    AKT activation controls cell survival in response to HDAC6 inhibition.

    HDAC6 is emerging as an important therapeutic target for cancer. We investigated mechanisms responsible for survival of tumor cells treated with a HDAC6 inhibitor. Expression of the 20 000 genes examined did not change following HDAC6 treatment in vivo. We found that HDAC6 inhibition led to an increase of AKT activation (P-AKT) in vitro, and genetic knockdown of HDAC6 phenocopied drug-induced AKT activation. The activation of AKT was not observed in PTEN null cells; otherwise, PTEN/PIK3CA expression per se did not predict HDAC6 inhibitor sensitivity. Interestingly, HDAC6 inhibitor treatment led to inactivating phosphorylation of PTEN (P-PTEN Ser380), which likely led to the increased P-AKT in cells that express PTEN. Synergy was observed with phosphatidylinositol 3-kinases (PI3K) inhibitor treatment in vitro, accompanied by increased caspase 3/7 activity. Furthermore, combination of HDAC6 inhibitor with a PI3K inhibitor caused substantial tumor growth inhibition in vivo compared with either treatment alone, also detectable by Ki-67 immunostaining and (18)F-FLT positron emission tomography (PET). In aggregate AKT activation appears to be a key survival mechanism for HDAC6 inhibitor treatment. Our findings indicate that dual inhibition of HDAC6 and P-AKT may be necessary to substantially inhibit growth of solid tumors

    Estimation of Reminant Gold Resources of Old Underground Workings by Indicator Approach – A Case Study

    The lack of geological control and old underground development at AngloGold Ashanti (Bibiani mine) in Ghana was a major setback for the conventional methods of resource estimation to produce accurate estimates of the deposit. Consequently, inverse distance weighting of indicator variables were used. This estimation approach, however, overestimated the boundaries of the old underground workings (voids) into inappropriate areas of the resource model and thereby caused significant underestimation of ore tonnage and overestimation of grade. This paper highlights the study that re-evaluated the Bibiani deposit using indicator kriging.     A holistic review of off-site construction literature published between 2008 and 2018

    Off-site construction (i.e., OSC) has become an emerging research domain in the recent decade. Through a three-step holistic review approach incorporating bibliometric search, scientometric analysis, and in-depth qualitative discussion, this study contributes to the body of knowledge in OSC by critically reviewing and summarizing: 1) the latest research keywords and main research topics in OSC; 2) the performance of OSC compared to that of conventional construction approach; 3) current research gaps in integrating OSC with other emerging construction concepts; and 4) future research directions in OSC. OSC is a domain that can be extended to cross-disciplinary research from the perspectives of engineering, management, and technology. Existing research have been focusing on many research disciplines, such as structural behaviors and joint connections of prefabricated components, scheduling and planning of off-site activities, as well as performance evaluation of OSC. However, further research is needed in integrating the emerging digital construction technology, integrated project delivery method, lean construction, and issues of sustainability of OSC. There are still limited studies linking OSC to the concept of Design for Manufacturing and Assembly. Future research should also adopt a larger database and allow for comprehensive evaluation of OSC performance

    Preclinical Applications of 3'-Deoxy-3'-[18F]Fluorothymidine in Oncology - A Systematic Review

    The positron emission tomography (PET) tracer 3'-deoxy-3'-[18F]fluorothymidine ([18F]FLT) has been proposed to measure cell proliferation non-invasively in vivo. Hence, it should provide valuable information for response assessment to tumor therapies. To date, [18F]FLT uptake has found limited use as a response biomarker in clinical trials in part because a better understanding is needed of the determinants of [18F]FLT uptake and therapy-induced changes of its retention in the tumor. In this systematic review of preclinical [18F]FLT studies, comprising 174 reports, we identify the factors governing [18F]FLT uptake in tumors, among which thymidine kinase 1 plays a primary role. The majority of publications (83 %) report that decreased [18F]FLT uptake reflects the effects of anticancer therapies. 144 times [18F]FLT uptake was related to changes in proliferation as determined by ex vivo analyses. Of these approaches, 77 % describe a positive relation, implying a good concordance of tracer accumulation and tumor biology. These preclinical data indicate that [18F]FLT uptake holds promise as an imaging biomarker for response assessment in clinical studies. Understanding of the parameters which influence cellular [18F]FLT uptake and retention as well as the mechanism of changes induced by therapy is essential for successful implementation of this PET tracer. Hence, our systematic review provides the background for the use of [18F]FLT in future clinical studies

    Investigating major subject research areas of Council for Scientific and Industrial Research Journals in Ghana

