Highly disordered histone H1-DNA model complexes and their condensates.
Disordered proteins play an essential role in a wide variety of biological processes, and are often posttranslationally modified. One such protein is histone H1; its highly disordered C-terminal tail (CH1) condenses internucleosomal linker DNA in chromatin in a way that is still poorly understood. Moreover, CH1 is phosphorylated in a cell cycle-dependent manner that correlates with changes in the chromatin condensation level. Here we present a model system that recapitulates key aspects of the in vivo process, and also allows a detailed structural and biophysical analysis of the stages before and after condensation. CH1 remains disordered in the DNA-bound state, despite its nanomolar affinity. Phase-separated droplets (coacervates) form, containing higher-order assemblies of CH1/DNA complexes. Phosphorylation at three serine residues, spaced along the length of the tail, has little effect on the local properties of the condensate. However, it dramatically alters higher-order structure in the coacervate and reduces partitioning to the coacervate phase. These observations show that disordered proteins can bind tightly to DNA without a disorder-to-order transition. Importantly, they also provide mechanistic insights into how higher-order structures can be exquisitely sensitive to perturbation by posttranslational modifications, thus broadening the repertoire of mechanisms that might regulate chromatin and other macromolecular assemblies
Detection of OH absorption against PSR B1849+00
We have searched for OH absorption against seven pulsars using the Arecibo
telescope. In both OH mainlines (at 1665 and 1667 MHz), deep and narrow
absorption features were detected toward PSR B1849+00. In addition, we have
detected several absorption and emission features against B33.6+0.1, a nearby
supernova remnant (SNR). The most interesting result of this study is that a
pencil-sharp absorption sample against the PSR differs greatly from the
large-angle absorption sample observed against the SNR. If both the PSR and the
SNR probe the same molecular cloud then this finding has important implications
for absorption studies of the molecular medium, as it shows that the statistics
of absorbing OH depends on the size of the background source. We also show that
the OH absorption against the PSR most likely originates from a small (<30

Genetic Modification of the Association between Peripubertal Dioxin Exposure and Pubertal Onset in a Cohort of Russian Boys
Background: Exposure to dioxins has been associated with delayed pubertal onset in both epidemiologic and animal studies. Whether genetic polymorphisms may modify this association is currently unknown. Identifying such genes could provide insight into mechanistic pathways. This is one of the first studies to assess genetic susceptibility to dioxins. Objectives: We evaluated whether common polymorphisms in genes affecting either molecular responses to dioxin exposure or pubertal onset influence the association between peripubertal serum dioxin concentration and male pubertal onset. Methods: In this prospective cohort of Russian adolescent boys (n = 392), we assessed geneâenvironment interactions for 337 tagging single-nucleotide polymorphisms (SNPs) from 46 candidate genes and two intergenic regions. Dioxins were measured in the boysâ serum at age 8â9 years. Pubertal onset was based on testicular volume and on genitalia staging. Statistical approaches for controlling for multiple testing were used, both with and without prescreening for marginal genetic associations. Results: After accounting for multiple testing, two tag SNPs in the glucocorticoid receptor (GR/NR3C1) gene and one in the estrogen receptor-α (ESR1) gene were significant (q < 0.2) modifiers of the association between peripubertal serum dioxin concentration and male pubertal onset defined by genitalia staging, although not by testicular volume. The results were sensitive to whether multiple comparison adjustment was applied to all geneâenvironment tests or only to those with marginal genetic associations. Conclusions: Common genetic polymorphisms in the glucocorticoid receptor and estrogen receptor-α genes may modify the association between peripubertal serum dioxin concentration and pubertal onset. Further studies are warranted to confirm these findings
Exploring the blueprint of photoprotection in Mycosporine-like Amino Acids
Microorganisms require protection against the potentially damaging effects of ultraviolet radiation exposure. Photoprotection is, in part, provided by mycosporine-like amino acids (MAAs). Previous reports have proposed that nonradiative decay mediates the impressive photoprotection abilities of MAAs. In this letter, we present the first ultrafast dynamics study of two MAAs, shinorine and porphyra-334. We demonstrate that, in aqueous solution, these MAAs relax along their S1 coordinates toward the S1/S0 conical intersection within a few hundred femtoseconds after photoexcitation and then traverse the conical intersection and vibrationally cool in approximately 1 ps through heat transfer to the solvent. This new insight allows a quintessential component of microbial life to be unraveled and informs the development of molecular photon-to-heat converters for a myriad of applications

