Using the cytokinesis-block micronucleus cytome assay to evaluate Chromosomal DNA damage in chronic renal patients undergoing Bicarbonate Haemodialysis and Haemodiafiltration
Introduction. Chronic Renal Failure (CRF) patients are considered to show genomic instability and are associated with a high risk of both cardiovascular diseases and cancer. We explored DNA damage due to two dialysis treatments in 20 patients undergoing bicarbonate haemodialysis, 20 undergoing haemodiafiltration and 40 healthy subjects.Methods. The cytokinesis-block micronucleus (MN) assay was performed on peripheral blood lymphocytes to evaluate genetic damage.Results. A higher frequency of MN in the dialysis groups compared with controls was found. The results do not show a relationship between genetic instability and the type, frequency and duration of haemodialysis. The average BD and HDF treatment time was respectively 3.8±6.3 and 3.7±3.9 yrs. CAT and scintigraphy was independently correlated with high levels of MN.Discussion. Overall, the frequency of MN in CRF patients undergoing dialysis therapy was observed to be higher. Further studies need to be performed on a larger number of patients and for a longer period
Skin Safety and Health Prevention: an Overview of Chemicals in Cosmetic Products
Introduction. Cosmetic products contain a wide range of chemicals to which we are exposed every day. The aim of the study was to determine the presence of potential dangerous substances which can cause adverse health effects by examining product labels.Methods. A total of 283 products were collected from various shops in Lecce (Italy) and divided into 3 categories: rinse-off, leave-on and make-up. The label of every product was examined and a list including fragrances, preservatives and other chemicals of concern was created.Results. Fragrances were present in 52.3% of the examined products, mostly limonene (76.9%) and linalool (64.6%) but also citronellol (34.1%), geraniol (31.5%), coumarin (30%) and hexyl cinnamal (29.2%). Preservatives showed a rate of 60% and the most frequently identified were phenoxyethanol (48.7%), sodium benzoate (35.6%), potassium sorbate (22%), methylparaben (15.2%) and MI/MCI (9.9%). The other chemicals of concern were detected in 58% of products; included PEGs (62.3%), acrylate copolymer (34%), petrolatum (17.2%), polysorbates (14,8%), BHT (14.7%), ethylhextyl methoxycinnamate (13.6%), benzophenone-1 (3.7%), benzophenone-3 (4.9%), BHA (1.6%), cocamide DEA and toluene (1.2%).Conclusions. The use of many of these substances is allowed within certain limits, due to their toxicity at higher concentrations. Other important aspects should be considered as, for instance, the possibility of long-term effects. On the other hand, other substances may induce several acute adverse side-effects, i.e. contact dermatitis and allergic reactions. For these reasons, an enhancement of the criteria used for cosmetics formulation is required since many chemicals used singularly or combined are potentially unsafe
Evaluation of Radon Exposure Risk and Lung Cancer Incidence/Mortality in South-Eastern Italy
Introduction: Radon and its decay products may cause substantial health damage after long-term exposure. The aim of the study was to perform a spatial analysis of radon concentration in the Salento peninsula, province of Lecce (South-eastern Italy) in order to better characterize possible risk for human health, with specific focus on lung cancer.Methods: Based on previous radon monitoring campaigns carried out in 2006 on behalf of the Local Health Authority (ASL Lecce) involving 419 schools and through the application of kriging estimation method, a radon risk map was obtained for the province of Lecce, in order to determine if areas with higher radon concentrations were overlapping with those characterized by the highest pulmonary cancer incidence and mortality rates.Results: According to our data, areas at higher radon concentrations seem to overlap with those characterized by the highest pulmonary cancer mortality and incidence rates, thus indicating that human exposure to radon could possibly enhance other individual or environmental pro-carcinogenic risk factors (i.e. cigarette smoking, air pollution and other exposures).Conclusions: The radon risk should be further assessed in the evaluation of the causes resulting in higher mortality and incidence rates for pulmonary cancer in Salento area vs Italian average national data. For these reasons, ASL Lecce in cooperation with ARPA Puglia and CNR-IFC has included the monitoring of individual indoor radon concentrations in the protocol of PROTOS case-control Study, aimed at investigating the role of different personal and environmental risk factors for lung cancer in Salento
Has VZV epidemiology changed in Italy? Results of a seroprevalence study
The aim of the study was to evaluate if and how varicella prevalence has changed in Italy. In particular a seroprevalence study was performed, comparing it to similar surveys conducted in pre-immunization era. During 2013–2014, sera obtained from blood samples taken for diagnostic purposes or routine investigations were collected in collaboration with at least one laboratory/center for each region, following the approval of the Ethics Committee. Data were stratified by sex and age. All samples were processed in a national reference laboratory by an immunoassay with high sensitivity and specificity. Statutory notifications, national hospital discharge database and mortality data related to VZV infection were analyzed as well. A total of 3707 sera were collected and tested. In the studied period both incidence and hospitalization rates decreased and about 5 deaths per year have been registered. The seroprevalence decreased in the first year of life in subjects passively protected by theirmother, followed by an increase in the following age classes. The overall antibody prevalence was 84%. The comparison with surveys conducted with the samemethodology in 1996–1997 and 2003–2004 showed significant differences in age groups 1–19 y. The study confirms that in Italy VZV infection typically occurs in children. The impact of varicella on Italian population is changing. The comparison between studies performed in different periods shows a significant increase of seropositivity in age class 1 – 4 years, expression of vaccine interventions already adopted in some regions
Risk Factors for Lung Cancer in the Province of Lecce: Results from the PROTOS Case–Control Study in Salento (Southern Italy)