    Journals are important source of relevant and quality information. They provide current source of up-to-date information to facilitate research, teaching, learning and knowledge dissemination. The objective of the study is to investigate the major subject research areas covered by the articles of the five journals in the Council for Scientific and Industrial Research. The study was undertaken in the CSIR of Ghana in which a major device for data collection was quantitative content analysis. This mode of data collection was determined after its validity and reliability among 1,430 journal articles from four CSIR Institutes was proven. Data collected were analysed using tables and graphs to group major publications. Results from the research indicated that cereal and legumes recorded the highest (318) disciplines in all the five journals in the CSIR

    The Emergence of Gated Communities in Ghana and their Implications on Urban Planning and Management

    Over the past few decades, gated communities have emerged as global commodity increasingly gaining patronage. In Sub-Sahara Africa, the city of Accra is fast recording the emergence of these communities. The research therefore examined the implications of the rising phenomenon in urban planning and management. By adopting a mixture of snow ball, simple random and purposive sampling and a face to face interview technique, the research accessed empirical data from planning institutions and households selected from three gated communities in the capital city of Accra. It was found that, gated communities in the capital have relatively good infrastructural facilities and services, the provision of which the real estate developers played a major role. However, a spatial policy gap on gated communities was identified in addition to missing legislative framework and low level of awareness on the part of planning institutions regarding the existence of some gated communities in the capital. In ensuring effective planning and management of gated communities in the urban arena as well as situating their occurrence within a broader spatial and socio-economic development of the country, the research concludes that, formulation of spatial policy and legislative framework and strengthening of planning institutions are requisite.     Intimate partner violence as a predictor of marital disruption in sub-Saharan Africa: A multilevel analysis of demographic and health surveys

    Intimate partner violence has gained momentum as health, social, and human right issue across the globe. Women within sub-Saharan Africa often do not report any case of violence due to the acceptance of violence which is rooted in their socio-cultural beliefs and practices. With a high prevalence of marital disruption in sub-Saharan Africa, it is important that we understand the role intimate partner violence plays in this phenomenon. Hence, this present study assessed the association between intimate partner violence and marital disruption among women in sub-Saharan Africa. Methods This study involved a cross-sectional analysis of data from the Demographic and Health Survey of 25 countries in sub-Saharan Africa. Multilevel binary logistic regression analysis was carried out and the results were presented as adjusted odds ratios (aOR) at 95% Confidence Interval (CI). Results The prevalence of physical, emotional, and sexual violence in the 25 countries considered in this study were 29.3%, 28%, and 11.5%, respectively. The highest prevalence of physical violence was in Sierra Leone (50.0%) and the lowest prevalence was in Comoros (5.7%). For emotional violence, the highest prevalence was in Sierra Leone (45.9%) and the lowest prevalence was in Comoros (7.9%). The highest prevalence of sexual violence was in Burundi (25.5%) and the lowest prevalence was in Comoros (1.8%). The average prevalence of marital disruption was 7.7%. This ranged from 1.3% in Burkina Faso to 20.2% in Mozambique. We found that women who had ever experienced physical violence were more likely to experience marital disruptions compared to those who had never experienced physical violence [aOR = 1.42, 95% CI = 1.35–1.50]. Women who had ever experienced sexual violence were more likely to experience marital disruption compared to those who had never experienced sexual violence [aOR = 1.29, 95% CI = 1.21–1.37]. Finally, women who had ever experienced emotional violence were more likely to experience marital disruption compared to those who had never experienced emotional violence [aOR = 1.86, 95% CI = 1.76–1.96]. Conclusion Findings from this study call for proven effective intimate partner violence reduction interventions such as strengthening laws against intimate partner violence in sub-Saharan Africa. Again, marital counseling and health education interventions should be implemented to address the role of intimate partner violence on the wellbeing of women and the stability of couples in sub-Saharan Africa

    A Study of the Publication pattern in CSIR- Plant Genetic Resources Research Institute

    Plant genetic resources (PGR) are those resources that are of benefit to man. They are plant materials containing useful characters of actual or potential values. They are the basic raw mate­rials for crop improvement today and for the future. This paper analysed changes in publication trend by scientists from 1981 to 2015 at the CSIR-Plant Genetic Resources Research Institute. It investigated major commodities covered by the articles produced by scientists of the Institute. The main area of study included publication formats covered by the articles in the Plant Ge­netic Resources Research Institute Reference (PGRRIREF Directory) (1981-2015). The study also highlighted recent research and development activities in each publication discipline in the PGRRIREF Directory. It is believed that information gathered from the analysis of this research, would increase the utilization of the crop plants in Ghana and beyond. Content analysis method and interviews were used for the study of the Publication pattern in CSIR-Plant Genetic Re­sources Research Institute. The findings indicated among others that, socio-economic (27.2%), horticulture (21.5%), root and tubers (18.5%) recorded increasing publication disciplines.
