    Model-guided design of ligand-regulated RNAi for programmable control of gene expression

    Progress in constructing biological networks will rely on the development of more advanced components that can be predictably modified to yield optimal system performance. We have engineered an RNA-based platform, which we call an shRNA switch, that provides for integrated ligand control of RNA interference (RNAi) by modular coupling of an aptamer, competing strand, and small hairpin (sh) RNA stem into a single component that links ligand concentration and target gene expression levels. A combined experimental and mathematical modelling approach identified multiple tuning strategies and moves towards a predictable framework for the forward design of shRNA switches. The utility of our platform is highlighted by the demonstration of fine-tuning, multi-input control, and model-guided design of shRNA switches with an optimized dynamic range. Thus, shRNA switches can serve as an advanced component for the construction of complex biological systems and offer a controlled means of activating RNAi in disease therapeutics

    The aim of this study is to examine population growth rates, and housing needs of Irbid city. For this purpose, the study took into consideration the housing needs in Irbid city. The study based on population census data used certain quantitative method to estimate population growth rates.and population projections in future , The annual decay rate of houses, and counted housing need resulted from decreasing occupancy rate to 2.5 person/room. The analysis of the study showed that the district witnessed rapid population growth. For instance, the population growth rate was 4.17% during 1979-1994. while it decreased to 2.05% during 1994-2004. The study, as a result, found that is 9.3% of population have occupancy rates of more than 3.5 person/room in the district. However, the study explained that the estimated housing needs for the whole district is 29684 housing unit during 2004-2024, which equals 1484 housing units annually

    Semi-hyperbolic mappings in Banach spaces

    The definition of semi-hyperbolic dynamical systems generated by Lipschitz continuous and not necessarily invertible mappings in Banach spaces is presented in this thesis. Like hyperbolic mappings, they involve a splitting into stable and unstable spaces, but a slight leakage from the strict invariance of the spaces is possible and the unstable subspaces are assumed to be finite dimensional. Bi-shadowing is a combination of the concepts of shadowing and inverse shadowing and is usually used to compare pseudo-trajectories calculated by a computer with the true trajectories. In this thesis, the concept of bi-shadowing in a Banach space is defined and proved for semi-hyperbolic dynamical systems generated by Lipschitz mappings. As an application to the concept of bishadowing, linear delay differential equations are shown to be bi-shadowing with respect to pseudo-trajectories generated by nonlinear small perturbations of the linear delay equation. This shows robustness of solutions of the linear delay equation with respect to small nonlinear perturbations. Complicated dynamical behaviour is often a consequence of the expansivity of a dynamical system. Semi-hyperbolic dynamical systems generated by Lipschitz mappings on a Banach space are shown to be exponentially expansive, and explicit rates of expansion are determined. The result is applied to a nonsmooth noninvertible system generated by delay differential equation. It is shown that semi-hyperbolic mappings are locally φ-contracting, where -0 is the Hausdorff measure of noncompactness, and that a linear operator is semi-hyperbolic if and only if it is φ-contracting and has no spectral values on the unit circle. The definition of φ-bi-shadowing is given and it is shown that semi-hyperbolic mappings in Banach spaces are φ-bi-shadowing with respect to locally condensing continuous comparison mappings. The result is applied to linear delay differential equations of neutral type with nonsmooth perturbations. Finally, it is shown that a small delay perturbation of an ordinary differential equation with a homoclinic trajectory is ‘chaotic’

    Optical control of spin coherence in singly charged (In,Ga)As/GaAs quantum dots

    Electron spin coherence has been generated optically in n-type modulation doped (In,Ga)As/GaAs quantum dots (QDs) which contain on average a single electron per dot. The coherence arises from resonant excitation of the QDs by circularly-polarized laser pulses, creating a coherent superposition of an electron and a trion state. Time dependent Faraday rotation is used to probe the spin precession of the optically oriented electrons about a transverse magnetic field. Spin coherence generation can be controlled by pulse intensity, being most efficient for (2n+1)pi-pulses.Comment: 5 pages, 4 figure

    Formulation of a 1D finite element of heat exchanger for accurate modelling of the grouting behaviour: Application to cyclic thermal loading

    This paper presents a comprehensive formulation of a finite element for the modelling of borehole heat exchangers. This work focuses on the accurate modelling of the grouting and the field of temperature near a single borehole. Therefore the grouting of the BHE is explicitly modelled. The purpose of this work is to provide tools necessary to the further modelling of thermo-mechanical couplings. The finite element discretises the classical governing equation of advection-diffusion of heat within a 1D pipe connected to ground nodes. Petrov-Galerkin weighting functions are used to avoid numerical disturbances. The formulation is able to capture highly transient and steady-state phenomena. The proposed finite element is validated with respect to analytical solutions. An example consisting of a 100 m depth U-pipe is finally simulated. A first continuous heating simulation highlights the nonsymmetric distribution of temperature inside and near the borehole. An estimation of the error on the results as a function of the resolution parameters is also carried out. Finally simulations of cyclic thermal loading exhibit the need to take into account all daily variations if the grouting behaviour must be modelled.     Decomposition of semigroup algebras

    Let A \subseteq B be cancellative abelian semigroups, and let R be an integral domain. We show that the semigroup ring R[B] can be decomposed, as an R[A]-module, into a direct sum of R[A]-submodules of the quotient ring of R[A]. In the case of a finite extension of positive affine semigroup rings we obtain an algorithm computing the decomposition. When R[A] is a polynomial ring over a field we explain how to compute many ring-theoretic properties of R[B] in terms of this decomposition. In particular we obtain a fast algorithm to compute the Castelnuovo-Mumford regularity of homogeneous semigroup rings. As an application we confirm the Eisenbud-Goto conjecture in a range of new cases.     Brucella Endocarditis with Splenic Abscess: A Report of the First Case Diagnosed in Korea

    Human brucellosis has a broad spectrum of clinical manifestations, which includes endocarditis, a focal complication that is uncommon yet responsible for the majority of associated deaths. The most successful treatment outcomes of Brucella endocarditis have been reported with usage of both antimicrobial agents and surgery. However, there are few reports on the treatment of Brucella endocarditis using antibiotics only. We report the first case in Korea of Brucella endocarditis with aortic valve vegetations and an accompanying splenic abscess, which were treated successfully with antibiotic therapy alone