Perceived control over condom use among sex workers in Madagascar: a cohort study
<p>Abstract</p> <p>Background</p> <p>Women's perceived control over condom use has been found to be an important determinant of actual condom use in some studies. However, many existing analyses used cross-sectional data and little quantitative information exists to characterize the relationships between perceived control and actual condom use among sex worker populations.</p> <p>Methods</p> <p>We assessed the association between measures of perceived condom use control and self-reported use of male condoms employing data from a longitudinal pilot study among 192 sex workers in Madagascar.</p> <p>Results</p> <p>In multivariable models, a lack of perceived control over condom use with a main partner and having a main partner ever refuse to use a condom when asked were both associated with an increased number of sex acts unprotected by condoms in the past week with a main partner (RR 1.86; 95% CI 1.21-2.85; RR 1.34; 95% CI 1.03-1.73, respectively). Conversely, no measure of condom use control was significantly associated with condom use with clients.</p> <p>Conclusion</p> <p>Perceived control over condom use was an important determinant of condom use with main partners, but not clients, among sex workers in Madagascar. Programs working with sex workers should reach out to main and commercial partners of sex workers to increase male condom use.</p

The QUaD Galactic Plane Survey. I. Maps and Analysis of Diffuse Emission
We present a survey of ~800 deg of the galactic plane observed with the QUaD telescope. The primary products of the survey are maps of Stokes I, Q, and U parameters at 100 and 150 GHz, with spatial resolution of 5' and 3'.5, respectively. Two regions are covered, spanning approximately 245°-295° and 315°-5° in the galactic longitude l and â4° < b < +4° in the galactic latitude b. At 0°.02 square pixel size, the median sensitivity is 74 and 107 kJy sr at 100 GHz and 150 GHz respectively in I, and 98 and 120 kJy sr for Q and U. In total intensity, we find an average spectral index of α = 2.35 ± 0.01(stat) ± 0.02(sys) for |b| †1°, indicative of emission components other than thermal dust. A comparison to published dust, synchrotron, and free-free models implies an excess of emission in the 100 GHz QUaD band, while better agreement is found at 150 GHz. A smaller excess is observed when comparing QUaD 100 GHz data to the WMAP five-year W band; in this case, the excess is likely due to the wider bandwidth of QUaD. Combining the QUaD and WMAP data, a two-component spectral fit to the inner galactic plane (|b| †1°) yields mean spectral indices of α s = â0.32 ± 0.03 and α = 2.84 ± 0.03; the former is interpreted as a combination of the spectral indices of synchrotron, free-free, and dust, while the second is largely attributed to the thermal dust continuum. In the same galactic latitude range, the polarization data show a high degree of alignment perpendicular to the expected galactic magnetic field direction, and exhibit mean polarization fraction 1.38 ± 0.08(stat) ± 0.1(sys)% at 100 GHz and 1.70 ± 0.06(stat) ± 0.1(sys)% at 150 GHz. Probing Cosmic Reionization and Molecular Gas Growth with TIME
Line intensity mapping (LIM) provides a unique and powerful means to probe
cosmic structures by measuring the aggregate line emission from all galaxies
across redshift. The method is complementary to conventional galaxy redshift
surveys that are object-based and demand exquisite point-source sensitivity.
The Tomographic Ionized-carbon Mapping Experiment (TIME) will measure the star
formation rate (SFR) during cosmic reionization by observing the redshifted
[CII] 158m line () in the LIM regime. TIME will
simultaneously study the abundance of molecular gas during the era of peak star
formation by observing the rotational CO lines emitted by galaxies at . We present the modeling framework that predicts the
constraining power of TIME on a number of observables, including the line
luminosity function, and the auto- and cross-correlation power spectra,
including synergies with external galaxy tracers. Based on an optimized survey
strategy and fiducial model parameters informed by existing observations, we
forecast constraints on physical quantities relevant to reionization and galaxy
evolution, such as the escape fraction of ionizing photons during reionization,
the faint-end slope of the galaxy luminosity function at high redshift, and the
cosmic molecular gas density at cosmic noon. We discuss how these constraints
can advance our understanding of cosmological galaxy evolution at the two
distinct cosmic epochs for TIME, starting in 2021, and how they could be

Parameter Estimation From Improved Measurements of the Cosmic Microwave Background From QUaD
We evaluate the contribution of cosmic microwave background (CMB) polarization spectra to cosmological parameter constraints. We produce cosmological parameters using high-quality CMB polarization data from the ground-based QUaD experiment and demonstrate for the majority of parameters that there is significant improvement on the constraints obtained from satellite CMB polarization data. We split a multi-experiment CMB data set into temperature and polarization subsets and show that the best-fit confidence regions for the ÎCDM six-parameter cosmological model are consistent with each other, and that polarization data reduces the confidence regions on all parameters. We provide the best limits on parameters from QUaD EE/BB polarization data and we find best-fit parameters from the multi-experiment CMB data set using the optimal pivot scale of k = 0.013 Mpc to be {hΩ, hΩ, H, A, n, Ï} = {0.113, 0.0224, 70.6, 2.29 Ă 10, 0.960, 0.086}.Astronom
