Inhibition of Progenitor Dendritic Cell Maturation by Plasma from Patients with Peripartum Cardiomyopathy: Role in Pregnancy-associated Heart Disease
Dendritic cells (DCs) play dual roles in innate and adaptive immunity based
on their functional maturity, and both innate and adaptive immune responses have
been implicated in myocardial tissue remodeling associated with
cardiomyopathies. Peripartum cardiomyopathy (PPCM) is a rare disorder which
affects women within one month antepartum to five months postpartum. A high
occurrence of PPCM in central Haiti (1 in 300 live births) provided the unique
opportunity to study the relationship of immune activation and DC maturation
to the etiology of this disorder. Plasma samples from two groups (n = 12) of
age- and parity-matched Haitian women with or without evidence of PPCM were
tested for levels of biomarkers of cardiac tissue remodeling and immune
activation. Significantly elevated levels of GM-CSF, endothelin-1, proBNP and
CRP and decreased levels of TGF- were measured in PPCM subjects relative
to controls. Yet despite these findings, in vitro maturation of normal human
cord blood derived progenitor dendritic cells (CBDCs) was significantly
reduced (p < 0.001) in the presence of plasma from PPCM patients relative
to plasma from post-partum control subjects as determined by expression of
CD80, CD86, CD83, CCR7, MHC class II and the ability of these matured CBDCs
to induce allo-responses in PBMCs. These results represent the first findings
linking inhibition of DC maturation to the dysregulation of normal physiologic
cardiac
tissue remodeling during pregnancy and the pathogenesis of PPCM
Vietnam military service history and prostate cancer
BACKGROUND: Three decades after US and Australian forces withdrew from Vietnam, there has been much public interest in the health consequences of service in Vietnam. One controversial question is whether the risk of prostate cancer amongst Vietnam veterans is increased. This paper examines relationships between military history, family history and risk of prostate cancer in a population-based case control study. METHODS: Cases were selected from the Cancer Registry of Western Australia as incident cases of histologically-confirmed prostate cancer, and controls were age-matched and selected from the Western Australian electoral roll. Study participants were asked to report any military service history and details about that service. RESULTS: Between January 2001 and September 2002, 606 cases and 471 controls aged between 40–75 years were recruited. An increased prostate cancer risk was observed in men reporting they were deployed in Vietnam although this was not statistically significant (OR = 2.12; 95% CI 0.88–5.06). An increased risk was also observed in men reporting prostate cancer in fathers (OR = 1.90; 95% CI 1.20–3.00) or brothers (OR = 2.05; 95% CI 1.20–3.50) diagnosed with prostate cancer. CONCLUSION: These findings support a positive association between prostate cancer and military service history in the Vietnam war and a first degree relative family history of prostate cancer
Genome Sequence Analyses of Pseudomonas savastanoi pv. glycinea and Subtractive Hybridization-Based Comparative Genomics with Nine Pseudomonads
Bacterial blight, caused by Pseudomonas savastanoi pv. glycinea (Psg), is a common disease of soybean. In an effort to compare a current field isolate with one isolated in the early 1960s, the genomes of two Psg strains, race 4 and B076, were sequenced using 454 pyrosequencing. The genomes of both Psg strains share more than 4,900 highly conserved genes, indicating very low genetic diversity between Psg genomes. Though conserved, genome rearrangements and recombination events occur commonly within the two Psg genomes. When compared to each other, 437 and 163 specific genes were identified in B076 and race 4, respectively. Most specific genes are plasmid-borne, indicating that acquisition and maintenance of plasmids may represent a major mechanism to change the genetic composition of the genome and even acquire new virulence factors. Type three secretion gene clusters of Psg strains are near identical with that of P. savastanoi pv. phaseolicola (Pph) strain 1448A and they shared 20 common effector genes. Furthermore, the coronatine biosynthetic cluster is present on a large plasmid in strain B076, but not in race 4. In silico subtractive hybridization-based comparative genomic analyses with nine sequenced phytopathogenic pseudomonads identified dozens of specific islands (SIs), and revealed that the genomes of Psg strains are more similar to those belonging to the same genomospecies such as Pph 1448A than to other phytopathogenic pseudomonads. The number of highly conserved genes (core genome) among them decreased dramatically when more genomes were included in the subtraction, suggesting the diversification of pseudomonads, and further indicating the genome heterogeneity among pseudomonads. However, the number of specific genes did not change significantly, suggesting these genes are indeed specific in Psg genomes. These results reinforce the idea of a species complex of P. syringae and support the reclassification of P. syringae into different species
