmacy Department of Lagos University Teaching Hospital (LUTH), Nigeria.Design: A total of 3800 prescriptions containing NSAIDs were analyzed for information on drug name, the number of NSAIDs per prescription, the presence of ACE inhibitors and diuretics alongside NSAIDs and NSAIDs prescribed in generic or brand names.Results: The results showed that Aspirin was the most frequently prescribed NSAID (62.2%) and 68.4% of the NSAIDs prescriptions studied were written in generic names. The total number of drugs per prescription was in most cases 3 or greater (84.6%). There were statistically significant (p ≀ 0.05) associations between the individual NSAID prescribed and whether they were prescribed in generics or brand names; individual NSAID prescribed and the frequency of co-prescription with an ACE inhibitor and a diuretic; types of NSAID prescribed and the cost in Naira.Conclusion: Though most of the prescribers complied with WHO standard in their prescriptions vis a vis generic prescription, avoidance of polypharmacy and avoidance of drug interactions and contraindications, there is obvious need for interventional measures or strategies to improve rational prescribing for some of the prescribers tailored towards rational prescription and use of drugsKeywords: NSAID, Prescription, Outpatient, Adverse reaction, Aspirin

    Polycyclic Aromatic Hydrocarbon, Haematological and Oxidative Stress Levels in Commercial Photocopier Operators In Lagos, Nigeria

    Background: Human exposure to hazardous substances in the environment has been known to play an important role in the pathogenesis of some diseases. Photocopying machines have become a cheap source of self-employment in Nigeria. For obvious reasons the highest level of patronage is encountered in the campuses of educational institutions. However, the persons who operate the machines are always exposed to possible hazards associated with the job without protective devices.Objective: This study investigated the levels of oxidative stress, polycyclic aromatic hydrocarbon (PAH) and haematological parameters in blood samples of photocopier operators.Methods: The experimental procedure involved 50 consented subjects selected based on some criteria. The haematological parameters, oxidative stress and PAH levels were determined using standard methods.Results: The results showed no significant difference (p ≄ 0.05) in the haematological parameters between the test subjects and the controls. However, there were duration on the job (yrs) dependent significant decrease in the level of superoxide dismutase (SOD) of the photocopier operators compared with the controls (> 5 years p≀ 0.0001; 4-5 years p≀0.001). The level of reduced glutathione (GSH) was significantly decreased across all lengths of duration on the job compared with the controls.Conclusion: The findings in this study revealed increased level of oxidative stress in photocopier operators with no significant change in haematological parameters. The health implication of operating photocopiers call for quick health education and intervention tailored to monitoring and guiding the photocopier operators. This will help to prevent or manage continuous exposure to the hazards of photocopying machines.Keywords: Photocopier operators, oxidative stress, PAH, haematology, Nigeria

    Premature rupture of membranes at term: immediate induction of labor versus expectant management

    Objective: To compare the maternal outcomes of immediate induction of labor with expectant management in women presenting with premature rupture of membranes (PROM) at term.Methods: One hundred and fifty two women with PROM at term were randomized into either immediate induction of labor with oxytocin or expectant management for a period of 12 hours. The primary outcome measure was the incidence of clinical endometritis in each group. Secondary outcomes were the mode of delivery, the neonatal outcome and the proportion of women in the expectant management group that progressed to spontaneous labor.Results: The immediate induction arm had a lower caesarean section rate, (7.9% vs 28.9%, P=0.001), higher spontaneous vaginal delivery rate (92.1% vs 71.1%; P=0.001) and lower incidence of clinical endometritis (0% vs 5.3%, P=0.006), when compared with the expectant management arm. The estimated duration of labor was shorter in the expectant management arm (8.9±2.17hours vs 10.6±2.35hours; P=<0.001). Neonatal morbidity rates were comparable in both groups.Conclusion: Immediate induction of labor in women with PROM at term resulted in significantly lower rate of infectious morbidity without increasing the risk of operative delivery.     Gastroprotective Effects of DAS-77 (a Phytomedicine) in Ulcer Models in Rats