In the province of Lecce (southern Italy), a higher incidence of lung cancer (LC) among men compared to regional and national data was reported. In a sub-area in the center of the province (cluster area), the incidence and mortality for LC was even higher. PROTOS is a case-control study aimed at investigating possible risk factors for LC in the province area. A total of 442 patients with LC and 1326 controls matched by sex and age living in the province of Lecce for at least 10 years were enrolled and georeferenced; they filled in a questionnaire with their personal information and exposures. For each risk factor, an Odds Ratio adjusted for all the other variables was calculated. The risk of LC increased with excessive use of alcohol in women, for those subjects with a family cancer history, for each increase in pack/year of cigarettes, for men more exposed considering the industrial district in the cluster area, and for those using pesticides in agriculture without wearing personal protective equipment. The higher incidence of adenocarcinoma in both sexes suggests that, in addition to cigarette smoking, concurrent exposures to other environmental, occupational, and life-style factors may play a role in increased cancer risk and should be more deeply explored
Emergence of unusual human rotavirus strains in Salento, Italy, during 2006–2007
<p>Abstract</p> <p>Background</p> <p>In recent years, rotavirus genotyping by RT-PCR has provided valuable information about the diversity of rotaviruses (RV) circulating throughout the world.</p> <p>The purpose of the present study was to monitor the prevalence of the different G and P genotypes of rotaviruses circulating in Salento and detect any uncommon or novel types.</p> <p>Methods</p> <p>During the period from January 2006 to December 2007, a total of 243 rotavirus positive stool samples were collected from children with diarrhoea admitted to four Hospitals in the province of Lecce (Copertino, Galatina, Gallipoli and Tricase).</p> <p>All the specimens were tested for RV by real time PCR and genotyped for VP7 (G-type) and VP4 (P-type) gene by reverse transcription (RT) and multiplex PCR using different type specific primers.</p> <p>Results</p> <p>In course of this study we identified 4 common G&P combinations viz. G2P[8], G1P[8], G2P[4] and G9P[8] amongst 59.8% of the typeable rotavirus positives.</p> <p>Rotavirus G2P[8] was recognized as the most widespread genotype during the sentinel-based survey in Salento.</p> <p>The detection of other novel and unusual strains, such as G2P[10], G4P[10], G8P[4], G9P[11] and G10P[8] is noteworthy.</p> <p>Furthermore, a significant number of mixed infections were observed during the survey period but G3P[8] rotaviruses were not detected.</p> <p>Conclusion</p> <p>This study highlights the genetic diversity among rotaviruses isolated from children in Salento and the emergence of some novel strains. Therefore, it is highly essential to continuously monitor for these strains so as to assess the impact of vaccines on RV strains circulating in Salento and understand the effect of strain variation on efficacy of presently available vaccines.</p
Lifestyles and socio-cultural factors among children aged 6-8 years from five Italian towns: The MAPEC-LIFE study cohort
Background: Lifestyles profoundly determine the quality of an individual’s health and life since his childhood.
Many diseases in adulthood are avoidable if health-risk behaviors are identified and improved at an early stage of
life. The aim of the present research was to characterize a cohort of children aged 6–8 years selected in order to
perform an epidemiological molecular study (the MAPEC_LIFE study), investigate lifestyles of the children that
could have effect on their health status, and assess possible association between lifestyles and socio-cultural factors.
Methods: A questionnaire composed of 148 questions was administered in two different seasons to parents of
children attending 18 primary schools in five Italian cities (Torino, Brescia, Pisa, Perugia and Lecce) to obtain
information regarding the criteria for exclusion from the study, demographic, anthropometric and health
information on the children, as well as some aspects on their lifestyles and parental characteristics. The results
were analyzed in order to assess the frequency of specific conditions among the different seasons and cities and
the association between lifestyles and socio-economic factors.
Results: The final cohort was composed of 1,164 children (50.9 boys, 95.4% born in Italy). Frequency of some
factors appeared different in terms of the survey season (physical activity in the open air, the ways of cooking
certain foods) and among the various cities (parents’ level of education and rate of employment, sport, traffic
near the home, type of heating, exposure to passive smoking, ways of cooking certain foods). Exposure to
passive smoking and cooking fumes, obesity, residence in areas with heavy traffic, frequency of outdoor play and
consumption of barbecued and fried foods were higher among children living in families with low educational
and/or occupational level while children doing sports and consuming toasted bread were more frequent in families
with high socio-economic level.
Conclusions: The socio-economic level seems to affect the lifestyles of children enrolled in the study including
those that could cause health effects. Many factors are linked to the geographical area and may depend on
environmental, cultural and social aspects of the city of residence

Epidemiology of jellyfish stings reported to summer health centres in the Salento peninsula (Italy).
The presence of jellyfish along coasts constitutes a potentially serious public health
problem because of the toxic effects of contact with them.
To determine the epidemiology of jellyfish stings in the Province of Lecce, Italy, in 2007.
The study involved the gathering and processing of data on patients seeking medical
treatment at summer first aid centres and hospitals along the coast of Salento following contact with
jellyfish.
In 2007, 446 bathers sought medical assistance following contact with jellyfish. The spatial
distribution indicates a greater frequency of stings sustained along the Adriatic coast, while the time
analysis shows patterns related to wind direction. Most of the injuries were seen in persons aged
between 1 and 10 years and in females. In 24% of cases, injuries were sustained to several parts of the
body, while single injuries were mainly to the lower limbs (33%). In 4.9% of cases, there were
complications, including one case of anaphylactic shock.
In the light of the significant level of medical assistance provided, there is a need to
identify suitable strategies that will guarantee the best response in terms of both prevention and
3 treatment