    Purpose: DAS-77 is a phytomedicine that contains the dried bark of Mangifera indica and root of Carica papaya. This study investigated the antiulcer effects of DAS-77 in rats.Methods: DAS-77 was administered orally twice daily for five consecutive days at doses of 50 - 400 mg/kg. Ulcer was induced in rats with ethanol, indomethacin, pylorus ligation (PL) and cold restraint stress (CRS). Ulcer scores were recorded based on examination of excised stomachs. Estimations of gastric content volume, pH and titratable acidity in the PL model and determination of the levels of antioxidants and malondialdehyde (MDA) in gastric tissues in the CRS model were also done.Results: In all the models, DAS-77 produced significant dose-dependent reductions in ulcer score. Peak effects were produced at the dose of 400 mg/kg with ulcer inhibition values of 98.57, 76.23, 99.28 and 96.70 % compared to 100.00, 93.79, 98.92 and 96.79 % for  misoprostol/cimetidine, respectively, for the ethanol, indomethacin, PL and CRS models. In the PL model, DAS-77 caused a significant increasein pH of gastric content but a reduction in volume and titratable acidity. At doses of 50 and 100 mg/kg in the CRS model, DAS-77 significantly increased the level of reduced glutathione (GSH) and diminishedMDA.Conclusion: The results obtained in this study suggest that DAS-77 possesses gastroprotective activity possibly due to reduced gastric secretion and acidity, and antioxidant activity.Keywords: DAS-77, Phytomedicine, Mangifera indica, Carica papaya, Gastroprotective effects, Ulcer

    Performance and carcass characteristics of Yankasa ram fed with variable levels of biscuit waste and Leucaena leucocephala based diets

    A study was conducted to find out the performance and carcass characteristics of sheep fed diet in which biscuits waste (BWM) and Leucaena leucocephala leaf hay (L/h) mixture were used to replace maize and wheat offal mixture at zero (control), 25, 50, 70 and 100% replacement levels. In a completely randomize design, the experimental diet where designated B0, B1, B2, B3, B4, respectively. The 20 growing Yankasa rams aged between 9 to 11 months with an initial average life weight of 12.17 ± 0.33 kg were randomly assign to the 5 dietary treatment with four rams per treatment. Data were collected to estimate dry matter intake (g/d /W0.7 kg), daily mean life weight gain (g/d), feed efficiency and carcass indices. Dietary replacement of maize and wheat offal mixture with biscuit waste meal and L. leucocelphala leaf hay mixture significantly (P< 0.05) influenced the entire variable measured. However, animal on diet B1 and B2 in which biscuits waste and L. leucocephala leaf mixture replaced 25 and 50% maize and wheat offal mixture, were better when compared to the control (B0) and other test diet in terms of performance and carcass characteristics. This was evident by higher DMI (77.18 and 76.84 g/d /W0.75 kg), ADWG (134.40 and 129.52 g/d), feed efficiency (0.199 and 0.195), live shrunk weight (21.50 and 20.10 kg), dressing percentage (85.81 and 84.33%), chilled carcass weight (18.33 and 16.83 kg), wholesale cuts leg (9.18 and 8.73%), rack (3.11 and 2.78%), BSF (4.53 and 3.83%), neck added (2.51 and 1.78%) and rib eye area (6.06 and 8.29) which were obtained from the animal on these two diets. Broadly, the result in the present study indicated that 25 and 75% replacement level of maize and wheat offal mixture with biscuits waste and L. leucocephala leaf hay mixture are ideal for ruminant animal production because it improve both the performance and carcass characteristics of rams.Key words: Performance, carcass, biscuit waste, Yankasa ram

    Effect of Moringa oleifera marinade on microbial stability of smoke-dried African Catfish (Clarias gariepinus)

    The study examined the antimicrobial effect of Moringa oleifera marinade on smoke-dried catfish stored at ambient temperature (37±20C) for two months. The experimental treatments are the control, 1%, 2% and 3% (w/v) Moringa oleifera Marinade (MOM) and 5% Brine (w/v) solutions. Seventy-five fishes of average weight of 260±8g were gutted, washed and randomly assigned to the treatments. Thereafter, the fishes were soaked in the treatments for 2 hours and later hot smoked for 12hours. After smoking, the fishes were stored in netted boxes and placed on laboratory shelves for two months. Microbial counts were conducted at 7-day interval while biochemical tests were conducted on the 8th week. Seven bacterial species namely; Staphylococcus sp, Bacillus sp, Klebsiella sp, Corynebacterium sp, Pseudomonas sp, Escherichia coli and streptococcus sp and six fungal species namely; Penicillium italicum, Cladosporium sp, Neurospora crassa, Candida sp, Aspergillus niger and Saccharomyces cerevisiae were observed in the study. There was a general increase in microbial load as storage progressed. However, the increment was pronounced in the control and brine treated fish samples. In all levels of MOM and 5% Brine, there was decrease in the bacterial and fungal counts as compared with the control samples. 3% MOM exhibited the highest antibacterial potency while 5% Brine exhibited the highest antifungal potency. Moringa oleifera marinade could be use to protect stored smoke-dried catfish from microbial spoilage thus limiting economic loss and possible heath risk to consumers.Key words: Moringa, marinade, bacteria, fungi, catfish, smoke-drie
